Whole-genome genotyping(WGG)stands as a pivotal element in genomic-assisted plant ***theless,sequencing-based approaches for WGG continue to be costly,primarily owing to the high expenses associated with library preparation and the laborious *** prior development of foreground and background integrated genotyping by sequencing(FBI-seq),we discovered that any sequence-specific primer(SP)inherently possesses the capability to amplify a massive array of stable and reproducible non-specific PCR products across the ***,we further improved FBI-seq by replacing the adapter ligated by Tn5 transposase with an arbitrary degenerate(AD)*** protocol for the enhanced FBI-seq unexpectedly mirrors a simplified thermal asymmetric interlaced(TAIL)-PCR,a technique that is widely used for isolation of flanking ***,the improved TAIL-PCR maximizes the primer-template mismatched annealing capabilities of both SP and AD *** addition,leveraging of next-generation sequencing enhances the ability of this technique to assay tens of thousands of genome-wide loci for any *** cost-effective,user-friendly,and powerful WGG tool,which we have named TAIL-PCR by sequencing(TAIL-peq),holds great potential for widespread application in breeding programs,thereby facilitating genome-assisted crop improvement.

Lipoteichoic acids(LTAs)are macroamphiphiles composed of alditol,lipid,phosphate,and carbohydrate *** to their inherent complexity,it is a severe challenge to access LTAs with structural integrity from natural sources for biological or immunological ***,we describe the first total synthesis of Lactococcus garvieae LTA(type II LTA),containing five distinct 1,2-cis gluco/galactopyranosidic linkages,via a novel additive-modulated O-glycosyl trichloroacetimidate preactivation glycosidation *** strategy features(1)high glycosidation yields and excellent 1,2-cis stereoselectivities independent of the donor anomeric configuration,(2)common and inexpensive reagents as promoters and additives,(3)application to standard glycosyl imidate donors without resorting to participating protection,and(4)general application to reactive and less reactive glycosyl ***,via the precise stereocontrolled construction of three galactopyranosidic and two glucopyranosidic bonds on a multigram scale,a series of structurally well-defined LTA molecules were successfully *** evaluation of these type II synthetic LTAs showed a structure–activity relationship in the stimulation of a proinflammatory response.

The inadequacy of tactile perception systems in humanoid robotic manipulators limits the breadth of available robotic ***,we designed a multifunctional flexible tactile sensor for robotic fingers that provides capabilities similar to those of human skin sensing *** sensor utilizes a novel PI-MXene/SrTiO_(3) hybrid aerogel developed as a sensing unit with the additional abilities of electromagnetic transmission and thermal insulation to adapt to certain complex ***,polyimide(PI)provides a high-strength skeleton,MXene realizes a pressure-sensing function,and MXene/SrTiO_(3) achieves both thermoelectric and infrared radiation response ***,via the pressure response mechanism and unsteady-state heat transfer,these aerogel-derived flexible sensors realize multimodal sensing and recognition capabilities with minimal *** can differentiate among 13 types of hardness and four types of material from objects with accuracies of 94%and 85%,respectively,using a decision tree *** addition,based on the infrared radiation-sensing function,a sensory array was assembled,and different shapes of objects were successfully *** findings demonstrate that this PI-MXene/SrTiO_(3) aerogel provides a new concept for expanding the multifunctionality of flexible sensors such that the manipulator can more closely reach the tactile level of the human *** advancement reduces the difficulty of integrating humanoid robots and provides a new breadth of application scenarios for their possibility.

10H2DA improves PCOS through autophagy-mediated mitochondrial fission dependent on the maintenance of calcium homeostasis between endoplasmic reticulum and mitochondria

Polycystic ovarian syndrome （PCOS） is the most principal reason for infertility in reproductive women， but no versatile treatment is feasible. Although trans-10-Hydroxy-2-decenoic acid （10H2DA） exhibits the anti-inflammatory， anti-cancerous and immunomodulatory properties， its role in improving PCOS remains elusive. This present study examined the effects of 10H2DA on the estrous cycle， ovarian morphology， reproductive hormone， glucose and insulin tolerances of PCOS mice， analyzed the binding between 10H2DA and free fatty acid receptor 4 （FFAR4）， and explored the regulation of 10H2DA on the c-Jun N-terminal kinase （JNK）/JUN （Jun proto-oncogene） signaling， cytosolic calcium ion (Ca2+)， endoplasmic reticulum （ER） Ca2+release， mitochondrial Ca2+， mitochondrial function， autophagy， lysosomal acidification， mitochondrial fission， secretion function and apoptosis of ovarian granulosa cells （GCs） using enzyme-linked immunosorbent assay， flow cytometry. western blotting， molecular docking， various fluorescent probes or plasmid， etc. The results found that 10H2DA alleviated the symptom of PCOS mice through improving the secretion dysfunction of GCs and protecting GCs against apoptosis. After binding to FFAR4 and inactivation of JNK/JUN signaling， 10H2DA restricted the release of ER Ca2+through targeting ryanodine receptors （RYRs） which had been identified as the downstream targets of JUN， diminished cytosolic Ca2+accumulation and prevented Ca2+influx into mitochondria via adjusting the mitochondrial calcium uptake 1 （MICU1） and mitochondrial calcium uniporter （MCU）. Concurrently， 10H2DA kept the integrity of mitochondrial function through maintaining mitochondrial Ca2+homeostasis and suppressed the discharge of mitochondrial reactive oxygen species （mtROS） into the cytosol through blocking the mitochondrial permeability transition pore opening to avoid the lipid peroxidation and ameliorate the secretory function and apoptosis of GCs. For cirrhotic refractory ascites,diuretics combined with albumin and vasoactive drugs are the first-line choice for ascites ***,their therapeutic effects are limited,and most refractory ascites do not respond to medication treat-ment,necessitating consideration of drainage or surgical ***-sequently,numerous drainage methods for cirrhotic ascites have emerged,including large-volume paracentesis,transjugular intrahepatic portosystemic shunt,peritoneovenous shunt,automated low-flow ascites pump,cell-free and concentrated ascites reinfusion therapy,and peritoneal catheter *** review introduces the advantages and disadvantages of these methods in different aspects,as well as indications and contraindications for this disease.

