Effects of Seed Pretreatment on Germination, Growth and Yield of <i>Momordica charantia</i>L.

Momordica charantia is a plant species widely used in food and traditional medicine. However, the germination techniques and the performances of M. charantia are poorly understood by the populations. This study aims to better understand the germination, growth and development characteristics, and yield of M. charantia. Three pretreatments of the seeds are used for the shortening of the pre-germination period: soaking the seeds in tap water for 24 hours (batch 2), in hot water at 80˚C (batch 3), for 24 hours and in the boiling water at 100˚C for 24 hours (batch 4). A control without soaking (batch 1) was also carried out. The seeds were sown in plastic boxes in the laboratory with sterile sand and directly in the nursery in polyethylene sleeves. The influence of the light factor (light and dark) and watering frequency (every day and every two days) on the growth and yield parameters was evaluated respectively in the laboratory and in the nursery. Germination, growth and yield parameters were determined. The results show that the light and frequency of watering factors have no influence (p > 0.05) on the rate and duration of seed germination unlike pretreatments. Thus, Batch1 and 2 exhibit significantly higher germination rates (70% ± 16%) than Lots 3 and 4 (21% ± 15%) in the laboratory and in the nursery. On the other hand in terms of germination time, batch 3 and 4 displayed shorter durations (14 days) than those batch1 and 2 (23 days). Growth parameters and yield (fruit and seeds) are strongly influenced by watering frequency, unlike pretreatments which only affect yield. Growth and yield parameters are significantly higher with daily watering than daily watering. Ultimately, this study showed that pretreatments have an influence on the duration and rate of germination as well as the frequency of watering on the performance of M. charantia

Bipolar Leads for Prevention of Phrenic Nerve Stimulation: Results from the ORPHEE Observational Study

Background: Up to one in three patients implanted with a cardiac resynchronization therapy-defibrillator (CRT-D) device experience phrenic nerve stimulation (PNS). Quadripolar leads are effective at reducing PNS, but compared to standard bipolar leads they have limitations related to maneuverability and high pacing thresholds. The ability of standard bipolar leads to overcome PNS is explored here. Methods: The French multicenter, observational study ORPHEE enrolled 90 CRT-D-eligible patients. Detection of PNS took place after satisfactory positioning of the LV bipolar lead (stable pacing threshold - LV ring, LV tip - RV ring and LV ring - RV coil) could prevent PNS from occurring in at least 90% of patients. Results: In 80 evaluable patients, PNS was reported in 12 patients (15%). Reprogramming overcame PNS in 10 patients: LV ring - RV coil in 8 patients;LV tip - LV ring in 1;and LV tip- RV ring in 1. As PNS was avoided in 78 of 80 patients (97.5%), the primary endpoint was significant (97.5% vs. 90%, p = 0.01). Conclusion: During CRT-D implantation, PNS occurred in 15% of patients. In most (97.5%) implanted patients, PNS could be avoided by vector reprogramming using a bipolar LV lead. For patients whose coronary sinus anatomy precludes the implantation of multi-electrode leads, bipolar leads are a suitable, reliable alternative.

Macrozoospermia,characterized by the presence of largeheaded spermatozoa usually carrying several flagella,is one of the most severe phenotypes of male *** in most cases,the gametes are chromosomally abnormal and cannot be used for assisted reproductive technologies(ART)(Ray et al.,2017).In 2007,subjects from consanguineous families were shown to carry the same homozygous deleterious AURKC variant(NM_001015878.2:c.144delC,later renamed c.145delC,***49TrpfsTer23),establishing AURKC as the first and main gene associated with macrozoospermia(Dieterich et al.,2007).Using flow cytometry,all spermatozoa from patients homozygous for the c.145delC variant were shown to be tetraploid,highlighting a cytokinesis blockage of the first meiotic divisions thus confirming that ART cannot be successful for these patients(Dieterich et al.,2009).

