Expression of triggering receptor on myeloid cell 1 and histocompatibility complex molecules in sepsis and major abdominal surgery

AIM： To evaluate the surface expression of triggering receptor on myeloid cell 1 （TREM-1）, class Ⅱ major histocompatibility complex molecules （HLA-DR）, and the expression of the splicing variant （svTREH-1） of TREM-1 in septic patients and those subjected to major abdominal surgery. METHODS： Using flow cytometry, we examined the surface expression of TREM-1 and HLA-DR in peripheral blood monocytes from 11 septic patients, 7 elective gastrointestinal surgical patients, and 10 healthy volunteers, svTREM-1 levels were analyzed by RT-PCR. RESULTS： Basal expression of TREM-1 and HLA- DR in healthy volunteers was 35.91±14.75 MFI and 75.8±18.3%, respectively. In septic patients, TREM-1 expression was 59.9±23.9 MFI and HLA-DR expression was 44.39±20.25%, with a significant difference between healthy and septic groups （P〈0.05） for both molecules. In the surgical patients, TREM-1 and HLA-DR expressions were 56.8±20.85 HFI and 71±13.8% before surgery and 72.65±29.92 MFI and 72.82±22.55% after surgery. TREM-1 expression was significantly different （P = 0.0087） between the samples before and after surgery and svTREM-1 expression was 0.8590± 0.1451 MF1, 0.8820±0.1460 MF1, and 2.210±0.7873 MF1 in the healthy, surgical （after surgery） and septic groups, respectively. There was a significant difference （P = 0.048） in svTREM-1 expression between the healthy and surgical groups and the septic group. CONCLUSION： TREM-1 expression is increased during systemic inflammatory conditions such as sepsis and the postoperative phase. Simultaneous low expression of HLA-DR molecules correlates with the severity of illness and increases susceptibility to infection. Additionally, TREM-1 expression is distinctly different in surgical patients at different stages of the inflammatory response before and after surgery. Context-dependent role of sirtuin 2 in inflammation

Sirtuin 2 is a member of the sirtuin family nicotinamide adenine dinucleotide(NAD~+)-dependent deacetylases, known for its regulatory role in different processes, including inflammation. In this context, sirtuin 2 has been involved in the modulation of key inflammatory signaling pathways and transcription factors by deacetylating specific targets, such as nuclear factor κB and nucleotide-binding oligomerization domain-leucine-rich-repeat and pyrin domain-containing protein 3(NLRP3). However, whether sirtuin 2-mediated pathways induce a pro-or an anti-inflammatory response remains controversial. Sirtuin 2 has been implicated in promoting inflammation in conditions such as asthma and neurodegenerative diseases, suggesting that its inhibition in these conditions could be a potential therapeutic strategy. Conversely, arthritis and type 2 diabetes mellitus studies suggest that sirtuin 2 is essential at the peripheral level and, thus, its inhibition in these pathologies would not be recommended. Overall, the precise role of sirtuin 2 in inflammation appears to be context-dependent, and further investigation is needed to determine the specific molecular mechanisms and downstream targets through which sirtuin 2 influences inflammatory processes in various tissues and pathological conditions. The present review explores the involvement of sirtuin 2 in the inflammation associated with different pathologies to elucidate whether its pharmacological modulation could serve as an effective strategy for treating this prevalent symptom across various diseases.

The reduction behavior and metallization degree of magnetite concentrate with agave bagasse were investigated in an inert *** effects of temperature,biomass content,and residence time on reduction experiments and metallization degree were investigated by X-ray diffraction and scanning electron *** with other types of biomass,agave bagasse had lower contents of nitrogen,sulfur,and ash.X-ray diffraction analysis showed that the metallization degree improved with increasing temperature and biomass *** metallization was achieved at 1100℃ for 30 min with 65:35 and 50:50 ratios of the magnetite concentrate to the agave *** results demonstrate that agave bagasse promotes the efficient metallization of magnetite concentrate without the external addition of a reducing ***,this biomass is a technical suitable alternative to replace fossil fuels in steelmaking.

Comparative Pharmacokinetic Study between Metformin Alone and Combined with Orlistat in Healthy Mexican Volunteers

Traditional treatment of type 2 diabetes mellitus (T2DM) has focused on correcting hyperglycemia. However, T2DM is almost always accompanied by other conditions and risk factors that affect its morbidity and mortality. Obesity represents one of the main risk factors associated in the T2DM and a modest weight loss around 5% is linked with significant reduction in the blood glucose levels. The objective of this study was to compare the pharmacokinetic parameters between two formulations of metformin, alone or combined with orlistat, in healthy volunteers. A single-blinded, single dose, two-period, two-sequence, crossover, randomized and balanced study design with a 7-day washout period was performed in 26 Mexican volunteers. Plasma samples were collected over a 24-hour period after administration of 500 mg metformin alone or combined with 60 mg orlistat in each period. A validated high-performance liquid chromatography coupled with a ultraviolet detector was used to analyze metformin concentration in plasma. Bioequivalence between metformin alone and metformin combined with orlistat was determined when the ratio for the 90% confidence intervals (CI) of area under the curve (AUC24h) and maximum concentration (Cmax) of the two formulations were within 80% and 125%. In the current study, the mean ± standard deviation (SD) of Cmax, AUC24h and AUC∞ of the formulation containing only metformin were 1.39 ± 0.44 μg/mL, 7.59 ± 3.17 μg h/mL and 8.48 ± 4.13 μg h/mL, respectively, while the mean ± SD of Cmax, AUC24h and AUC∞ of the formulation containing metformin and orlistat were 1.38 ± 0.48 μg/mL, 7.80 ± 2.83 μg h/mL and 9.13 ± 4.29 μg h/mL, respectively. The parametric 90% CI for Cmax and AUC24h were 87.5-109.3 and 88.7-124.7, respectively. These results suggest that both formulations are bioequivalent and there is not pharmacokinetic interaction between metformin and orlistat.

