Radiotherapy and the cellular DNA damage response: current and future perspectives on head and neck cancer treatment

Incidences of head and neck squamous cell carcinoma(HNSCC)have been on the rise in the last few decades,with a significant risk factor being human papillomavirus(HpV)type-16/18 infection,particularly in the development of oropharyngeal ***(RT)is an important treatment modality for HNSCC,where it promotes extensive cellular DNA damage leading to the therapeutic *** has been well-established that HpV-positive HNSCC display better response rates and improved survival following RT compared to HpV-negative *** differential radiosensitivity has been largely associated with altered cellular DNA damage response mechanisms in HpV-positive HNSCC,and particularly with the signaling and repair of DNA double strand ***,other factors,particularly hypoxia present within the solid cancer,have a major impact on relative ***,recent approaches aimed at enhancing the radiosensitivity of HNSCC have largely centered on targeting key proteins involved in DNA repair,DNA damage checkpoint activation,and hypoxia *** studies have utilised in vitro and in vivo models of HpV-positive and HpV-negative HNSCC and examined the impact of specific inhibitors against the targets in combination with radiation in suppressing HNSCC cell growth and ***,accumulating evidence has shown that targeting enzymes including poly(ADp-ribose)polymerase,ataxia telangiectasia and Rad-3 related,DNA-dependent protein kinase catalytic subunit,and checkpoint kinase 1 can radiosensitise HNSCC cells which should be taken forward in further preclinical studies,with the goal of optimizing the future effective RT treatment of HNSCC.

Highly efficient GaAs solar cells by limiting light emission angle

In a conventional flat plate solar cell under direct sunlight,light is received from the solar disk,but is re-emitted *** isotropic emission corresponds to a significant entropy increase in the solar cell,with a corresponding drop in ***,using a detailed balance model,we show that limiting the emission angle of a high-quality GaAs solar cell is a feasible route to achieving power conversion efficiencies above 38%with a single *** highest efficiencies are predicted for a thin,light trapping cell with an ideal back reflector,though the scheme is robust to a non-ideal back *** with a conventional planar cell geometry illustrates that limiting emission angle in a light trapping geometry not only allows for much thinner cells,but also for significantly higher overall efficiencies with an excellent rear ***,we present ray-tracing and detailed balance analysis of two angular coupler designs,show that significant efficiency improvements are possible with these couplers,and demonstrate initial fabrication of one coupler design.

A rare case of failed healing in previously burned skin after a secondary burns

Background:patients presenting with large surface area burns are common in our practice;however, patients with a secondary large burn on pre-existing burn scars and grafts are rare and not reported. Case presentation:We report on an unusual case of a patient sustaining a secondary large burn to areas previously injured by a burn from a different mechanism. We discuss the potential implications when managing a case like this and suggest potential biological reasons why the skin may behave differently. Our patient was a 33-year-old man who presented with a 5%TBSA burn on skin scarred by a previous 40%total body surface area (TBSA) burn and skin grafts. Initially assessed as superficial partial thickness in depth, the wounds were treated conservatively with dressings;however, they failed to heal and became infected requiring surgical management. Conclusions:Burns sustained in areas of previous burn scars and grafts may behave differently to normal patterns of healing, requiring more aggressive management and surgical intervention at an early stage.

AN ECOSYSTEM MODEL ANALYSIS OF THE EFFECT OF MINE TAILINGS ON THE EUpHOTIC ZONE OF A pELAGIC ECOSYSTEM

A computer analysis was performed on experimental results obtained when mine tailings were added to seawater. The Mixed Upper-Layer Ecotrophie Simulation (MULES) model was tested by changing the extinction coefficient and the abundance of heterotrophic zooflagellates. Increasing the extinction coefficient resulted in a delay of phytoplankton growth, an increase in zooplankton standing stock and better growth of autotrophic flagellates compared with diatoms. Zooflagellates in the ecosystem influence the growth of zooplankton; secondary production by zooplankton was markedly depressed at low levels of zooflagellates. These results are believed to be of general significance for the diagnosis of suspended sediment effects on planktonic ecosystems.

