Expression of Notch pathway components (Numb, Itch, and Siah-1) in colorectal tumors: A clinicopathological study

BACKGROUND The role of the Notch pathway in carcinogenesis and tumor progression has been demonstrated in many organs,including the ***,studies aimed at developing therapies targeting this pathway in various cancers require the identification of several factors that may play a role in regulating Notch-1 *** Numb,Itch,and seven in absentia homolog-1(Siah-1)have been shown to contribute to the regulation of Notch signaling,their role in colorectal carcinogenesis and tumor progression has not been fully elucidated to *** To evaluate Numb,Itch,and Siah-1 expression in colorectal tumors to clarify their relationship with Notch-1 expression and their role in carcinogenesis and tumor *** Expression of Notch-1,Numb,Itch,and Siah-1 was investigated in 50 colorectal carcinomas,30 adenomas,and 20 healthy colonic tissues by immunohistochemistry and quantitative real-time polymerase chain reaction(PCR)*** In contrast to Notch-1,which is expressed at higher levels in tumor tissues and adenomas,expression of Numb,Itch,and Siah-1 was stronger and more frequent in normal mucosa(P<0.01).There was a positive correlation between Notch-1 expression and high histological grade,the presence of lymph node metastasis,and advanced-stage tumors,whereas expression of Numb,Itch,and Siah-1 was absent or reduced in tumors with these clinicopathological parameters(P<0.05).In survival analysis,expression of Notch was related to poor prognosis but that of Numb,Itch,and Siah-1 correlated with improved survival(P<0.05).Multivariate analysis revealed Notch-1 expression and loss of Numb expression to be independent prognostic parameters together with lymph node metastasis(P<0.05).CONCLUSION Our findings support the role of Notch-1 in colorectal carcinoma and indicate that loss of Numb,Itch,and Siah-1 expression is associated with *** data also suggest that these three proteins might be involved in the Notch-1 pathway during colo

This study was designed to investigate whether Petroselinum crispum(PC)extract has protective effects on the brain in the scopolamine-induced Alzheimer’s disease(AD)rat *** rats were divided into;control,scopolamine(1 mg/kg,i.p.),galantamine(1.5 mg/kg,i.p.)and PC extract(2 g/kg,p.o.)-treated scopolamine *** day 14,the novel object recognition test(NORT)and Morris water maze test(MWMT)were performed and then the rats were ***-induced cognitive impairments observed in the NORT and MWMT,significantly improved with PC extract and galantamine *** reduced M_(1) receptor expression,Bcl-2/Bax ratio,and glutathione levels in the hippocampus and frontal cortex,while malondialdehyde levels,caspase-3/9 expressions,and acetylcholinesterase(AChE)activity were found to be *** the other hand,PC and galantamine treatments reversed these *** conclusion,PC extract has shown an ameliorative effect on the spatial and recognition memory,M_(1) receptor expression,apoptosis,oxidative stress,and increased AChE ***,it was concluded that PC could prevent AD-like conditions and can be used as a functional ***,since animal models do not completely mimic those of humans,based on the data obtained in this study,the importance of PC on human AD should be demonstrated in future studies.

Objective: To prepare and characterize poly lactic-co-glycolic acid(PLGA) nanoparticles loaded with soluble leishmanial antigen or autoclaved leishmanial antigen and explore in vitro and in vivo immunogenicity of antigen encapsulated nanoparticles. Methods: Water/oil/water double emulsion technique was employed to synthesize PLGA nanoparticles, and scanning electron microscopy, Fourier transform infrared spectroscopy and Zeta-potential measurements were used to identify the characteristics of nanoparticles. Cytotoxicity of synthetized nanoparticles on J774 macrophage were investigated by MTT assays. To determine the in vitro immunostimulatory efficacies of nanoparticles, griess reaction and ELISA was used to measure the amounts of NO and cytokines. During the in vivo analysis, Balb/c mice were immunized with vaccine formulations, and protective properties of nanoparticles were measured by Leishman Donovan unit in the liver following the infection. Cytokine levels in spleens of mice were determined by ELISA. Results: MTT assay showed that neither soluble leishmanial antigen nor autoclaved leishmanial antigen encapsulated nanoparticles showed cytotoxicity against J774 macrophage cells. Contrary to free antigens, both autoclaved leishmanial antigen-nanoparticle and soluble leishmanial antigen-nanoparticle formulations led to a 10 and 16-fold increase in NO amounts by macrophages, respectively. Leishman Donovan unit calculations revealed that soluble leishmanial antigen-nanoparticles and autoclaved leishmanial antigen-nanoparticles yielded 52% and 64% protection against visceral leishmaniasis in mouse models. Besides, in vitro and in vivo tests demonstrated that by increasing IFN-γ and IL-12 levels and inhibiting IL-4 and IL-10 secretions, autoclaved leishmanial antigen-nanoparticles and soluble leishmanial antigennanoparticles triggered Th1 immune response. We compare the efficacy of intratesticular ozone therapy with intraperitoneal ozone therapy in an experimental rat model. For this purpose, 24 rats were divided into four groups including sham-operated, torsion/detorsion, torsion/detorsion plus intraperitoneal ozone （O-IP）, and torsion/detorsion plus intratesticular ozone （O-IT）. The O-IP ozone group received a 4 mg kg-1 intraperitoneal injection of ozone, and the O-IT group received the same injection epididymally. At 4 h after detorsion, the rats were sacrificed and orchiectomy materials were assessed histopathologically. Spermatogenesis in the seminiferous tubules and damage to the Sertoli cells were histopathologically evaluated in the testes using the Johnsen scoring system, i-NOS and e-NOS activities in the testis tissue were also evaluated. Torsion-detorsion caused a decreased Johnsen score and increased apoptosis of spermatogonial and Sertoli cells. Ozone injection prevented increases in Johnsen score and i-NOS level, e-NOS level of the O-IP group was significantly lower than that of the O-IP group, and i-NOS level of the O-IT group was significantly lower than that of the O-IP group. Local ozone therapy is more effective than systemic ozone therapy at improving IRI-related testicular torsion. Our study is the first to show that the efficacy of intratesticular implementation of ozone therapy is higher than that of intraperitoneal ozone therapy.

