The Growth Study of Vero Cells in Different Type of Microcarrier

The fact of microcarrier (MC) culture introduces new possibilities and makes possible the practical high-yield culture of anchorage-dependent cells has generated a considerable focus in this study. The objective of this research was to study the comparison of Vero cell growth on different types of commercial microcarriers;Cytodex-1, Cytodex-3, Hil-lex? II and Plastic Plus in spinner vessel and two liters bioreactor cultured for 96 hours. Biological performance of the microcarrier in RPMI media showed the preference of Vero cell grew on Cytodex 3 microcarriers with highest maximum viable cell number (2.4 × 105 cells/ml) followed by Cytodex 1, Hillex and Plustic Plus. Vero cell on Cyto-dex-3 data in spinner flask was compared in bioreactor and result showed higher viable cell number in biorector. Thus, this dextran-crosslink gelatin microcarrier (Cytodex 3) provided the best surface for cell attachment and fast proliferation. At the end of this cell growth improvement will be used for virus transfection producing a vaccine in bioreactor.

Objective: To determine the etiologies of tropical acute febrile illness(TAFI) in West Pahang, Malaysia and to investigate morbidity and mortality factors in relation to TAFI. Methods: A multicenter prospective cohort study was conducted between January and June 2016 in six district hospitals throughout the western part of Pahang State in Peninsular Malaysia. A total of 336 patients answered a standardized questionnaire and blood samples were collected for laboratory confirmation of infectious etiology. Descriptive analysis and logistic regression were performed to identify factors associated with TAFI. Results: A total of 336 patients were included. The patients were mainly Malays(70.2%), males(61.3%), aged(44.6±17.4) years, with more than half(58.9%) presenting with gastrointestinal symptoms. The majority were diagnosed with dengue(35.7%) while malaria(4.5%) was the least frequent. The in-hospital mortality due to TAFI was 9.2%. Patients with meliodosis had five times higher mortality [Adjusted OR: 5.002, 95% CI:(1.233, 20.286)]. Patients with comorbidities such as cardiovascular symptoms(P<0.001) and renal replacement therapy initiation(P<0.001) were significantly associated with in-hospital mortality in all TAFI. Conclusions: The etiology of TAFI in the western Pahang includes dengue, leptospirosis, malaria and melioidosis, which carry the highest risk of in-hospital mortality. The presence of cardiovascular symptoms may be used to assess the disease severity in TAFI, but more studies are needed in the future.

An image retrieval system was developed purposely to provide an efficient tool for a set of images from a collection of images in the large database that matches the user’s requirements in similarity evaluations such as image content similarity, edge, and colour similarity. Retrieving images based on the contents which are colour, texture, and shape is called content-based image retrieval (CBIR). This paper discusses and describes about the colour features technique for image retrieval systems. Several colour features technique and algorithms produced by the previous researcher are used to calculate the similarity between extracted features. This paper also describes about the specific technique about the colour basis features and combined features (hybrid techniques) between colour and shape features.

Flood Risk Pattern Recognition Using Chemometric Technique: A Case Study in Muda River Basin

This study constructs downscaling statistical model in analyzing the hydrological modeling in the study area which faces the risk of flood occurrence as the impact of climate change. The combination of chemometric method and time series analysis in this study show that even during the monsoon season, rainfall and stream flow are not the major contribution towards the changing of water level in the study area. Based on Correlation Test, it shows that suspended solid and water level show high correlation with p-value < 0.05. Factor Analysis being carried out to determine the major contribution to the changes of Water Level and the result show that Suspended Solid shows a strong factor pattern with value 0.829. Based on Control Chat Builder for time series analysis, the Upper Control Limit for water level and suspended solid are 7.529 m and 1947.049 tons/day and the Lower Control Limit are 6.678 m and 178.135 tons/day. This shows that human development in the area gives high impact towards climate change and risk of flood in the study area which commonly faces flood during monsoon season.

Malaria control programme utilizing indoor residual spraying of chemical insecticide is only effective if a high coverage of targeted area is achieved. The effectiveness of the residual spraying, on the other hand, relies on the efficacy and residual activity of the insecticides applied, which to a certain extent are influenced by the nature of the sprayed surfaces. The bioefficacy of indoor residual-sprayed deltamethrin wettable granule (WG) formulation for the control of malaria was compared with the current dose of deltamethrin wettable powder (WP) in malaria endemic areas in Balai Ringin, Sarawak. Doses of 20 mg/m2 WP (control), 20 mg/m2 WG, 30 mg/m2 WG and 40 mg/m2 WG were sprayed separately on different surfaces namely, wooden, rough-bamboo, smooth-bamboo and brick surfaces. Residual activity of WP and WG formulations was tested against lab-bred Anopheles maculatus using WHO standard procedure. Deltamethrin at 30 mg/m2 WG exhibited the highest sustainable level of effectiveness against An. maculatus (An. maculatus mortality was between 95% - 100%) up to week 60 post-spraying when sprayed on smooth- bamboo surface. These results indicated that 30 mg/m2 WG could be an ideal concentration for controlling malaria vector effectively up to 15 months of which long-lasting residual spraying was envisaged. The usual two spraying cycles per year with 20 mg/m2 deltamethrin WP could be replaced with 30 mg/m2 deltamethrin WG since the long residual activity was achieved by employing a single spraying only.

Genotoxic Potential of Shrimp Pastes (Belacan) Extracts Using <i>Umu</i>Test

This study was conducted to observe the genotoxic effects of aqueous, methanol, hexane and dichloromethane extracts of “belacan” (shrimp paste) taken from three local districts in Melaka, Malaysia (Kelemak, Batang Tiga & Pantai Puteri). The umu test which was used as the screening test was conducted with and without the presence of metabolic activation system. Without the presence of metabolic activation system, aqueous extracts from Kelemak showed mutagenicity activity at 5 mg/ml with IR (Induction Rate) = 1.52 ± 0.57 and the methanol extracts showed mutagenic activities at 0.625 mg/ml and 5 mg/ml, which the IR was the highest at 5 mg/ml (2.08 ± 0.09). On the other hand, samples from Batang Tiga, Melaka showed mutagenic effects at all five concentrations for the dichloromethane extract, with IR = 2.09 ± 0.64 as the highest value at 1.25 mg/ml. Methanol extracts also showed positive results at 1.25 mg/ml and 2.5 mg/ml with IR = 1.70 ± 0.33 and IR = 2.12 ± 0.51 respectively, and aqueous extract at 0.625 mg/ml with IR = 1.54 ± 0.48 and 5 mg/ml with IR = 1.74 ± 0.50. There was a significant difference of the mean values of IR between the four different types of “belacan” extracts from Batang Tiga (p < 0.05). All four “belacan” extracts from Pantai Puteri, Melaka did not show any mutagenic effect. With the presence of metabolic activation system, there was no mutagenic effect observed in all four extracts from the three districts. Further study to analyze the contents in the food samples should be done in the future to determine the possible contents in the food samples that might be responsible for the mutagenic activities.
