Stress enhances expression of calcium-binding proteins and nMDAr subunit genes in the rat hippocampus

Backgrounds:Oxidative stress impairs the function of calcium-binding proteins and deregulates calcium signaling in living *** have previously explored the overexpression of calcium-binding protein genes in a reactive oxygen and nitrogen species-induced in vitro cell model of stress that leads to ***,in in vivo models,low levels of stress leads to depressive-like ***,we aimed to analyze gene expression of major calcium-binding proteins(calcineurin,calmodulin,calsyntenin,synaptotagmin,and calreticulin)and n-methyl-D-aspartic acid(nMDA)receptor subunits(glutamate receptor ionotropic[Glun]Glun1,Glun2A,and Glun2B)in the hippocampus of stress-induced ***:Six-week-old male Wistar rats were assigned to two stress induction groups and a control group without stress(n=6).Stress was induced by using H_(2)O_(2)(3%in water)or by immobilization(using a sticky mat)over a period of 30 *** of calcium-binding protein genes in the hippocampus,antioxidant assays,structural alterations in hippocampal neurons,and depressive-like behavior were ***:Expression of genes encoding calcium-binding proteins calcineurin,calsyntenin,synaptotagmin and nMDA receptor subunit Glun1 was enhanced in both chemical and physical stress-induced rats compared with control rats(4.25±0.05 vs.1.03±0.02,press-induced rats,neurons in the CA2 region of the hippocampus were fewer and appeared disorganized compared with control ***,stressinduced rats showed decreased mobility and lower sucrose preference in behavioral studies compared with control ***:Lower levels of reactive oxygen and nitrogen species(rOnS)can also lead to stress in rats by affecting their calcium signaling,buffering capacity in the neurons leading to depressive symptoms.

Tablets are one of the most comm only used solid dosage forms taken by *** preparati on of high-quality tablets requires an understanding of the preparation process and elucidation of how the physical and mechanical properties change as a function of the preparation *** work aims to investigate the impact of microwave irradiation drying and conventional drying methods(including freeze,convective and vacuum drying)on the hardness,tensile strength and friability of tablets made from a multi-component formulation containing naproxen sodium,microcrystalline cellulose,and *** results show that tablets subjected to microwave drying had the secondhighest tensile strength and hardness of 1.296 MPa and 67 n,*** tablets subjected to vacuum drying had the lowest tensile strength and hardness of 1.21 MPa and 64 n,*** friability index values for the tablets derived from the microwave and freeze-drying methods werer the tablets subjected to convective drying and vacuum drying methods were>l%.Microwave drying was observed to be an efficient method to produce naproxen sodium-containing tablets with satisfactory mechanical *** findings confirm that the drying method plays an essential role in the improvement or degradation of the mechanical properties of tablets.

We report here for the first time the composition and mineralogical studies on a new meteorite, which fell in Dhayala ki Chappar （24°58°n, 73° 48°27°E） 5 km nW of nathdwara in south rajasthan, India, on Dec. 25th, 2012. Mineralogical and compositional studies were carried out on a representative piece of the nathdwara meteorite sample. The mineralogical composition of the meteorite has been found to be olivine （42 45 vol.%）, feldspar （10-15 vol.%）, orthopyroxene （23-25 vol.%）, troilite （6 8 vol.%）, and titanium bearing minerals （6-8 vol.%）. Our investigations show that the nathdwara meteorite belongs to H6 group of ordinary chondrites.

Characterization of amphiboles from the Kola super-deep borehole, russia by raman and infrared spectroscopy

We present here for the first time,the raman and infrared spectroscopic investigation of amphiboles from the World's deepest borehole,the Kola super-deep borehole,at the depth of 11.66 *** Kola Super-deep borehole(SG-3)(henceforth referred as KSDB)is located in the northwest of the Kola Peninsula in the northern frame of the Pechenga structure,*** was drilled in the north-eastern part of the Baltic Shield(69о5’n,30о44’E)and reached a depth of 12.262 *** has been drilled in the northern limb of the Pechenga geosyncline composed of rhythmically inter-bedded volcanogenic and tuffaceous-sedimentary strata extending to the nW at 300°–310°and dipping to SW at angles of 30°–50°.The SG-3 geological section is represented by two complexes–Proterozoic and *** facies is dominant in the depth region from 6000 m to 12,000m to the *** raman spectra of the sample reveal abundant presence of plagioclase and *** most distinct raman peak in this study indicates the tremolite-ferro-actinolite rich enrichment of the borehole samples at this depth corroborating earlier conventional petrographic studies.