The actin-bundling protein Fascin-1 modulates ciliary signalling

primary cilia are microtubule-based cell organelles important for cellular communication. Since they are involved in the regulation of numerous signalling pathways, defects in cilia development or function are associated with genetic disorders, collectively called ciliopathies. Besides their ciliary functions, recent research has shown that several ciliary proteins are involved in the coordination of the actin cytoskeleton. Although ciliary and actin phenotypes are related, the exact nature of their interconnection remains incompletely understood. Here, we show that the protein BBS6, associated with the ciliopathy Bardet–Biedl syndrome, cooperates with the actin-bundling protein Fascin-1 in regulating filopodia and ciliary signalling. We found that loss of Bbs6 affects filopodia length potentially via attenuated interaction with Fascin-1. Conversely, loss of Fascin-1 leads to a ciliary phenotype, subsequently affecting ciliary Wnt signalling, possibly in collaboration with BBS6. Our data shed light on how ciliary proteins are involved in actin regulations and provide new insight into the involvement of the actin regulator Fascin-1 in ciliogenesis and cilia-associated signalling. Advancing our knowledge of the complex regulations between primary cilia and actin dynamics is important to understand the pathogenic consequences of ciliopathies.

关于地震波通过时触发的地震数与主震引起的静态应力变化触发的地震数的相对比例问题,一直存在着争议。静态应力变化应该对地震发生概率有长期影响,而由地震波通过引起的动态应力变化则不会。两种机制都应该会提高某些地方的地震活动率,但只有静态应力变化计算可预测地震活动率降低的影区。因此,鉴别出主震后受到抑制的地震活动就可判断静态应力变化过程。我们注意到原则上静态应力变化理论只能预测影区内特殊震源机制地震受到抑制,而非总体地震发生率的降低。因此,可用主震前、后因一定机制类型地震受到抑制而引起的平均震源机制的变化来表示应力影区的特征。我们考察了从哈佛矩张量(CMT)目录中抽取的119个MS≥7主震的±2°半径范围内、主震发生前后5年内的平均震源机制。仅在两例中发现由地震受到抑制引起的显著的平均震源机制变化。不过,通过数据叠加,我们能从统计上分辨出特定震源机制的主震后地震受到了显著抑制。这表明,虽然静态应力影区信号微弱,但它们的确出现在全球目录中。

Three-dimensional numerical modeling of single geocellreinforced sand

This paper summarizes the development of a three-dimensional numerical model for analyzing single geocell-reinforced *** this model,the infill soil was modeled using the Duncan-Chang model,which can simulate non-linearity and stress-dependency of *** was modeled using linearly elastic plate elements,which can carry both bending and membrane stresses.A linear interface stress-strain relationship with a MohrCoulomb yield criterion was adopted to model the interface friction between the geocell wall and the *** modeling the geocell and the soil separately,the interaction between the soil and the geocell can be accurately *** verify this model,a plate load test was conducted in the laboratory,in which a 12-cmthick sand layer reinforced by a single geocell was subjected to a vertical load from a circular steel *** load-displacement curves and the horizontal tensile strain of the geocell were recorded during the test.A numerical model was created according to the setup of the load *** numerical results compared reasonably well with the test data.

patient reported outcome measures(pROMs)provide a valuable means of measuring outcomes subjectively from a patient's perspective,facilitating the assessment of service quality across healthcare providers,and assisting patients and clinicians in shared decision *** primary aim of this systematic review was to critically appraise all historic studies evaluating patient reported quality of life,in adult patients undergoing laparoscopic cholecystectomy for symptomatic *** secondary aim was to perform a quality assessment of cholecystectomy-specific pROM-validation studies.A literature review was performed in pubMed,Google ScholarTM,the Cochrane Library,Medline,CINAHL,EMBASE and psychINFO databases up to September *** characteristics,pROM-specific details and a bias assessment were summarised for non-validation studies.A COnsensus-based Standards for the selection of health Measurement INstruments(COSMIN)analysis was performed to assess the methodological quality of identified pROM-validation *** one studies were found to evaluate health-related quality of life(HRQoL)after laparoscopic *** 94.1%of these studies included pROMs as a primary outcome measure,provided level 1 evidence through randomised controlled trials(RCTs).There was significant variation in the selection and reporting of pROMs,with no studies declaring patient involvement in pROM selection,and 88.2%of studies failing to document the management of missing data points,or non-returned surveys(33.3%).In the 6 pROM-validation studies identified,only 5 psychometric properties were evaluated,the findings of which were limited due to the small number of *** systematic review identifies a lack in consistency of study design and pRO reporting in clinical *** an increasing number of studies are being performed to evaluate pROs,a lack of adherence to existing pRO administration and reporting guidelines is continuing to negatively affect st