Dear editor,Radiotherapy is a cornerstone in the management of anal or rectal cancer. Because elderly patients are often excluded from clinical trials, little is known about radiotherapy’s therapeutic index (efficacy/toxicity ratio) in the geriatric population [1]. Still, the ageing of population imposes the challenge to treat older cancer patients and probably to adjust their treatment [2]. A few studies reported data on radiation-induced toxicities in nonagenarian patients, but data on efficacy are still scarce. For rectal cancer, preoperative radiotherapy programs were suggested to be feasible in elderly patients [3–5], with a preference for short protocols without chemotherapy [4]. For anal cancer, the safety of radiochemotherapy was only reported in limited sets of elderly patients [6]. The objective of the present study was to report efficacy and toxicity data on the radiotherapy treatment of nonagenarian patients with anal or rectal *** analyzed the data of 34 nonagenarian patients with anal or rectal cancers: 27 (79.4%) with a rectal adenocarcinoma and 7 (20.6%) with an anal canal squamous cell carcinoma.

Arsenic trioxide(As2O3)is recently found to have therapeutic potential in systemic sclerosis(SSc),a life-threatening multi-system fibrosing autoimmune disease with type I interferon(IFN-I)*** activated plasmacytoid dendritic cells(pDCs)are responsible for IFN-I secretion and are closely related with fibrosis establishment in *** this study,we showed that high concentrations of As2O3 induced apoptosis of pDCs via mitochondrial pathway with increased BAX/BCL-2 ratio,while independent of reactive oxygen species ***,at clinical relevant concentrations,As2O3 preferentially inhibited IFN-αsecretion as compared to other cytokines such as TNF-α,probably due to potent down-regulation of the total protein and mRNA expression,as well as phosphorylation of the interferon regulatory factor7(IRF7).In addition,As2O3 induced a suppressive phenotype,and in combination with cytokine inhibition,it down-regulated pDCs’capacity to induce CD4+T cell proliferation,Thl/Th22 polarization,and B cell differentiation towards ***,chronically activated pDCs from SSc patients were not resistant to the selective IFN-αinhibition,and regulatory phenotype induced by ***,our data suggest that As2O3 could target pDCs and exert its treatment efficacy in SSc,and more autoimmune disorders with IFN-I signature.

No consensus exists regarding the role of radiotherapy in the management of gynecologic cancer in nonagenarian *** retrospectively reviewed the outcomes of 19 consecutive nonagenarian patients with gynecologic cancer(6 endometrial cancers,6 cervical cancers,4 vulvar cancers,and 3 vaginal cancers) who were treated with *** was performed mainly in a palliative setting(n = 12;63.2%),with a median dose of 45 Gy(range,6-76 Gy).Infrequent major acute or late toxicities were *** 19 patients,9(47.4%) experienced tumor progression,5(26.3%) experienced complete response,2(10.5%) experienced stable disease and/or partial *** last follow-up,12 patients(63.2%) had died;most deaths(n = 9) occurred because of the *** results suggest that radiotherapy is feasible in the treatment of nonagenarian patients with gynecologic cancer.

Radiotherapy is a cornerstone of anticancer treatment. However in spite of technical evolutions, important rates of failure and of toxicity are still reported. Although numerous pre-clinical data have been published, we address the subject of radiotherapy-stem cells interaction from the clinical efficacy and toxicity perspective. On one side, cancer stem cells(CSCs) have been recently evidenced in most of solid tumor primary locations and are thought to drive radio-resistance phenomena. It is particularly suggested in glioblastoma, where CSCs were showed to be housed in the subventricular zone(SVZ). In recent retrospective studies, the radiation dose to SVZ was identified as an independent factor significantly influencing overall survival. On the other side, healthy tissue stem cells radio-destruction has been recently suggested to cause two of the most quality of life-impacting side effects of radiotherapy, namely memory disorders after brain radiotherapy, and xerostomia after head and neck radiotherapy. Recent publications studying the impact of a radiation dose decrease on healthy brain and salivary stem cells niches suggested significantly reduced long term toxicities. Stem cells comprehension should be a high priority for radiation oncologists, as this particular cell population seems able to widely modulate the efficacy/toxicity ratio of radiotherapy in real life patients.