Chronic model of tympanic perforation in rats with mitomycin C and dexamethasone

A rat model of chronic tympanic membrane perforation was developed to be used in the search of new materials for the sealing of these perforations. A longitudinal study was carried out in rats subjected to incisional myr-ingotomy followed by the application of mitomycin C alone or with dexamethasone. Rats were checked at days 3, 7,1 0,1 4 and weekly thereafter until perforation closure, for up to 6 months. The addition of dexamethasone is a key component in order to obtain a chronic opening. Myringotomies treated w ith saline had a mean healing time of 8.5 days. At 8 weeks, between 62.5% and 77.7% of tympanic membranes treated w ith mitomycin C and dexamethasone remained perforated and at 6 months this number fell to 21.4%. This technique is able to maintain most tympanic membrane perforations patent for at least 8 weeks. This rat model is adequate for its use in preclinical or translational research.

Safety and Efficacy of Oral Mirodenafil in Mexican with Erectile Dysfunction

Erectile dysfunction is treated with 5-phospodiesterase inhibitors as Mirodenafil, which has shown its efficacy and safety in Koreans, however;no information in other populations is available. An open clinical trial study was designed to evaluate the efficacy and safety in real life of a fixed-dose of Mirodenafil in Mexican patients with erectile dysfunction. Forty-seven male patients received a 100 mg tablet of Mirodenafil, during 12 weeks. Primary outcome efficacy measure was the percentage of male patients with successful intercourse. Secondary outcomes measures included patient satisfaction, mood and self-esteem level. Safety assessments included laboratory tests, vital signs, physical examination, 12-lead electrocardiogram recordings, and incidence of adverse events by patients. Oral administration of Mirodenafil improved in an 80% - 90% the number of successful intercourses from 7 to 84 days of treatment. Moreover, patients reported a significant increment in their sexual satisfaction, mood and self-esteem. Mirodenafil treatment did not modify vital signs nor anthropometric parameters during 84 days. Mild headache was the most frequent adverse event (17.0%) and there were no severe adverse events during pharmacological treatment. Data suggest that oral Mirodenafil is safety, well tolerated and effective in the Mexican population with erectile dysfunction.

细小颗粒从纸张表面剥落及其在印刷橡皮布上的积聚(或称掉毛掉粉),是新闻纸印刷中遇到的最复杂的问题之一,且在高填料再生纸中更为严重。纳米纤维素纤维(NFC)通过增加纤维与纤维之间以及纤维内部的结合而成为高填料纸的增强剂具有巨大的潜力,目前受到极大关注。然而,添加NFC对掉毛掉粉的影响尚未得到研究。本研究的目的是量化NFC对新闻纸掉毛掉粉的影响。通过在含有14%(质量分数)灰分的再生纸的浆中添加不同浓度的PCC和NFC来评估NFC作为掉毛掉粉控制剂的效果。结果表明,添加1.5%(质量分数)的NFC,再生纸的掉毛掉粉减少了47%以上;助留助滤也得到了改善。

In the last years, with the advent of Smart Grids, many research and demonstration projects have seen the light in order to involve electric system in the implementation of advances in information and communication technologies, in order to improve network efficiency, reliability, security and quality of service. Before new implementations are performed, previous results have to be studied, and taken into account. In this line, this work presents the methodology, results and conclusions of the evaluation of the Smart Grid functionalities developed by 5 different DSOs during DISCERN project in order to select the optimal, cost-effective and most replicable solutions for the strategic development of the intelligence at medium voltage (MV) and low voltage (LV) networks.

Degradation of Gesaprim Herbicide by Heterogeneous Photocatalysis Using Fe-Doped TiO2

Fe-doped TiO2 was prepared by the sol gel method and characterized by X-ray diffraction. All the Fe-doped TiO2 were composed of an anatase crystal form. The activity of the Fe-doped TiO2 for the degradation of the gesaprim commercial herbicide (which contains atrazine as active compound and formulating agents) was studied by varying the iron content during UV (15 W), visible light and solar irradiations. The visible light came from commercial saving energy lamps (13, 15 and 20 Watts). The gesaprim degradation rate depended on the iron content in the photo catalyst. The Fe-doped TiO2 (0.5% by weight of TiO2) showed higher TOC removal under visible light and was more active than the undoped TiO2 photo catalyst under the light irradiation sources tested. Over 90% of chemical oxygen demand abatement was achieved with both UV and visible light but less time was required to decrease the chemical oxygen demand content by using the catalyst doped with iron at 0.5% under visible light. It was observed that the degradation of gesaprim increased by increasing the iron content in the catalyst under visible light.
