Lipase.producing Aspe.gillus was isolate. from soil colle.te. from a re.use.dump site.locate. at Awka, Anambra State. Nige.ia using standard microbiology and bioche.ical te.hnique.. Crude.e.tract of lipase.was produce. afte. a succe.sful scre.ning of the.isolate. using mine.al broth containing p-NPP through subme.ge. fe.me.tation syste. with optimize. physiologic conditions. Thre. ste.s of purification we.e.carrie. out: Ammonium sulphate. dialysis and ge. filtration (se.hade. G-150). Crude.e.tract was pre.ipitate. by using 70% saturation of ammonium sulphate.at pH 6.0 which gave.the.optimum pre.ipitation of the.prote.n with spe.ific activity of 260.56 U/mg. Pre.ipitation using ammonium sulphate.carrie. out at pH 6.5 and 8.0 gave.spe.ific activity of 217 U/mg of the.prote.n. The.pre.ipitate. we.e.furthe. de.alte. through dialysis for twe.ve.hours and spe.ific activity of 343.20 U/mg was re.orde. from the.dialysate.afte.wards. Furthe. purification was done.by using se.hade. G-150 and spe.ific activity of 490.55 U/mg was re.orde. from the.active.poole. fractions. The.purification table.showe. a 2.32 purification folds of lipase.was gotte. afte. ge. filtration (se.hade. G-150) with a lipase.pe.ce.tage.yie.d of 2.00%. The.spe.ific activity of lipase.incre.se. from 211.81 to 490.55 U/mg. Characte.ization of β-galactosidase.gave.optima pH and te.pe.ature.of the.e.zyme.at 6.0 and 60°C re.pe.tive.y. Kine.ic constants: Km and Vmax value. we.e.obtaine. at various conce.trations of p-NPP whe.e.0.32 mM and Vmax of 200.00 μmol/min re.pe.tive.y. Ca2+ and Co2+ showe. gre.te. e.fe.t on lipase.activity in a conce.tration-de.e.de.t manne. (0.03 - 0.05 M) whe. compare. to Mn2+ and Fe.sup>2+. The.re.ults from this study have.shown that lipase.produce. from filame.tous Aspe.gillus has a wide.range.of activity ove. physiologic conditions in re.ards to industrial and clinical standard ope.ational proce.ure..

Activity data and e.ission factors are.critical for e.timating gre.nhouse.gas e.issions and de.ising e.fe.tive.climate.change.mitigation strate.ie.. This study de.e.ope. the.activity data and e.ission factor in the.Fore.try and Othe. Land Use.Change.(FOLU) subse.tor in Malawi. The.re.ults indicate.that “fore.tland to cropland,” and “we.land to cropland,” we.e.the.major land use.change. from the.ye.r 2000 to the.ye.r 2022. The.fore.tland ste.dily de.line. at a rate.of 13,591 ha (0.5%) pe. annum. Similarly, grassland de.line. at the.rate.of 1651 ha (0.5%) pe. annum. On the.othe. hand, cropland, we.land, and se.tle.e.ts ste.dily incre.se. at the.rate.of 8228 ha (0.14%);5257 ha (0.17%);and 1941 ha (8.1%) pe. annum, re.pe.tive.y. Furthe.more. the.re.ults indicate.that the.“grassland to fore.tland” change. we.e.highe. than the.“fore.tland to grassland” change., sugge.ting that fore.t re.rowth was occurring. On the.e.ission factor, the.re.ults inte.e.tingly indicate.that the.e.was a significant incre.se.in carbon se.ue.tration in the.FOLU subse.tor from the.ye.r 2011 to 2022. Carbon se.ue.tration incre.se. annually by 13.66 ± 0.17 tCO2 e.ha/yr (4.6%), with an unce.tainty of 2.44%. The.e.ore. it can be.conclude. that the.e.is pote.tial for a Carbon marke. in Malawi.

Me.hanical e.aluation of AZ80 Magne.ium Alloy in Cast Wrought Form

Wrought magne.ium alloy AZ80 with a thick se.tion of 20 mm was pre.are. by sque.ze.casting (SC) and pe.mane.t ste.l mold casting (PSMC). The.porosity me.sure.e.ts of the.SC and PSMC de.icte. that SC AZ80 had a pore.conte.t of 0.52%, which was 77% lowe. than 2.21% of PSMC AZ80 counte.part. The.YS, UTS, e.sub>f, e.and stre.gthe.ing rate.of cast AZ80 we.e.de.e.mine. by me.hanical pulling. The.e.gine.ring stre.s ve.sus strain be.de. line. showe. that SC AZ80 had a YS of 84.7 MPa, a UTS of 168.2 MPa, 5.1% in e.sub>f, and 25.1 GPa in modulus. But, the.YS, UTS and e.sub>f of the.PSMC AZ80 spe.ime. we.e.only 71.6 MPa, 109.0 MPa, 1.9% and 21.9 GPa. The.findings of the.me.hanical pulling e.ide.tly de.icte. that the.YS, UTS, e.sub>f and e.of SC AZ80 we.e.18%, 54%, 174% and 15% highe. than PSMC counte.part. The.compute. re.ilie.ce.and toughne.s sugge.te. that the.SC AZ80 e.hibite. gre.te. re.istance.to te.sile.loads during e.astic de.ormation and posse.se. highe. capacity to absorb e.e.gy during plastic de.ormation compare. to the.PSMC AZ80. At the.be.inning of pe.mane.t change. the.stre.gthe.ing rate.of SC AZ80 was 10,341 MPa, which was 9% gre.te. than 9489 MPa of PSMC AZ80.
