Signal transducer and activator of transcription 3 (STAT3) is a recently characterized transcription factor which is essential to liver regeneration. We have previously reported that hepatic stimulator substance (HSS), a novel growthpromoting substance, phosphorylated the epidermal growth factor (EGF) receptors and activated downstream ras-MAp kinase (extracellular signal-regulated kinases, ErK1/2) cascade. However, whether HSS signal is related to STAT3 pathway remains unclear. The present study is aiming to explore the regulatory effect of activation of ErK1/2 evoked by HSS on STAT3 phosphorylation and STAT3 signaling. Human hepatoma cell line HepG2 was stably transfected with HSS cDNA and HSS expression was measured by Northern blot. The results showed that the transfection of HSS into HepG2 resulted in remarkable increase in cellular proliferation as compared with the non-transfected cells, and it was further proved that the cellular proliferation in the HSS-transfected cells was related to ErK1/2 a

BACKGrOUND: Alcoholic hepatitis (AH) is a necrotizing inflammatory process caused by alcoholic liver injury. It carries a significant short-term mortality. The management of AH is challenging. Although corticosteroids have been demonstrated to exert anti-inflammatory and antifibrotic effects, their efficacy for the treatment of AH remains debatable. DATA SOUrCES: A literature search was performed of MEDLINE, ScienceDirect, SpringerLink and Wiley InterScience using the key words 'alcoholic hepatitis', 'alcoholic liver disease', and 'corticosteroids'. The available data reported in the relevant literature were analyzed. rESULTS: More than 17 controlled trials and at least 13 meta-analyses have reported the efficacy of corticosteroids in the treatment of AH in the past 40 years. Many were poorly designed and used different inclusion/exclusion criteria, making it difficult to reach a consensus. In this review, we summarized all the controversial data in the past decade and analyzed the potential causes for the varying therapeutic effects of corticosteroids in AH. The focus of the controversy has changed from 'whether steroids are beneficial or harmful for AH patients' to 'how to accurately identify responders to steroids early and rationalize corticosteroid treatment'. An early response to glucocorticoids, as determined by calculating the Lille score after 7 days of treatment, has been shown to be a clinically useful indicator. Moreover, down-regulation of steroid sensitivity, risk of infection, and a rational therapeutic strategy of corticosteroids in AH patients are all crucial for therapeutic ***: An early and accurate determination of steroid sensitivity is important. Besides, we need to overcome the down-regulation of steroid sensitivity, reduce the infection risk and rationalize the therapeutic strategy of corticosteroids. A fresh perspective is needed on the use of corticosteroids in AH patients.

To investigate the potential of cardiomyogenic differentiation of rat bone marrow stromal cells (MSCs), they were exposed to 5-azacytidine tretments (single/repeat) at varying concentrations (3, 5, 10 μmol/L) and the fates of the cells were analyzed by immunocytochemistry, western blot and the reporter gene of enhanced cyan fluorescent protein (ECFp) under the control of ventricular myosin light chain 2 (MLC2v) promoter. MSCs were also cocultured with cardiomyocytes for periods up to 16 days, the expression of cardiac myosin heavy chain (MHC) and troponin I (Tn I) proteins was analyzed. After the induction with 5-azacytidine, neither spontaneously beating cells nor myotubes were found; MHC and Tn I proteins were also undetectable and no ECFp-positive MSCs were detected. But when cocultured with cardiomyocytes, spontaneously contracting MSCs were observed and cardiac specific proteins could be detected. The results proved that the novel effects of 5-azacytidine on the cardiomyogenic differentiation of MSCs should be questioned and a direct intercellular communication with cardiomyocytes is necessary for MSCs to differentiate into cardiomyocytes. The second envelope glycoprotein （E2） of hepatitis C virus has been shown to bind human target cells and has become a major target for the development of anti-HCV vaccines. Anti-E2 antibodies have been suggested to be of clinical significance in diagnosis, treatment and prognosis of hepatitis C. However, large-scale expression and purification of E2 proteins in mammalian cells is difficult. AIM: To investigate the possible relationships between alterations in circadian rhythm of melatonin, cortisol andbronchial asthma. METHODS: Salivary melatonin and cortisol were measured simultaneously by radioimmunoas-say in 10 mild intermittent or persistent patients, 11 moderate-to-severe persistent asthma patients, and 15 controlsubjects. Twelve salivary samples were collected in a series during a 24-h period in each subject. rESULTS: Theresults showed overall lower levels of salivary melatonin in asthma patients compared with control subject(pplitude, peak-level, and baseline of salivary melatonin were significantly lower in mild intermit-tent or persistent (pprate-to-severe persistent asthma patients (ppared withcontrol group. The 24-h mean level of salivary cortisol was greatly lower and the acrophase was markedly delayedin patients with mild intermittent or persistent asthma (prate-to-severe persistent asthma (pppared with control subject. CONCLUSION: Disordered circadian rhythms of salivary melato-nin and cortisol were found in asthma patients, which may be involved in the pathogenesis of bronchial asthma.

