a.M: To sea.ch tools of high blood pressure in the model of nitric oxide (NO)-defective hypertension, a.d the study focused on the effect of rilmenidine, a.onist of imida.oline receptors, which wa. suggested to modula.e centra. sympa.hetic outflow. METHODS: Three experimenta. groups, ea.h consisting of 7 ra.s, were used: (I) ra.s with inhibition of NO syntha.e (NOS) by NG-nitro-L-a.ginine methyl ester (L-Na.E) 40 mg·kg-1·d-1 for 4 weeks in drinking wa.er, (II) ra.s with inhibited NOS a. in group I , plus a.onist of imida.oline receptors rilmenidine 3 mg·kg-1·d-1 for 4weeks by ga.a.e, a.d(III) control ra.s. Systolic blood pressure wa.mea.ured weekly noninva.ively. a. the end of experiment a.rtic ring isometric tension wa. followed, NOS expression (a.rta. left ventricle), a.d NOS a.tivity (left ventricle a.d bra.n) were determined. RESULTS: In the group I systolic blood pressure in- crea.ed significa.tly, a.rtic ring rela.a.ion to a.etylcholine wa. significa.tly a.tenua.ed. Rilmenidine a.ministered simulta.eously with L-Na.E (group II) prevented the increa.e of blood pressure which did not differ significa.tly from control va.ues; a.rtic ring rela.a.ion to a.etylcholine did not differ from control. No cha.ge in NOS expres- sion (a.rta.a.d left ventricle) wa. found in groups I a.d II. Significa.t decline in NOS a.tivity (left ventricle a.d bra.n) wa.found in groups I a.d II. CONCLUSION: Rilmenidineha. a.ema.ka.le role inNO-defective hypertension, possibly by inhibiting centra. sympa.hetic outflow a.d by a.fecting receptors in va.cula. smooth muscle a.so. The prime ca.se of hypertension in this experimenta. model - the compromised production of NO due to inhibition of NOS - wa. not a.fected by rilmenidine.
