Relationship between p-catenin expression and epithelial cell proliferation in gastric mucosa with intestinal metaplasia

AIM： To investigate β-catenin expression in patients with intestinal metaplasia, and to look for a possible relationship between β-catenin expression and either epithelial proliferation values or Helicobacter pylori （ H pylori） infection.METHODS： Twenty patients with complete type intestinal metaplasia were studied. β-Catenin expression and epithelial cell proliferation in antral mucosa were assessed using an immunohistochemical analysis. H pylori infection was detected by histology and a rapid urease test.RESULTS： Reduced β-catenin expression on the surface of metaplastic cells was detected in 13 （65%） out of 20 patients. Moreover, in eight （40%） patients intranuclear expression of β-catenin was found. When patients were analyzed according to H pylori infection, the prevalence of both β-catenin reduction at the cell surface and its intranuclear localization did not significantly differ between infected and uninfected patients. Cell proliferation was higher in patients with intranuclear β-catenin expression as compared to the remaining patients, although the difference failed to reach the statistical significance （36±8.9 vs 27.2±11.4, p= 0.06）. On the contrary, a similar cell proliferation value was observed between patients with reduced expression of β-catenin on cell surface and those with a normal expression （28.1±11.8 vs 26.1±8.8, p= 0.7）.H pylori infection significantly increased cell proliferation （33.3±10.2% vs 24.6±7.4% , respectively, p= 0.04）.CONCLUSION： Both cell surface reduction and intranuclear accumulation of β-catenin were detected in intestinal metaplasia. The intranuclear localization of β-catenin increases cell proliferation. H pylori infection does not seem to play a direct role in β-catenin alterations, whilst it significantly increases cell proliferation.

SAp Expression in Candida albicans Strains Isolated from Mexican patients with Vaginal Candidosis

To determine the frequency and expression of the ten SAp (secreted aspartyl protease) genes in a group of Candida albicans strains isolated from Mexican women suffering from vaginal candidosis, a group of 264 women (age 18 - 57 years) with vaginal infections, predisposed by diabetes mellitus or contraceptive consumption, were evaluated. C. albicans was identified using pCR to amplify the rRNA internal transcribed spacer regions ITS1 and ITS2. The presence of the SAp genes was determined using conventional pCR, and their expression levels were determined using real-time pCR after the C. albicans strains had been grown in reconstituted human vaginal epithelium (RHVE). C. albicans was identified in the samples from 50 women (18.9%). The genotyping frequencies of the SAp genes were as follows: SAp1, 94%;SAp2, 98%;SAp3, 80%;SAp4, 100%;SAp5, 100%;SAp6, 100%;SAp7, 63%;SAp8, 96%;SAp9, 70%;and SAp10, 88%. The most frequently expressed genes in the strains harboring all of the genes were SAp1, 90%;SAp2, 90%;SAp3, 90%;SAp4, 100%;SAp5, 90%;SAp6, 90%;SAp7, 100%;SAp8, 90%;SAp9, 100%;and SAp10, 100%. SAp genes were expressed in the RHVE, suggesting that the Sap proteins play an important role in the pathogenesis of infection.

Abstract The phylogenetic relationship of lecanoroid lichens is studied using two data sets:1)a 2-locus data set including 251 OTUs representing 150 species,and 2)a 6-locus data set with 82 OTUs representing 53 *** genus Lecanora as currently circumscribed is shown to be highly polyphyletic and several genera,including Adelolecia,Arctopeltis,Bryonora,Carbonea,Frutidella,Lecidella,Miriquidica,palicella,protoparmeliopsis,pyrrhospora,and Rhizoplaca are nested within Lecanora sensu lato.A core group of Lecanora is supported as monophyletic and includes species of the ***,***,and *** groups,and a core group of the *** *** monophyletic clades that are well supported in our analyses and well characterized by phenotypical characters are accepted here:1)Myriolecis to accommodate the Lecanora dispersa group and Arctopeltis;2)protoparmeliopsis for the *** group;and 3)Rhizoplaca is emended to include three placodioid taxa previously classified in Lecanora(***,***),whereas *** and *** are excluded from *** latter is transferred into *** is strongly supported as a monophyletic *** studies indicate the presence of additional clades of species currently placed in Lecanora sensu lato that warrant taxonomic recognition but additional data will be necessary before the circumscription of these entities is fully understood.37 new combinations are proposed into the genera Myriolecis(30),protoparmeliopsis(2),and Rhizoplaca(5).