Antioxidative effect of propofol during cardiopulmonary bypass in adults

AIM: To investigate the antioxidative potential of propofol (an intravenous anesthetic with a chemical structure similarto phenol-basedfree radicalscavengers suchas vitaminE) during cardiopulmonary bypass(CpB). METHODS: Thirty adult patients referred for elective cardiac procedure with CpB were included and randomly allocated to a propofol group or a control group. patients in the propofol group received propofol (0.1 mg·kg-1·min-1) intrave- nously for anesthesia maintenance, whereas those allocated to the control group received fentanyl 10 μg/kg intrave- nously and inhaled enflurane (1 %-1.5 %). Blood samples were collected at 7 time points: before the start of CpB, at 30 and 60 min of CpB, at the conclusion of CpB, 10 min after the administration of protamine, and 12 and 24 h after the cessation of CpB. plasma levels of free F2-isoprostanes (sensitive markers of free radicals production) and complement C5a were determined by mass-spectrometricassay and enzyme immunoassay, respectively. Neu- trophil adhesion to endothelial cells was observed at ×200 magnificationunder a light microscope. rESULTS: Levels of F2-isoprostanes, complement C5a and neutrophil adhesion rate increased significantly during and after CpB in both groups. There were significantly higher levels of F2-isoprostanes, C5a, and more neutrophils adhering to endothelial cells in the control group than those in the propofol group, respectively. CONCLUSION: Cardio- pulmonary bypass is associated with a great production of damaging free radicals. propofol may be beneficial both as an anesthetic and as a potent free radical scavenger in patients presenting pathologies associated with free radical reactions during CpB.

BACKGrOUND:Whether a major liver resection is safe has been judged mainly from the patient’s hepatic ***,a safe limit for liver resection does not exist *** study aimed to construct a new scoring system as a guide to determine a safe limit for liver resection and avoid liver dysfunction after ***:Eighty-six patients with hepatocellular carcinoma who had undergone hepatectomy in west china hospital from March 2007 to June 2010 were *** patients were classified according to the levels of total bilirubin after hepatectomy and the parameters in the perioperative period were *** operating characteristic (rOC) analysis was made to assess the liver function compensatory (LFC) value to predict liver dysfunction of the patients after *** value is defined as the preoperative KICG value×22.487+standard remnant liver volume (SrLV)×***:patients were classified into group Ⅰ (normal group,n=69) and group Ⅱ (with total bilirubin >85.5 μmol/L for 7 days after hepatectomy,n=17) based on the levels of total bilirubin after *** II was further divided into two subgroups:recovered subgroup (n=14) and fatal subgroup (n=3).There were no significant differences in preoperative data or intraoperative findings except the indocyanine green test parameters (KICG and ICG r15) and *** analysis showed that the sensitivity and specificity of an LFC value ≤13.01 were 94.1% and 82.6% respectively for predicting liver dysfunction of the patients after ***:The LFC value appears to be a good predictor of postoperative liver dysfunction in patients who undergo hepatectomy for *** expected LFC value of 13.01 seems to be a safe limit for liver resection.

The description and differentiation of the so-called "Cold" and "Hot" natures, the primary "Drug Naure" of Chinese medicine, is the focus of theoretical research. In this study, the divergency between the "Cold" and the "Hot" natures was investigated through examining the temperature tropism of mice affected by Coptis chinensis Franch and its processed materials by using a cold/hot plate differentiating technology. After exposure to C. chinensis Franch, the macroscopic behavioral index of the remaining rate (rr) on a warm pad (40℃ ) significantly increased (propism. The internal indexes of adenosine triphosphatase (ATpase) activity and oxygen consuming volume decreased significantly (ppability of energy metabolism. This external behavior of Hot tropism might reflect the internal Cold nature of C. chinensis Franch. However, the processed materials of C. chinensis Franch exhibited a different Cold nature in temperature tropism compared with crude C. chinensis Franch (CC): the Cold nature of bile-processed C. chinensis Franch (BC) enhanced while the ginger-processed C. chinensis Franch (GC) changed inversely. The changing sequence was consistent with the theoretical prognostication. It is indicated that the external Cold & Hot natures of Chinese medicine may possibly reflect in an ethological way for the changes of animal’s temperature tropism which might be internally regulated by the body’s energy metabolism.
