Emotions,Perceptions and Health Behaviors of Adult Congenital Heart Disease Patients during COVID-19 in New York City

Background:Adults with congenital heart disease(ACHD)have increased prevalence of mood and anxiety *** are limited data regarding the influence of the COVID-19 pandemic on the mental health and health behaviors of these ***:The purpose is to evaluate the perceptions,emotions,and health behaviors of ACHD patients during the COVID-19 ***:In this cross-sectional study of ACHD patients,we administered surveys evaluating self-reported emotions,perceptions and health *** regressions were performed to determine the adjusted odds of displaying each perception,emotion and health behavior based on predictor ***:Ninety-seven patients(mean age 38.3 years,46.4%female,85.6%moderate or complex lesion)completed the *** majority of patients reported feeling moderately or very sad(63.1%),and 48.4%of patients identified themselves as feeling moderately or very *** majority of patients perceived their risk of COVID-19 as moderate or *** were more likely to report feeling sad and anxious(95%CI 1.06–10.96,p-value 0.039,and 95%CI 1.44–15.30,p-value=0.012,respectively),and were associated with higher odds of having a perceived increased risk of COVID-19(95%CI 1.33–10.59,p-value 0.012).There was no association between ACHD anatomic or physiologic classification and perceptions,emotions and health ***:Females were more likely to report feeling sad,anxious and an increased risk of COVID-19 in comparison to *** findings indicate the need for mental health support and promotion of health behaviors during the pandemic amongst all ACHD patients,regardless of underlying condition.

Development of ^(18)F-labeled radiotracers for neuroreceptor imaging with positron emission tomography

Positron emission tomography （PET） is an in vivo molecular imaging tool which is widely used in nuclear medicine for early diagnosis and treatment follow-up of many brain diseases. PET uses biomolecules as probes which are labeled with radionuclides of short half-lives, synthesized prior to the imaging studies. These probes are called radiotracers. Fluorine-18 is a radionuclide routinely used in the radiolabeling of neuroreceptor ligands for PET because of its favorable half-life of 109.8 min. The delivery of such radiotracers into the brain provides images of transport, metabolic, and neurotransmission processes on the molecular level. After a short introduction into the principles of PET, this review mainly focuses on the strategy of radiotracer development bridging from basic science to biomedical application. Successful radiotracer design as described here provides molecular probes which not only are useful for imaging of human brain diseases, but also allow molecular neuroreceptor imaging studies in various small-animal models of disease, including genetically- engineered animals. Furthermore, they provide a powerful tool for in vivo pharmacology during the process of pre-clinical drug development to identify new drug targets, to investigate pathophysiology, to discover potential drug candidates, and to evaluate the pharmacokinetics and pharmacodynamics of drugs in vivo.

Until recently,with the exception of coeliac disease,gastroenterologists have not been particularly interested in the role of diet in the management of gastrointestinal ***,patients have always felt that diet must play a part in their symptoms and,in the absence of any medical interest,have turned to alternative dietary practitioners for help,which can often have no evidence ***,with the advent of the FODMAP diet(fermentable oligosaccharides,disaccharides,monosaccharides,and polyols)and the realisation that diet can have a profound effect on the microbiome,medical opinion is now ***,research on the various diets that are now available is often completely *** are carbohydrate binding proteins which are widely distributed in nature and are found in a whole variety of commonly consumed *** seems likely that the exclusion of lectins from the diet could become the next“food fashion”for alternative practitioners to promote,especially as there is some evidence to suggest that certain lectins may be harmful to *** is,therefore,the purpose of this viewpoint to try and stimulate research on the dietary effects of lectins,which is currently minimal,so that we can pre-empt a situation where we are unable to give patients or the public evidence based advice on this topic.

Ti-Zr-Hf-Nb-Ta-Sn high-entropy alloys with good properties as potential biomaterials

Calcium at the Cell Wall-Cytoplast Interface

Attention is given to the role of Ca^2＋ at the interface between the cell wall and the cytoplast, especially as seen in pollen tubes. While the cytoplasm directs the synthesis and deposition of the wall, it is less well appreciated that the wall exerts considerable self control and influences activities of the cytoplasm. Ca^2＋ participates as a crucial factor in this two way communication. In the cytoplasm, a [Ca^2＋] above 0.1 μM, regulates myriad processes, including secretion of cell wall components. In the cell wall Ca^2＋ , at 10μM to 10 mM, binds negative charges on pectins and imparts structural rigidity to the wall. The plasma membrane occupies a pivotal position between these two compartments, where selective channels regulate influx of Ca^2＋ , and specific carriers pump the ion back into the wall. In addition we draw attention to different factors, which either respond to the wall or are present in the wall, and usually generate elevated[Ca^2＋ ] in the cytoplasm. These factors include：（i） stretch activated channels; （ii） calmodulin; （iii） annexins; （iv） wall associated kinases; （v） oligogalacturonides; and （vi） extracellular adenosine 5＇-triphosphate. Together they provide evidence for a rich and multifaceted system of communication between the cytoplast and cell wall, with Ca^2＋ as a carrier of information.