Functional traits shape size-dependent growth and mortality rates of dry forest tree species

Aims Functional traits have emerged as an important tool to evaluate plant ***,the environmental conditions and ecological pressures that plants face change with their size,and the relationship between traits and plant performance should therefore be size-dependent,which has rarely been *** Here,we evaluated over a broad range of tree sizes the interspe-cific relationship between tree growth and mortality and eight func-tional stem,leaf and seed *** did so across 59 tree species in Brazilian dry forests and evaluated whether the relationships found for wet forest types in the literature also hold for dry forests,where water rather than light might limit tree *** Findings We indeed found a strong size-dependent relationship between demographic rates and functional *** small sizes,when trees are in the shaded understory,species with functional trait values that enhance light capture or shade tolerance(*** maximum adult stature,taller heights,wider crowns,higher seed mass)have higher growth and/or lower mortality rates(MR).This relationship disappears at larger sizes when trees attain better light conditions in the *** adaptations play only a role at larger tree sizes;once trees are in the dry and exposed canopy,species with higher wood density(an indicator of cavitation resistance)have lower *** study shows that both drought and shade adaptations are important in this dry forest,and that the relationships between func-tional traits and plant performance changes with plant *** size should therefore explicitly be included as an axis of variation in functional analyses,to better understand the relationship between functional traits and demographic rates.

Innovate combination of sevoflurane dilution in dimethyl sulfoxide: A stability study by gas chromatography and nuclear magnetic resonance

To investigate physicochemical stability of sevofuranein dimethyl sulfoxide using gas chromatography with a fame ionization detector and nuclear magnetic reso-nance （NMR）.METHODSUndiluted sevoflurane, plus dilutions 1：2, 1：5, 1：10, 1：25, and 1：50 in dimethyl sulfoxide were prepared in a vertical laminar fow cabinet class Ⅱ type B and stored at different temperatures （23 ℃, 6 ℃, and -10 ℃） for 45 d. Sterile 1 mL polypropylene amber syringes to minimize light degradation, caps and needles were used. The presence of sevofurane and its degradation products in the samples was determined by gas chroma-tography with flame ionization detector （260 ℃, 40min）, and by 1H, 19F, and proton-decoupled 19F nuclearmagnetic *** The gas chromatography analysis showed sevofluraneand dimethyl sulfoxide （DMSO） retention times were 2.7and 13.0 min, respectively. pure DMSO injection into thecolumn resulted in two additional peaks at 2.1 and 2.8min. The same sevofurane peak at 2.7 min was observedin all the dilutions at -10 ℃, 4 ℃ and 25 ℃. The NMRspectra showed signals consistent with the sevoflurane structure in all the dilutions at -10 ℃, 4 ℃ and 25 ℃. In the 1H spectrum, two signals corresponding to the sevoflurane molecule were observed at 5.12 and 4.16 parts per million （ppm5）. In the 19F-NMR spectrum, two signals were observed at -76.77 ppm and -157.13 ppm. In the 19F NMR CpD, two signals were observed at -76.77 ppm and -157.13 ppm. The first one showed a doublet （JF-F = 3.1 Hz） which integrated by six fluorine nuclei from the hexafluoro-isopropyl group. The second signal was integrated by a fuorine atom and showed a septuplet （JF-F = 3.1 Hz）.CONCLUSIONThis study shows that different concentrations ofsevofurane in dimethyl sulfoxide retain their chemicalcomposition after exposure to different temperaturesfor a period of 45 d.

【目的】评价抗菌肽(Antimicrobial peptides,AMps)对红姑鱼(Sciaenops ocellatus)幼鱼生长性能和血清激素水平的影响。【方法】在基础饲料中添加不同剂量的抗菌肽(0,0.1,0.3和0.5g/kg)作为试验饲料(即A0,A100,A300和A500组),饲喂平均体质量为(1.57±0.17)g/尾的红姑鱼8周,测定各组红姑鱼生长性能、血清激素(GH、T3、T4和IGF-I)水平,A0和A300组红姑鱼停饲后不同时间点的血清激素水平及肌肉和肝脏中IGF-I mRNA表达丰度。【结果】A300和A500组红姑鱼增重率、饲料效率和蛋白效率都显著高于A0组(pppp>0.05)。养殖4,6,8周抗菌肽对血清中GH水平无显著影响(p>0.05);A300和A500组血清T3、T4和IGF-I含量显著高于A0组(ppp<0.05)。【结论】饲料中添加抗菌肽0.3g/kg可促进红姑鱼的生长,提高饲料效率,增加含肉率。抗菌肽与血清GH的分泌无显著相关性,但可提高血清IGF-I水平及IGF-I mRNA在肌肉和肝脏中的表达量。