The corrosion mechanism of 510L low alloy steel treated by acid-cleaned surface(ACS)and eco-pickled surface(EPS)techniques in three simulated solutions(S0:atmospheric environment;S1:soil environment;S2:industrial environment)and the influence of interaction between different corrosive anions on corrosion were *** results show that the total corrosion rates of samples in three simulated solutions were in order of S2>S0>S1,which is simultaneously correlated with initial corrosion dissolution processes as well as after the formation of corrosion ***3−will inhibit the initial corrosion owing to the formation of films,whereas HSO3−will accelerate the dissolutions of the matrix based on the synergistic action of HSO3−and Cl−.On the other hand,there is no significant difference in corrosion rates between the samples treated by ACS and EPS *** EPS technique that is safe,reusable and environmentally friendly can be further widely used in future work.

Double active sites promoting hydrogen evolution activity and stability of CoRuOH/Co_(2)P by rapid hydrolysis

Cobalt-based phosphides show excellent hydrogen evolution reaction(HER)performance,however,improving the intrinsic activity and stability of it in alkaline electrolyte still remains a ***,CoRuOH/Co_(2)P/CF with heterojunction structure was developed by means of molten salt and rapid hydrolysis(30 s).The OH-from rapid surface hydrolysis of Co_(2)P as a hydrogen adsorption site can facilitate the formation of thin CoRuOH layer as a water dissociation site,which may bring out better synergistic effect for alkaline ***,the covering of CoRuOH can improve the stability of Co_(2)P for *** drives at 100 mA/cm^(2),it only requires overpotential of 81 mV in 1.0 mol/L KOH(25℃).Even at higher current density(1000 mA/cm^(2)),CoRuOH/Co_(2)P/CF can also operate stability for at least 100 *** coupling with NiFe-LDH/IF in a two-electrode system,the voltage of NiFe-LDH/IF(+)||CoRuOH/Co_(2)P/CF(-)at 1000 mA/cm^(2)is merely 1.77 V with 100 h,demonstrating great potential for water *** implementation of this work provides a new strategy and reference for the further improvement of transition metal phosphides as HER electrocatalysts.

Clinical characteristics of acute non-varicose upper gastrointestinal bleeding and the effect of endoscopic hemostasis

BACKGROUND Acute non-variceal upper gastrointestinal bleeding(ANVUGIB)constitutes a prevalent emergency within Gastroenterology,encompassing 80%-90%of all gastrointestinal hemorrhage *** condition is distinguished by its abrupt onset,swift progression,and notably elevated mortality *** To gather clinical data from patients with ANVUGIB at our hospital in order to elucidate the clinical characteristics specific to our institution and analyze the therapeutic effectiveness of endoscopic *** We retrospectively retrieved the records of 532 patients diagnosed with ANVUGIB by endoscopy at our hospital between March 2021 and March 2023,utilizing our medical record *** pertaining to general patient information,etiological factors,disease outcomes,and other relevant variables were meticulously collected and *** Among the 532 patients diagnosed with ANVUGIB,the male-to-female ratio was 2.91:1,with a higher prevalence among ***,43.6%of patients presented with black stool as their primary complaint,while 27.4%had hematemesis as their initial *** admission,17%of patients exhibited both hematemesis and black stool,while most ANVUGIB patients primarily complained of overt gastrointestinal *** routine blood examinations at admission revealed that 75.8%of patients had anemia,with 63.4%experiencing moderate to severe anemia,and 1.5%having extremely severe anemia(hemoglobin<30 g/L).With regard to etiology,53.2%of patients experienced bleeding without a definitive trigger,24.2%had a history of using gastric mucosa-irritating medications,24.2%developed bleeding after alcohol consumption,2.8%attributed it to improper diet,1.7%to emotional excitement,and 2.3%to fatigue preceding the bleeding ***-induced ANVUGIB was more prevalent in the elderly than middle-aged and young individuals,while bleeding due to alcohol consumption showed the opposite ***,diet-related bleeding was more common
