Deep learning identification of novel autophagic protein-protein interactions and experimental validation of Beclin 2-Ubiquilin 1 axis in triple-negative breast cancer

Background:Triple-negative breast cancer (TNBC), characterized by its lack of traditional hormone receptors and HER2, presents a significant challenge in oncology due to its poor response to conventional therapies. Autophagy is an important process for maintaining cellular homeostasis, and there are currently autophagy biomarkers that play an effective role in the clinical treatment of tumors. In contrast to targeting protein activity, intervention with protein-protein interaction (PPI) can avoid unrelated crosstalk and regulate the autophagy process with minimal interference pathways.Methods:Here, we employed Naive Bayes, Decision Tree, and k-Nearest Neighbors to elucidate the complex PPI network associated with autophagy in TNBC, aiming to uncover novel therapeutic targets. Meanwhile, the candidate proteins interacting with Beclin 2 were initially screened in MDA-MB-231 cells using Beclin 2 as bait protein by immunoprecipitation-mass spectrometry assay, and the interaction relationship was verified by molecular docking and Co-IP experiments after intersection. Colony formation, cellular immunofluorescence, cell scratch and 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) tests were used to predict the clinical therapeutic effects of manipulating candidate PPI.Results:By developing three PPI classification models and analyzing over 13,000 datasets, we identified 3733 previously unknown autophagy-related PPIs. our network analysis revealed the central role of Beclin 2 in autophagy regulation, uncovering its interactions with 39 newly identified proteins. Notably, the Co-IP studies identified the substantial interaction between Beclin 2 and Ubiquilin 1, which was anticipated by our model and discovered in immunoprecipitation-mass spectrometry assay results. Subsequently, investigations showed that overexpressing Beclin 2 increased Ubiquilin 1, promoted autophagy-dependent cell death, and inhibited proliferation and metastasis in MDA-MB-231 cells.

Removal of Cr (VI) from Tannery Effluent and Aqueous Solution by Sequential Treatment with Microfungi and Basidiomycete-Degraded Sawdust

Removal of Cr (VI) from aqueous solution and tannery effluent in sequence with Cr (VI) resistant microfungi (Aspergillus niger, Penicillium chrysogenum) and sawdust degraded by basidiomycete (Gloeophyllum sepiarium) was investigated in the laboratory. Initial or primary treatment with microfungi reduced 200 mg/l Cr (VI) in aqueous solution by 64.6% - 78.2% while a markedly lower 0.52 mg/l Cr (VI) in tannery effluent was reduced by 72.4% - 84.6%. However, the residual Cr (VI) in both aqueous solution and tannery effluent was reduced to a non-detectable level after secondary treatment by passage through basidiomycete-degraded sawdust column. The recovery of 65.4% - 87.7% of the Cr (VI) removed by treatment microfungi by elution indicated adsorption as the major mechanism for Cr (VI) removal. The microfungi reduced BoD in tannery effluent by 85.3 ± 5.6 - 92.7 ± 6.8 and concomitantly removed Cr (VI), hence it is hypothesized that non-Cr (VI) constituents of tannery effluent may have interfered with biosorption of Cr (VI) by treatment microfungi. It is concluded that the two-stage sequential treatment process may be of potential cost-saving stratagem for removal of chromium from industrial wastes.

Effect of Methanol Extract of <i>Gomphrena celesioides</i>on Chloroquine-Induced Hepatotoxicity and oxidative Stress in Male Wistar Rats

