Hepsin,a transmembrane serine protease abundant in renal endothelial cells,is a promising therapeutic target against several cancers,particularly prostate *** is involved in the release and polymerization of uromodulin in the urine,which plays a role in kidney stone *** this work,we design new potential hepsin inhibitors for high activity,improved specificity towards hepsin,and promising ADMET *** ligands were developed in silico through a novel hierarchical *** pipeline explicitly accounts for off-target binding to the related serine proteases matriptase and HGFA(human hepatocyte growth factor activator).We completed the pipeline incorporating ADMET properties of the candidate inhibitors into custom multi-objective optimization *** ligands designed show excellent prospects for targeting hepsin via the blood stream and the urine and thus enable key experimental *** computational pipeline proposed is remarkably costefficient and can be easily adapted for designing inhibitors against new drug targets.

随着遥感技术的发展,使用现代遥感技术工具来分析和监测生态空间中的空间现象需要更多的关注。土壤,作为全球生态系统的组成部分,是构成地球的最上层,是植物、动物和其他生物的家园,为了保持其可持续性功能,需要不断地进行研究。土壤结构参数与太阳能(光)能量相互作用产生种类型的反应,分别是吸收、反射和透射。在土壤参数与光能相互作用的三种反应中,反射在遥感分析和估算土壤固有性质方面起着至关重要的作用。土壤颗粒大小是土壤结构参数中的一个协调元素,它决定了土壤的化学和生物学特性的相互作用。在土壤中的其他的土壤参数的性质,取决于被调查土壤的粒度分布,这就导致了土壤颗粒大小所反映的能量程度的变化。土壤表面的BRF为遥感土壤特性的评估提供了基础,有助于土壤资源的有效利用。本研究主要分析不同角度测量的不同土壤颗粒大小的BRF分布。由于土壤表面反射是环境监测、评价和精准农业的重要组成部分。本研究将有助于解释和理解光能的复杂相互作用及其对土壤参数(特别是土壤颗粒尺寸)的影响,以及从土壤表面测量的反射能量如何进行土壤有效分类和表征。为了研究不同土壤种类和土壤粒径对反射的影响,我们选择了三种不同类型的土壤样本。其中两个土壤样本为黑土、栗钙土,包括14个粒径范围,从2毫米-0.027 mm依次降序筛分得到。另一个土壤样本为风沙土,包含6个粒径范围,从0.525 mm到0.075mm依次降序筛分得到。样品的反射光谱测量使用东北师范大学实验室角谱仪系统(NENULGS)进行,在可见和近红外波长范围内进行探测。同时,测量了不同视角双向反射系数(BRFs)。之后,在选定波长下,将土壤表面BRF和ARF的非朗伯现象与之前的研究进行比较,以验证本研究测量的准确性。并定量分析了各粒径土壤在可见光和近红外波段最低点方向上反射的平均BRF,以实现其他角度检测数据的标准化。此外,研究发现,与之前的研究一样,反射峰和最小反射的所有粒径的BRF都在前向散射方向。接着,计算了3个土样0.3mm粒径的多角度ARF相对差值。观测到最大的相对ARF差出现在后向散射方向,最小的相对ARF差出现在前向散射方向。黑土颗粒大小在可见光和近红外波段均表现出最大和最小的ARF相对差异。此外,3个样品的粒径变化表明,土壤表面各向异性特征是检验和分析土壤粒径变化的重要参数之一。我们对土壤样品的有效粒径与最低点的BRF进行相关分析,并在所有视角下进行评估。结果表明,风沙土在各观测角度下的平均线性关系最强(R=0.84),栗钙土的线性关系次之(R=0.64),黑土的线性关系最弱(R=0.41)。黑土出现的弱线性关系主要与有机质含量较高有关。风沙土的自然对数相关系数(R)最弱。基于风成沙土最小粒径分布假设,进一步证实了这种弱关系。栗钙土和黑土的自然对数关系分别最强和较强。结果表明,粒径分布的体积决定了使用BRF进行有效定量检测土壤表面的统计工具类型。本研究所采用的方法有助于理解不同视角下不同土壤表面的反射光能量。该方法的合理应用对土壤的特征、制图和分类具有重要意义,有助于土壤的有效利用。

Influence of BA and IBA or NAA Combinations on Micropropagation of Cryptolepis sanguinolenta