Dear Editor, We would like to present a rare case ofpolyorchidism. Polyorchidism is an unusual abnormality of the genital tract and until date, there are approximately 178 case reports of polyorchidism in the literature？ However, the most of patients are asymptomatic and usually diagnosed incidentally. Some may present as inguinal hernia （30%）, maldescended testis （15% to 30%）, testicular torsion （13%） and hydrocele （9%）, varicocele （1%）, hypospadias （〈1%）, anomalous urogenital union （〈1%）, and malignancy （〈1%）.2 Management of polyorchidism is still controversial. Important factors of polyorchidism management are testis location, size and anatomical organization of the testicular drainage system and the age of the patient.

Neuropilins(NRPs) are highly conserved transmembrane glycoproteins that possess pleiotropic functions. Neuropilin-1(NRP1) and its homologue neuropilin-2 interact as coreceptors with both class 3 semaphorins and vascular endothelial growth factor and are involved in neuronal guidance and angiogenesis, respectively. The contribution of NRPs to tumor angiogenesis has been highlighted in previous studies, leading to the development of NRP antagonists as novel anti-angiogenesis therapies. However, more recent studies have demonstrated that NRPs have a much broader spectrum of activity in the integration of different pathways in physiological and pathological conditions. A few studies investigated the role of NRPs in both malignant and non-neoplastic liver diseases. In normal liver, NRP1 is expressed in hepatic stellate cells and liver sinusoidal endothelial cells. NRP1 expression in hepatocytes has been associated with malignant transformation and may play an important role in tumor behavior. A contribution of NRPs in sinusoidal remodeling during liver regeneration has been also noted. Studies in chronic liver diseases have indicated that, besides its influence on angiogenesis, NRP1 might contribute to the progression of liver fibrosis owing to its effects on other growth factors, including transforming growth factor β1. As a result, NRP1 has been identified as a promising therapeutic target for future antifibrotic therapies based on the simultaneous blockade of multiple growth factor signaling pathways. In this review, the structure of NRPs and their interactions with various ligands and associated cell surface receptors are described briefly. The current understanding of the roles of the NRPs in liver diseases including tumors, regeneration and fibrogenesis, are also summarized.

With the development of aircraft equipment, conventional navigation is the shift from performance based navigation (PBN). As is known, conventional navigation is based on ground-based navigation aids;however, PBN is based on aircraft avionics and performance. In this paper, a new method called Point Merge System (PMS) considered as one of PBN procedures will be introduced. PMS has many benefits related to fuel savings and emission reductions by implementing Continuous Descent Approaches (CDAs). A new PMS standard arrival route (STAR) model will be designed in radar simulation and it will be suggested.

Aim: The aim of this study was to determine the demographical and clinical features of patients with cerebral palsy (CP) attending to our outpatient or inpatient clinics. Materials and Methods: Children admitted to Inonu University Faculty of Medicine Department of Physical Medicine and Rehabilitation outpatient or inpatient clinics, Malatya, Turkey with the diagnosis of CP during the study period were included in the study. Age, sex, etiological factors, clinical classifications, and epidemiological characteristics such as socio-economic status as well as the problems associated with CP were analyzed in all patients. Results: Of the total of 130 patients with mean ages 51.05 ± 36.06 months, 51 were girls and 79 were boys. The most common etiologic risk factors were bleeding and presence of threatened miscarriage in prenatal period;asphyxia, low birth weight and prematurity in perinatal period;and neonatal convulsions and hyperbilirubinemia in the postnatal period. Consanguineous marriage was present in 24.6% of families. In clinical classification it was seen that 45 subjects were spastic quadriplegic (34.6%), 41 were spastic diplegic (31.5%), 15 were dyskynetic (11.5%), 14 were spastic hemiplegic (10.8%), 10 were mixed type (7.7%) and 5 were hypotonic/ataxic (3.8%). Conclusion: Prenatal risk factors constitute the foremost risk factors in CP etiology. CP incidence can be lowered by close follow up of the newborns who has been considered as high risk newborns due to the presence of risk factors during pregnancies or deliveries, and by increasing the number of neonatal intensive care units. Besides, it is important to raise public awareness about consanguinity marriages in our country.