Hepatic injury has been reported to be associated with chloroquine therapy. Gomphrena celesioides has been claimed to have pleiotropic protective properties in the liver by traditional herbal practitioner but there is no scientific evidence to this claim. This investigation therefore sought to evaluate the effect of Gomphrena celesioides administration on chloroquine-induced hepatic injury in rats. Forty adult male rats were divided into five groups of eight rats and were treated orally once daily. Rats in group one received 1 ml/kg body weight of 0.9% normal saline;rats in group two received 250 mg/kg body weight of chloroquine for three days;groups three, four and five rats were pre-treated with 200 mg/kg, 400 mg/kg and 800 mg/kg body weight of methanol extract of Gomphrena celesiodes for three days and on the fourth day were given 250 mg/kg body weight of chloroquine for three days. The experiment lasted for seven days. Liver injury was manifested biochemically by a significant increase in serum level or activities of hepatic markers (aminotransferases, alkaline phosphate, bilirubin, cholesterol and gamma glutamyl transferase). In addition, hepatic tissue from chloroquine-treated rats showed a significant increase in lipid peroxidation with a decrease in hepatic superoxide dismutase, catalase, glutathione peroxidase, glutathione-S-transferase and glutathione reservoirs. Moreover, the liver histopathologic evaluation revealed significant in chloroquine-treated rats. Gomphrena celesioides administration significantly alleviated chloroquine-induced pathologic changes in serum biochemistry and liver tissue. The results also suggest that Gomphrena celesioides possesses protective properties against chloroquine-induced liver injury via mitigation of drug-induced oxidative stress and its consequent events.

Solution of Some Second order ordinary Differential Equations Using a Derived Algorithm

We emphasized explicitly on the derivation and implementation of a new numerical algorithm scheme which gave stable results that show the applicability of the method. In this paper, we aimed to solve some second order initial value problems of ordinary differential equations and compare the results with the theoretical solution. Using this method to solve some initial value problems of second order ordinary differential equations, we discovered that the results compared favorably with the theoretical solution which led to the conclusion that the new numerical algorithm scheme derived in the research is approximately correct and can be prescribed for any related ordinary differential equations.

Tannin-Rich Extract of <i>Chasmanthera dependens</i>Stem Potential in Piroxicam-Induced Nephrotoxicity in Adult Male Wistar Rats

Piroxicam is commonly used as anti-inflammatory and pain relieving drug;however, its side effects include fluid retention, renal damage and heart failure. This study aimed at evaluating the nephroprotective role of different doses of the tannin-rich extract of Chasmanthera dependens stem (TRECDS) on piroxicam-induced nephrotoxicity in adult male Wistar rats. Thirty-two adult rats were divided into four groups of eight rats per group and treated orally for ten days. Rats in group one received 0.5 ml normal saline (0.9% v/v) and served as normal control group. Rats in group two received 20 mg/kg body weight piroxicam alone. Rats in groups three and four received 20 mg/kg body weight of piroxicam with concomitant administration of 200 and 400 mg/kg body weight of TRECDS. At the expiration of the experiment, rats were sacrificed and the kidney was removed. Renal function was evaluated. The results showed that administration of piroxicam alone caused a significant elevation in the serum concentrations of albumin, creatinine, total protein, urea concentrations and the activity of renal nucleotidase with a reduction in the activity of glucose-6-phosphate dehydrogenase (G6PD) when compared to normal control (p ore, renal tissue from the piroxicam alone treated group revealed a significant decrease in the activities of renal superoxide dismutase, catalase, glutathione peroxidase and glutathione-S-transferase as well as reduced glutathione with concomitant increase in lipid peroxidation and hydrogen peroxide generation. In addition, histological assessment of the renal tissue showed noticeable damage in piroxicam alone treated group. However, concomitant administration of TRECDS showed a dose-dependent reduction in the concentrations and the activity of the kidney markers with significant increase in the activities of G6PD and restores the antioxidant status of the kidney. Effect of Ceo_2 and Al_2o_3 on the activity of Pd/Co_3o_4/cordierite catalyst in the three-way catalysis reactions(Co/No/C_nH_m)

The present article studies the effect of Ceo2 and A1203 on the activity of Pd/Co304/cordierite catalyst in conversion of No, Co, CnHm. The catalysts were characterized by temperature programmed reduction with hydrogen, X-ray diffraction, X-ray photoelectron spectroscopy and transmission electron microscopy. It is shown that the effect of Ceo2 on the properties of Pd/C03 o4/cordierite catalyst depends on preparation method. The catalyst obtained by co-deposition of cerium and cobalt oxides has higher activity in Co oxidation （Co ＋ 02 and Co ＋ No） and total hexane oxidation （C6H14 ＋ 02）. Such phenomenon is probably caused by more than stoichiometric amount of formed oxygen vacancies, an increase in both mobility of surface oxygen and dispersity of components in the catalytic composition. It is demonstrated that Ceo2 addition promotes the So2 resistance of Pd/C03 o4/cordierite. The second support decreases the activity of Pd/Co3oa/cordierite catalyst in the reactions of Co and C6H14 with oxygen because of CoA1204 formation.