The study being the first of its kind established an efficient protocol for micropropagation of Cryptolepis sanguinolenta, an important endangered medicinal plant species, used in the treatment of Malaria. For shoot induction, semi hard wood nodal segments were maintained on MS (Murashige and Skoog) nutrient medium supplemented with MS vitamins, 30 g/L sucrose, 3% gelrite and various auxin and cytokinin combinations. Treatments involved 6-benzyladenine (BA) at 0.5, 1.0, 1.5, 2.0, 2.5 or 3.0 mg/L in combination with 0.1 mg/L Indole 3-butyric acid (IBA) or Naphthaleneacetic acid (NAA). Control consisted of hormone free MS medium. BA and IBA combinations were found to be more efficient in shoot regeneration than the BA and NAA combinations. Cultures maintained on MS medium supplemented with 3 mg/L BA, in combination with 0.1 mg/L IBA recorded the highest shoot induction (100%), mean shoot length (1.28 cm) and mean number of nodes per explant (2.5). This, however, did not differ significantly from cultures maintained on 2 or 2.5 mg/L BA and 0.1 mg/L IBA supplemented MS medium. regenerated shoots were transferred onto different media for root induction. Treatments consisted of full strength MS medium augmented with either 0.5 or 0.1 mg/L IBA, hormone free half strength MS medium and half strength MS medium augmented with 0.01 mg/L IBA. After six weeks of culture, no rooting was recorded in all treatments with the exception of half strength MS medium supplemented with 0.01 mg/L IBA, which recorded 60% rooting. regenerated plantlets were successfully weaned and established in the greenhouse.

Shea nuts play an important role in food security for rural folks within sub-Sahara Africa, serving as the main source of income for many people living in Northern Ghana. Unfortunately, the full economic potential of the Sheanut tree has not been fully realized due to the difficulty involved in its domestication. This difficulty in vegetatively propagating sheanut trees has greatly hindered its cultivation and the realization of its true economic potential. Two experiments were conducted to investigate the effects of rooting media and varying indole 3-butyric acid (IBA) concentrations on adventitious root formation in cuttings taken from coppiced sheanut trees. results indicated that 3000 ppm produced significantly (p 0.05) better rooting (57.5%) than 5000 ppm (30%), 7000 ppm (45.0%) and the control (7.5%). Although the levels of soluble sugars (SS) and total free phenols (TFp) in the cutting were significantly (p 0.05) higher at the end of the experiment (after IBA treatment) comp.red to the start (prior to IBA treatment), the SS and TFp trends observed did not clearly explain the rooting differences found between the IBA levels investigated. Callus formation was significantly (p 0.05) higher (35.0%) in the control (no IBA). Generally, callus formation decreased with increasing IBA concentration. In the rooting media experiment, rooting was significantly (p 0.05) higher in the rice husk medium (35.0%) comp.red to that in the palm fiber (18.3%), saw dust (14.1%) and top soil (16.7%) media.

In spite of the economic potential of the sheanut tree (Vitellaria paradoxa), its domestication still has not been achieved due to the long juvenile period of seed propagated plants and the absence of a reliable vegetative propagation method. Three experiments were conducted by using a modified container layering technique to investigate the effects of season (rainy and dry season), light treatments (etiolation and natural light) and indole 3-butyric acid (IBA) on root formation in juvenile and mature sheanut trees. The effect of light treatments on the internal conditions such as level of sugars (soluble, insoluble and total sugars) and total free phenols in layered sheanut shoots was also considered. Rooting was generally lower in the mature trees (27.9%) comp.red to that in juvenile 4 year-old plants (40.9%). Etiolation increased the levels of total sugars and phenols in shoots of 4-year-old plants and mature fruit bearing trees, but this increase did not significantly enhance rooting in both the juvenile and mature sheanut trees. The container layering technique holds promise as a method for the asexual propagation of sheanut planting material. Roots formed using this method looked healthy and were vertically orientated giving layered shoots a better chance at survival.

A growing number of studies are now emphasizing the critical importance of learning and knowledge accumulation for firm-level competitiveness. Despite the growing awareness, relatively fewer project-based firms have institutionalized mechanisms to systematically capture new project knowledge and re-use it to improve the execution of subsequent projects. The peculiarity of projects presents unique challenges that make the cognitive approach to learning difficult to implement. As such, researchers are recommending the social constructivist perspective of learning as the most viable strategy for cultivating learning within and across projects. However, scant work has been undertaken from this sociological perspective to analyze how temporary organizations manage knowledge arising from and relating to projects. From this standpoint, the aim of this paper is to discuss the learning mechanisms of construction firms. The study adopted a quantitative strategy by employing a questionnaire survey into the learning practices of construction projects in Ghana. Drawing on preliminary findings from the literature, the study proposes a model for cultivating learning within projects from the social constructivist viewpoint. In the model, project management practitioners can purposefully nurture or structure a project learning activity through four mechanisms viz.: institutionalization, externalization, socialization and internalization. The proposed model is subsequently validated in an empirical study into the learning practices during the implementation of construction projects in Ghana. Based on the empirical results, it seems that knowledge sharing and transfers through the four aforementioned learning mechanisms proposed by the model are highly regarded within project management practice in Ghana.
