Dynamic culture improves MSC adhesion on freeze-dried bone as a scaffold for bone engineering

AIM: To investigate the interaction between mesenchymal stem cells (MSCs) and bone grafts using two different cultivation methods: static and dynamic. METHODS: MSCs were isolated from rat bone marrow. MSC culture was analyzed according to the morphology, cell differentiation potential, and surface molecular markers. Before cell culture, freeze-dried bone (FDB) was maintained in culture for 3 d in order to verify culture medium pH. MSCs were co-cultured with FDB using two different cultivation methods: static co-culture (two-dimensional) and dynamic co-culture (three-dimensional). After 24 h of cultivation by dynamic or static methods, histological analysis of Cell adhesion on FDB was performed. Cell viability was assessed by the Trypan Blue exclusion method on days 0, 3 and 6 after dynamic or static culture. Adherent cells were detached from FDB surface, stained with Trypan Blue, and quantified to determine whether the cells remained on the graft surface in prolonged non-dynamic culture. Statistical analyses were performed with SpSS and a p potential for adipogenic and osteogenic differentiation of MSC cultures. Rat MSCs were positive for CD44, CD90 and CD29 and negative for CD34, CD45 and CD11bc. FDBs were maintained in culture for 3 d and the results showed there was no significant variation in the culture medium pH with FDB compared to pure medium pH (p > 0.05). In histological analysis, there was a significant difference in the amount of adhered cells on FDB between the two cultivation methods (p day 0, the cell viability in the dynamic system was significantly higher than in the static system (p days 0, 3 and 6 after dynamic culture (p day 6

Resveratrol attenuates metabolic,sperm,and testicular changes in adult Wistar rats fed a diet rich in lipids and simple carbohydrates

Development of Sustainable Solutions for Zebra Mussel Control Through Chemical product Engineering

The zebra mussel is an important aquatic pest that causes great damage to freshwater-dependent industries, due to biofouling. The main goal of the project discussed here is to develop improved solutions to control this species. Three approaches have been explored in an attempt to design innovative application strategies for existing biocides：（i） encapsulation of toxins; （ii） combination of toxins; （iii） investigation of the seasonal variation of the species＇ tolerance to toxins. In this paper, the principles behind these approaches and the major results on each topic are presented. The benefits of adopting a chemical product engineering approach in conducting this project are also discussed.

The study of thermal comfort in the built environment is of great relevance since it stimulates the development of more sustainable buildings suited to the local climate and able to meet the human need for *** objective of this research was to develop,construct and test a passive heating system adaptable to existing buildings,reducing the need for major interventions and increasing thermal comfort in the indoor *** adopted methodological approach was a case study in a single-family residence located in the Brazilian city of petrópolis,a mountainous region with a humid subtropical climate and a rigorous *** proposed passive heating system is totally isolated,thus mitigating air infiltration and promoting increase of temperature in the internal environment through the absorption of solar energy and greenhouse *** kind of solution is especially interesting for residents of this region,since most of the city buildings are not adequately prepared to handle low ***,given local climatic conditions,residents need to spend a lot of money on the acquisition and operation of electric or gas heating *** results indicated that the developed system,in fact,increased the temperature of the studied room when compared to an adjacent room,which did not receive the *** findings of this paper,therefore,provide a valuable reference for experts and practitioners in the selection of heating systems to be used in cold regions,and proved that passive systems can provide thermal comfort at the same time that optimize the interaction of the building with the local ecosystem.

Composition and antimicrobial activity of Cistus munbyi essential oil: an endemic plant from Algeria

The chemical composition and antimicrobial potential of Cistus munbyi essential oil were studied for the first time. GC and GC–MS analysis revealed 48 compounds representing 96.98% of the oil. Terpinen-4-ol(23.75%), meta-Cymene(17.30%), and Sabinene(12.38%)were the major constituents. Antimicrobial activity was evaluated against nine pathogens using the disc diffusion and broth micro-dilution methods. Results show that *** essential oil possesses strong antimicrobial activity against all strains, regardless if Gram-positive or Gramnegative bacteria, or yeast, with MICs values not exceeding10(mg/ml). In addition to its efficacy, C. munbyi essential oil has an unusual antimicrobial potency which is attributed to its specific chemical composition. Thus, findings presented here suggest that endemic C. munbyi contains a very interesting essential oil that may be valuable in several areas.

Two-Dimensional Multi-Domain Hybrid Spectral-WENO Methods for Conservation Laws

Association between the CYBA and NOX4 genes of NADpH oxidase and its relationship with metabolic syndrome in non-alcoholic fatty liver disease in Brazilian population

Background: Oxidative stress has been implicated in the progression of severe forms of non-alcoholic fatty liver disease(NAFLD). NADpH oxidase produces reactive oxygen species. In the present study, we investigated for the first time two single nucleotide polymorphisms(SNps) in the regulatory region of genes encoding NADpH oxidase 4(NOX4) and p22 phox(CYBA) in ***: A total of 207 biopsy-proven NAFLD patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(NASH)(n = 180)] were evaluated. Genomic DNA was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of ***: Associations of CYBA-675 T/A with high-density lipoprotein(HDL)(TT vs TA vs AA; p p patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(ALT)(CA + AA vs CC; p = 0.02). However, there was no association of SNps in the CYBA and NOX4 genes encoding the NADpH oxidase system proteins and the presence of NASH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, p p presence of ***: There were associations between the presence of the allele A in the NOX4 SNp and a higher concentration of ALT in the NAFLD population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGL and lower HDL in NASH patients. The presence of metabolic syndrome was associated with advanced degrees of fibrosis in NAFLD patients.

Nonalcoholic fatty liver disease(NAFLD) has been identified as one of the most prevalent chronic liver disease in adults and children populations. NAFLD is usually associated with the metabolic syndrome(MS), which is chiefly related to insulin resistance and its consequences. Insulin resistance has a crucial role in the pathogenesis of hepatic steatosis and potentially nonalcoholic steatohepatitis(NASH). Because of the contemporary epidemics of MS and obesity, the burden of NAFLD is also expected to rise. Unhealthy diets, such as the so-called western diet, are enriched in fructose, trans-fatty acids and saturated fat and seem to be associated with the development of NAFLD. In human studies, certain dietary sugars, particularly fructose, are used as a substrate for lipogenesis leading to hepatic fatty infiltration, inflammation, and possibly fibrosis. Other investigations have shown that fat consumption especially cholesterol and trans/saturated fatty acids are also steatogenic and seem to increase visceral adiposity. The identification of specific dietary components that favor the development of NASH could be important for the management of this disorder. This review focuses on the effects of different dietary approaches to prevent and treat NAFLD emphasizing the macronutrients and energy composition.