Macitentan (MAC) is a pulmonary arterial hypertension (PAH) drug marketed as a tablet and often has stability issues in the final dosage form. Quantitative determination of MAC and its associated impurities in tablet dosage form has not been previously reported. This study quantified impurities present in Macitentan tablets using a binary solvent-based gradient elution method using reversed phase-high performance liquid *** developed method w as validated per International Conference on Harmonization (ICH) guidelines and the drug product w as subjected to forced degradation studies to evaluate stability. The developed method efficiently separated the drug and impurities (48 min) w ithout interference from solvents,excipients,or other impurities. The developed method met all guidelines in all characteristics w ith recoveries ranging from 85%-115%,linearity w ith r 2≥0. 996 6,and substantial robustness. The stability-indicating nature of the method w as evaluated using stressed conditions (hydrolysis:1 n HCl at 80℃/15 min; 1 n naOH at 25℃/45 min; humidity stress (90%relative humidity) at 25℃for 24 h,oxidation:at 6%(v/v) H2O2,80℃/15 min,thermolysis:at105℃/16 h and photolysis:UV light at 200 Wh/m2; Fluorescent light at 1. 2 million luxh). Forced degradation experiments show ed that the developed method w as effective for impurity profiling. All stressed samples w ere assayed and mass balance w as> 96%. Forced degradation results indicated that MAC tablets w ere sensitive to hydrolysis (acid and alkali) and thermal conditions. The developed method is suitable for both assay and impurity determination,w hich is applicable to the pharmaceutical industry.

To increase the accurate finishing productivity of Inconel 600, plain turning by Tantalum thin film hard coated K.20 carbide insert is studied. The main focus of this investigation is the turning performance of Inconel 600 with coated cutting insert by studying the relation ships among machining conditions such as feed rate, tool wear, cutting speed, depth of cut and work piece surface quality and material removal rate. Machining experiments are conducted on self centering medium duty lathe under different machining conditions. The result provides some useful information.

有些工厂排放的废水中常含有大量的氟化物。对于含氟废水的处理,通常采用钙盐（特别是石灰）或钙盐加铝盐的沉淀处理法。氟与钙作用生成微溶性的氟化钙沉淀物。由于该沉淀物具有一定的溶解度,所以根据溶解度计算以化学计量的氟和钙溶液中仍有4.3×10-4M（相当于8.18 mg/1）的游离氟离子存在。若钙盐过量,因同离子效应,可使游离氟离子浓度进一步降低。但是实验表明对于含氟浓度小于3×10-3M（相当于60mg/1）的溶液并非如此。rohrer（1974）发现,即使水中不含其它干扰成分。

Perlecan,a heparan sulfate proteoglycan,acts as a mechanical sensor for bone to detect external *** of perlecan increases the risk of osteoporosis in patients with Schwartz-Jampel Syndrome(SJS)and attenuates loading4nduced bone formation in perlecan deficient mice(Hypo).Considering that intracellular calcium[Ca2+]i is an ubiquitous messenger controlling numerous cellular processes including mechanotransduction,we hypothesized that perlecan deficiency impairs bone’s calcium signaling in response to *** test this,we performed real-time[Ca2+]i imaging on in situ osteocytes of adult murine tibiae under cyclic loading(8 n,Figure 1).relative to wild type(WT),Hypo osteocytes showed decreases in the overall[Ca2+]i response rate(-58%),calcium peaks(-33%),cells with multiple peaks(-53%),peak magnitude(-6.8%),and recovery speed to baseline(-23%).rnA sequencing and pathway analysis of tibiae from mice subjected to one or seven days of unilateral loading demonstrated that perlecan deficiency significantly suppressed the calcium signaling,ECM-receptor interaction,and focal adhesion pathways following repetitive *** in the endoplasmic reticulum(Er)calcium cycling regulators such as ryr1/ryanodine receptors and Atp2a1/Sercal calcium pumps were identified in Hypo *** together,impaired calcium signaling may contribute to bone’s reduced anabolic response to loading,underlying the osteoporosis risk for the SJS patients.
