purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RpE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from polycaprolactone (pCL), and different concentrations of gelatin (5%, 15% and 30%). ARpE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARpE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARpE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARpE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARpE-19 (RpE65 and ZO-1) qRT-pCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of p 0.05 was considered to be statistically significant. Results ARpE-19 cells grew on pCL/Gelatin membranes mainly in pCL/30% gelatin combination, which had not cytotoxic effect. RpE65/cytokeratin-18/ and actin-positive ARpE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARpE-19 cells fused among, suggesting the presence of tight junctions. The expression of RpE65 and ZO-1 was unchanged. Conclusions: pCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RpE-cells and exhibited RpE-like morphology. Non-participation of asymptomatic candidates in screening protocols reduces early diagnosis and worsens prognosis of colorectal cancer

The Agatsuma et al’s study shows that despite the evidence of the benefits of an early colorectal cancer(CRC)diagnosis,through screening in asymptomatic subjects,up to 50%of candidates reject this option and many of those affected are diagnosed later,in advanced *** efficacy of screening programs has been well-established for several years,which reduces the risk of CRC morbidity and mortality,without taking into account the test used for screening,or other ***,a significant proportion of patients remain unscreened,so understanding the factors involved,as well as the barriers of the population to adherence is the first step to possibly modify the participation *** barriers could include a full range of social and political aspects,especially the type of financial provision of each health *** Japan,health services are universal,and this advantageous situation makes it easier for citizens to access to these services,contributing to the detection of various diseases,including ***,the symptomatic CRC group had a lower early-stage diagnosis rate than the patients detected during follow-up for other comorbidities,and symptomatic and cancer screening groups showed similar early-stage diagnosis.

Calibration of the DEM sliding friction and rolling friction parameters of a cohesionless bulk material

The discrete element method(DEM)has become a valuable tool for understanding the mechanical behaviour of granular assemblies,however,the accuracy of the DEM simulations depends on several interaction parameters such as the sliding friction and rolling friction ***,these parameters are estimated using bulk calibration approach(BCA)where the draw down test has been suggested as an effective way to approach these *** test provides up to four bulk criteria,the angle of repose,shear angle,mass flow rate and the mass loss which are used to narrow down the possible ***,there is still more research needed around it to fully understand how this methodology *** experimental and numerical study was carried out using the draw down test to assess the influence of different mass flow rates and particle shapes on DEM parameters for a cohesionless bulk *** is concluded that use of multi-sphere particles and three aperture sizes in the draw down test to calibrate the sliding and rolling friction parameters of a cohesionless bulk material can converge to a small feasible region in which a single combination of the friction coefficients can be *** calibrated sliding and rolling friction coefficients were validated using multi-sphere particles,where the maximum deviation was 5.9%from the experimental values.

Does enhanced recovery after surgery programs improve clinical outcomes in liver cancer surgery?