Diclofenac-Induced Kidney Damage in Wistar Rats: Involvement of Antioxidant Mechanism

Kidney damage has been associated with administration diclofenac, a phenylacetic acid derivative belonging to the nonsteroidal anti-inflammatory drugs (NSAIDs), which is commonly used for the treatment of various diseases such as rheumatoid arthritis, ankylosing spondylitis, acute muscle pain conditions and osteoarthritis. This study investigated the exact mechanism of diclofenac in renal toxicity by determining the involvement of oxidative stress in rats. Adult male Wistar rats were divided into two groups of eight rats in each group and orogastrically treated for three days. Group 1 served as the normal control and received normal saline (0.9% w/v) and group 2 received 40 mg/kg body weight of diclofenac for three days. Administration of diclofenac caused degeneration of the kidney of rats as evidenced by significant elevation in the serum levels of creatinine, urea, albumin, uric acid, protein and electrolytes and the activities of renal-5’-nucleotidase and glucose-6-phosphate-dehydrogenase (G6PDH) compared with control. Furthermore, administration of diclofenac decreased the activities of superoxide dismutase (SoD), catalase, glutathione peroxidase (GPx), and glutathione-S-transferase (GST) and the level of glutathione with concomitant increase in hydrogen peroxide (H2o2) and malondialdehyde (MDA) levels in the kidney of the diclofenac treated groups compared with control. These findings reveal that administration of diclofenac may impair kidney functions through induction of oxidative stress.

B3Y-FETAL effective interaction in the folding analysis of elastic scattering of ^(16)o+^(16)o

In this paper,a new M3Y-type effective nucleon–nucleon interaction,derived based on the lowest order constrained variational approach(LoCV)and termed B3Y-Fetal,has been used in DDM3Y1,BDM3Y1,BDM3Y2,and BDM3Y3 density-dependent versions in a heavy ion(HI)optical potential based on four types of a real folded potential and a phenomenological Woods–Saxon imaginary potential to study the elastic scattering of the^(16)o+^(16)o nuclear system within the framework of the optical model(oM)by computing the associated differential cross sections at various incident *** results of the folding analyses have shown the DDB3Y1-Fetal and BDB3Y1-Fetal,out of the four folded potentials,give a reasonably better description of the elastic data of the nuclear *** best-fit folded potentials are followed,in performance,by the BDB3Y2-Fetal,with the BDB3Y3-Fetal potential coming *** performance trend was also demonstrated by the optical potentials based on the M3Y-Reid ***,the best-fit folded potentials,renormalized by a factor NRof approximately 0.9,have been shown to reproduce the energy dependence of the real optical potential for^(16)o scattering found in previous optical model analyses creditably *** excellent agreement with previous works,they have also been identified in this work to belong to the family of deep refractive potentials because they have been able to reproduce and consistently describe the evolution of Airylike structures,at large scattering angles,observed in the^(16)o scattering data at different ***,a comparison of the performances of B3Y-Fetal and M3Y-Reid effective interactions undertaken in this work has shown impressive agreement between them.

Mn2+and the trivalent europium(Eu3+)-doped MgGa2o4 ceramics are characterized using a multi-experimental *** formation of spinel-structured ceramics is ascertained from X-ray diffraction(XRD)*** investigations with transmission electron microscopy(TEM)show irregularly shaped grains and grain boundaries with a homogeneous distribution of Eu3+*** inability of Eu activator to penetrate the bulk of ceramic grains is inferred from positron annihilation lifetime spectroscopy *** Eu doping is shown to enhance the positron trapping rate due to the occupancy of vacancy-type defects at ceramic grains by Eu3+*** Mn2+and Eu3+doped samples show a broad multi-color luminescence in 350–650 nm range under 240 nm and 270–300 nm *** emission is concluded to originate from host defects,whereas green emission and narrow lines in the red region of the spectrum are attributed to Mn2+and Eu3+ions,*** asymmetry around Eu3+ions can be concluded from the photoluminescence and positron annihilation lifetime spectra analysis.
