Calcitriol analog ZK191784 ameliorates acute and chronic dextran sodium sulfate-induced colitis by modulation of intestinal dendritic cell numbers and phenotype

AIM: to investigate the effects of ZK1916784, a low calcemic analog of calcitriol on intestinal inflammation. MEtHODS: Acute and chronic colitis was induced by dextran sodium sulfate (DSS) according to standard procedures. Mice were treated intraperitoneally with ZK1916784 or placebo and colonic inflammation was evaluated. Cytokine production by mesenterial lymph node (MLN) cells was measured by ELISA. Immunohistochemistry was performed to detect intestinal dendritic cells (DCs) within the colonic tissue, and the effect of the calcitriol analog on DCs was investigated. RESULtS: treatment with ZK191784 resulted in significant amelioration of disease with a reduced histological score in acute and chronic intestinal inflammation. In animals with acute DSS colitis, down- regulation of colonic inflammation was associated with a dramatic reduction in the secretion of the proinflammatory cytokine interferon (IFN)-γ and a significant increase in intereleukin (IL)-10 by MLN cells. Similarly, in chronic colitis, IL-10 expression in colonic tissue increased 1.4-fold when mice were treated with ZK191784, whereas expression of the th1-specific transcription factor t-beta decreased by 81.6%. Lower numbers of infiltrating activated CD11c+ DCs were found in the colon in ZK191784-treated mice with acute DSScolitis, and secretion of proinflammatory cytokines by primary mucosal DCs was inhibited in the presence of the calcitriol analog. CONCLUSION: the calcitriol analog ZK191784 demonstrated significant anti-inflammatory properties in experimental colitis that were at least partially mediated by the immunosuppressive effects of the derivate on mucosal DCs.

Is the clinical head impulse test helpful in cochlear implantation candidacy evaluation?

Heat shock protein 90 inhibition by 17-DMAG lessens disease in the MRL/Ipr mouse model of systemic lupus erythematosus

Elevated expression of heat shock protein 90 （HSP90） has been found in kidneys and serum of systemic lupus erythematosus （SLE） patients and MRLIMp-FasIprIFasJpr（MRLIIpr） autoimmune mice. We investigated if inhibition of HSP90 would reduce disease in MRL/ Ipr mice. In vitro, pretreatment of mesangial cells with HSP90 inhibitor Geldanamycin prior to immune-stimulation showed reduced expression of IL-6, IL-12 and NO. In vivo, we found HSP90 expression was elevated in MRL/Ipr kidneys when compared to C57BL/6 mice and MRIJIpr mice treated with HSP90 inhibitor 17-DMAG. MRIJIpr mice treated with 17-DMAG showed decreased proteinuria and reduced serum anti-dsDNA antibody production. Glomerulonephritis and glomerular IgG and C3 were not significantly affected by administration of 17-DMAG in MRIJIpr. 17-DMAG increased CD8＋ t cells, reduced double-negative t cells, decreased the CD4/CD8 ratio and reduced follicular B cells. these studies suggest that HSP90 may play a role in regulating t-cell differentiation and activation and that HSP90 inhibition may reduce inflammation in lupus.

Bone wound healing is a highly dynamic and precisely controlled process through which damaged bone undergoes repair and complete regeneration. External factors can alter this process, leading to delayed or failed bone wound healing. the findings of recent studies suggest that the use of selective serotonin reuptake inhibitors(SSRIs) can reduce bone mass, precipitate osteoporotic fractures and increase the rate of dental implant failure. With 10% of Americans prescribed antidepressants, the potential of SSRIs to impair bone healing may adversely affect millions of patients’ ability to heal after sustaining trauma. Here, we investigate the effect of the SSRI sertraline on bone healing through pre-treatment with(10 mg·kg-1sertraline in drinking water, n = 26) or without(control, n = 30) SSRI followed by the creation of a 5-mm calvarial defect. Animals were randomized into three surgical groups:(a) empty/sham,(b) implanted with a DermaMatrix scaffold soak-loaded with sterile PBS or(c) DermaMatrix soak-loaded with542.5 ng BMP2. SSRI exposure continued until sacrifice in the exposed groups at 4 weeks after surgery. Sertraline exposure resulted in decreased bone healing with significant decreases in trabecular thickness, trabecular number and osteoclast dysfunction while significantly increasing mature collagen fiber formation. these findings indicate that sertraline exposure can impair bone wound healing through disruption of bone repair and regeneration while promoting or defaulting to scar formation within the defect site.