Dielectric confinement on exciton binding energy and nonlinear optical properties in a strained Zn_(1-x_(in))Mg_(x(in))Se/Zn_(1-x(out))Mg_(x(out))Se quantum well

The band offsets for a Zn1-xin Mgxin Se/Zn1-xout Mgxout Se quantum well heterostructure are determined using the model solid theory. The heavy hole exciton binding energies are investigated with various Mg alloy contents. The effect of mismatch between the dielectric constants between the well and the barrier is taken into account. The dependence of the excitonic transition energies on the geometrical confinement and the Mg alloy is discussed. Non-linear optical properties are determined using the compact density matrix approach. The linear, third order non-linear optical absorption coefficient values and the refractive index changes of the exciton are calculated for different concentrations of magnesium. The results show that the occurred blue shifts of the resonant peak due to the Mg incorporation give the information about the variation of two energy levels in the quantum well width.

The pollen tube clear zone: Clues to the mechanism of polarized growth

Pollen tubes usually exhibit a prominent region at their apex called the ＂clear zone＂ because it lacks light refracting amyloplasts. A robust, long clear zone often associates with fast growing pollen tubes, and thus serves as an indicator of pollen tube health. Nevertheless we do not understand how it arises or how it is maintained. Here we review the structure of the clear zone, and attempt to explain the factors that contribute to its formation. While amyloplasts and vacuolar elements are excluded from the clear zone, virtually all other organelles are present including secretory vesicles, mitochondria, Golgi dictyosomes, and the endoplas-mic reticulum （ER）. Secretory vesicles aggregate into an inverted cone appressed against the apical plasma membrane. ER elements move nearly to the extreme apex, whereas mitochondria and Golgi dictyosomes move less far forward. The cortical actin fringe assumes a central position in the control of clear zone formation and maintenance, given its role in generating cytoplasmic streaming. Other likely factors include the tip-focused calcium gradient, the apical pH gradient, the influx of water, and a host of signaling factors （small G-proteins）. We think that the clear zone is an emergent property that depends on the interaction of several factors crucial for polarized growth.

Formation of the multi-subunit oxygen-evolving photosystem II （PSII） complex involves a number of auxiliary protein factors. In this study we compared the localization and possible function of two homolo- gous PSU assembly factors, Psb28-1 and Psb28-2, from the cyanobacterium Synechocystis sp. PCC 6803. We demonstrate that FLAG-tagged Psb28-2 is present in both the monomeric PSII core complex and a PSII core complex lacking the inner antenna CP43 （RC47）, whereas Psb28-1 preferentially binds to RC47. When cells are exposed to increased irradiance, both tagged Psb28 proteins additionally associate with oligo- meric forms of PSII and with PSII-PSI supercomplexes composed of trimeric photosystem I （PSI） and two PSII monomers as deduced from electron microscopy. The presence of the Psb27 accessory protein in these complexes suggests the involvement of PSI in PSII biogenesis, possibly by photoprotecting PSII through energy spillover. Under standard culture conditions, the distribution of PSII complexes is similar in the wild type and in each of the single psb28 null mutants except for loss of RC47 in the absence of Psb28-1. In comparison with the wild type, growth of mutants lacking Psb28-1 and Psb27, but not Psb28-2, was retarded under high-light conditions and, especially, intermittent high-light/dark conditions, emphasizing the physiological importance of PSII assembly factors for light acclimation.

Deletion or underexpression of the Y-chromosome genes CDY2 and HSFY is associated with maturation arrest in American men with nonobstructive azoospermia

Maturation arrest （MA） refers to failure of germ cell development leading to clinical nonobstructive azoospermia. Although the azoospermic factor （AZF） region of the human Y chromosome is clearly implicated in some cases, thus far very little is known about which individual Y-chromosome genes are important for complete male germ cell development. We sought to identify single genes on the Y chromosome that may be implicated in the pathogenesis of nonobstructive azoospermia associated with MA in the American population. Genotype-phenotype analysis of 132 men with Y-chromosome microdeletions was performed. Protein-coding genes associated with MA were identified by visual analysis of a genotype-phenotype map. Genes associated with MA were selected as those genes within a segment of the Y chromosome that, when completely or partially deleted, were always associated with MA and absence of retrievable testicular sperm. Expression of each identified gene transcript was then measured with quantitative RT-PCR in testicular tissue from separate cohorts of patients with idiopathic MA and obstructive azoospermia. Ten candidate genes for association with MA were identified within an 8.4-Mb segment of the Y chromosome overlapping the AZFb region. CDY2and HSFYwere the only identified genes for which differences in expression were observed between the MA and obstructive azoospermia cohorts. Men with obstructive azoospermia had 12-fold higher relative expression of CDY2transcript （1.33__.0.40 vs. 0.11＋_0.04; P=O.O003） and 16-fold higher expression of HSFYtranscript （0.78__.0.32 vs. 0.05_0.02; P=O.O005） compared to men with MA. CDY2 and HSFYwere also underexpressed in patients with Sertoli cell only syndrome. These data indicate that CDY2and HSFYare located within a segment of the Y chromosome that is important for sperm maturation, and are underexpressed in testicular tissue derived from men with MA.