Dear Editor, y-Aminobutyric acid （GABA） is a non-protein amino acid （AA） metabolized via the GABA shunt; a three-enzyme pathway that includes glutamate decarboxylase （GAD）, GABA transaminases （GABA-t）, and succinic semialdehyde dehydrogenase （SSADH）（Bown and Shelp, 1997; Bouch~ and Fromm, 2004）. the majority of work on GABA, to date, has focused on its role as an inhibitory neurotransmitter in mammals. In plants, however, due to its intermediate posi- tion between carbon （C） and nitrogen （N）, the metabolism of GABA has been suggested as a modulator of C-N balance （Fair et al., 2008）. In addition, the numerous observations of a rapid accumulation of GABA in response to （a）biotic stresses has prompted different hypotheses to be postu- lated on its role including a regulator of cytosolic pH and a herbivore deterrent （Bouche and Fromm, 2004; Roberts, 2007）. Additionally, a signaling role has been hypothesized in plants, though evidence remains elusive and the debate on the dual function of GABA as a signaling molecule and as a metabolite in plants remains ongoing. In the present work, we report changes in the metabolite and transcript profiles in Arabidopsis seedlings exposed to exogenous GABA. Liquid-grown seedlings were cultured under C and N limitation to reveal a possible role of GABA as either C or N substrate and to ascertain its influence on transcriptional programs.

Localized Prostate Cancer Patients’ Information in Urology Outpatient Practice—Results of a Nationwide Prospective Study in Germany

Purpose: Many studies demonstrate the importance of an empathic communication with prostate cancer patients and show various information needs. Most of the studies are cross sectional. the present analysis aims to 1) provide detailed prospective data on the actual quality of patient information provided by urologists in private practice in Germany;and 2) explore sociodemographic, communicative, and health- and treatment related determinants of the quality of provider-patient-communication. Methods: HAROW is a prospective, observational study designed to collect clinical data and patient reported outcomes of different treatment options (hormonal therapy, active surveillance, radiation, operation, watchful waiting) for newly diagnosed patients with localized prostate cancer under real conditions. At 6-month intervals, general clinical data, patient reported outcomes and patients’ assessment of patient-physician-communication are documented. A total of 1893 questionnaires at t0 (initial diagnosis) and t1 (six months after diagnosis) were analyzed. A linear regression model was estimated. Results: the mean age of the men was 68.38 years, and most lived with their spouse or partner (94.1%). Most patients were informed about treatment options (t0 96%, t1 93.2%), but much fewer received information about self-help groups (t0 36.4%, t1 45.8%), rehabilitation (t0 59.9%, t1 68%), and second medical opinion (t0 57.2%, t1 59.3%). Older men (p = 0.02) and men living alone (p = 0.048) received less information than others. Physician empathy (p = 0.000) and prostatectomy treatment (p = 0.020) were positively associated with receiving more information. Conclusion: there is room for improvement in terms of informing prostate cancer patients about rehabilitation, second medical opinion and self-help groups. Urologists being empathetic share more information with their patients. Some patients receive less information than others, especially older patients living alone.

Incidence of Subclinical Hypothyroidism in Cardiac Surgery Patients. Comparison of Presentation Characteristics, Hospital and Medium-term Outcomes with Euthyroid Patients

Subclinical hypothyroidism (SCHt) is common, with an occurrence of up to 10% of the adult population and defined biochemically only by elevated tSH and normal t4. SCHt affects negatively on lipid and carbohydrate metabolism increasing the risk of ischemic heart disease, affects negatively on cardiac performance and have a close correlation with renal function. the aim of this study is to compare presentation characteristics and outcomes between euthyroid pts and pts with SCHt who underwent cardiac surgery. Methods: 474 pts from June 2003 through September 2004 had tSH and t4 measured. 365 pts were euthyroid (Group 1), 41 pts had SCHt (Group 2). Groups were compared by demographics and EuroSCORE (ES) risk profiles. Operative and hospital outcomes were compared as was follow-up mortality up to 96 months. Results: there were more females in Group 2, p = 0.04, more pts with CHF and number of NYHA III-IV pts (p ts in Group 2 had elevated s-crea (p trial fibrillation (p = 0.007). Comparing the Groups by EuroSCORE (ES) showed higher risk scores in Group 2 pts (Additive ES 6.8 vs 8.5 and Logistic ES 12.3% vs 18.1%, p = 0.01 and 0.03). Hospital mortality was higher in Group 2 (12.2% vs 4.1%, p = 0.04) and the number of pts needing extended care was higher in Group 2 (p = 0.01). Follow up mortality was doubled in Group 2 pts up to 96 months compared to Group 1 (p tation characteristics and risk scores are different and worse in SCHt pts compared with euthyroid pts. Hospital and follow-up mortality are increased in SCHt pts.

trong>Objectivestrong>: This retrospective study evaluated 1) benefits of single nucleotide polymorphism (SNP)-based chromosomal microarrays (CMAs) in the diagnosis of complete hydatidiform mole (CHM) and partial HM (PHM) in products of conception (POC) and amniotic fluid (AF) specimens, and 2) frequency of whole-genome uniparental disomy (wgUPD) and triploidy in POC and AF specimens received at a US national reference laboratory. Methods: We reviewed consecutive 2138 POC and 3230 AF specimens and identified the cases with wgUPD and triploidy which are associated with molar pregnancy. Results: Of 2138 consecutive POC specimens tested, SNP-based CMA detected wgUPD in 10 (0.47%) and triploidy in 84 (3.93%). Of the 10 wgUPD cases, 9 (90%) were confirmed as CHM. Of 3230 consecutive AF specimens, the array detected wgUPD in 1 case (0.03%) and triploidy in 11 (0.34%). Conclusions: SNP-based microarray allows detection of wgUPD in POC and AF specimens at a US national reference laboratory. Correctly diagnosing HM and differentiating CHM from PHM are important for clinical management. The effective SNP-based CMA detection of wgUPD in CHM may enable physicians to monitor patients at risk for gestational trophoblastic disease and neoplasm. Conventional chromosome analysis of POC has a high failure rate, cannot be performed on formalin-fixed paraffin embedded samples, and cannot detect wgUPD. Further multi-institutional collaborative assessmen on accuracy, cost-effectiveness, and adequate access to SNP-based CMA, may lead this testing platform to be considered as the first-tier analysis tool for POC specimens, including those showing PHM or CHM.

Meridional Distributions of Historical Zonal Averages and their Use to Quantify the Global and Spheroidal Mean Near-Surface temperature of the terrestrial Atmosphere

the zonal averages of temperature (the so-called normal temperatures) for numerous parallels of latitude published between 1852 and 1913 by Dove, Forbes, Ferrel, Spitaler, Batchelder, Arrhenius, von Bezold, Hopfner, von Hann, and Börnstein were used to quantify the global (spherical) and spheroidal mean near-surface temperature of the terrestrial atmosphere. Only the datasets of Dove and Forbes published in the 1850s provided global averages below 〈t〉=14°C, mainly due to the poor coverage of the Southern Hemisphere by observations during that time. the global averages derived from the distributions of normal temperatures published between 1877 and 1913 ranged from 〈t〉=14.0°C (Batchelder) to 〈t〉=15.1°C (Ferrel). the differences between the global and the spheroidal mean near-surface air temperature are marginal. to examine the uncertainty due to interannual variability and different years considered in the historic zonal mean temperature distributions, the historical normal temperatures were perturbed within ±2σ to obtain ensembles of 50 realizations for each dataset. Numerical integrations of the perturbed distributions indicate uncertainties in the global averages in the range of ±0.3°C to ±0.6°C and depended on the number of available normal temperatures. Compared to our results, the global mean temperature of 〈t〉=15.0°C published by von Hann in 1897 and von Bezold in 1901 and 1906 is notably too high, while 〈t〉=14.4°C published by von Hann in 1908 seems to be more adequate within the range of uncertainty. the HadCRUt4 record provided 〈t〉≌?13.7°C for 1851-1880 and 〈t〉=13.6°C for 1881-1910. the Berkeley record provided 〈t〉=13.6°C and 〈t〉≌?13.5°C for these periods, respectively. the NASA GISS record yielded 〈t〉=13.6°C for 1881-1910 as well. these results are notably lower than those based on the historic zonal means.
