Thalidomide ameliorates portal hypertension via nitric oxide synthase independent reduced systolic blood pressure

AIM: Portal hypertension is a common complication of liver cirrhosis and significantly increases mortality and *** reports have suggested that the compound thalidomide attenuates portal hypertension(PHT).However, the mechanism for this action is not fully *** hypothesis is that thalidomide destabilizes tumor necrosis factor α(TNFα) mR NA and therefore diminishes TNFα induction of nitric oxide synthase(NOS) and the production of nitric oxide(NO).To examine this hypothesis, we utilized the murine partial portal vein ligation(PVL) PHT model in combination with endothelial or inducible NOS isoform gene knockout ***: Wild type, inducible nitric oxide synthase(i NOS)-/- and endothelial nitric oxide synthase(e NOS)-/-mice received either PVL or sham surgery and were given either thalidomide or *** nitrate(total nitrate, NOx) was measured daily for 7 d as a surrogate of NO *** TNFα level was quantified by enzyme-linked immunosorbent ***α m RNA was quantified in liver and aorta tissue by reverse transcription-polymerase chain *** was determined by recording splenic pulp pressure(SPP) and abdominal aortic flow after 0-7 *** to thalidomide was determined by measurement of SPP and mean arterial pressure(MAP).ReSULTS: SPP, abdominal aortic flow(Qao) and plasma NOx were increased in wild type and i NOS-/-PVL mice when compared to sham operated control *** contrast, SPP, Qao and plasma NOx were not increased in e NOS-/- PVL mice when compared to sham *** TNFα level in both sham and PVL mice was below the detection limit of the commercial eLISA ***, the effect of thalidomide on serum TNFα levels was undetermined in wild type, e NOS-/-or i NOS-/- *** acutely increased plasma NOx in wild type and e NOS-/- mice but not i NOS-/-***, thalidomide temporarily(0-90 min) decreased mean arterial pressure, SPP and Qao in wild type, e NOS-/- and i NOS-/- PVL mice, aft

Characterization of Gibberellin Receptor Mutants of Barley （Hordeum vulgare L.）

The sequence of Gidl （a gene for a gibberellin （GA） receptor from rice） was used to identify a putative orthologue from barley. This was expressed in e. coil, and produced a protein that was able to bind GA in vitro with both structural specificity and saturability. Its potential role in GA responses was investigated using barley mutants with reduced GA sensitivity （gsel mutants）. Sixteen different gsel mutants each carried a unique nucleotide substitution in this sequence. In all but one case, these changes resulted in single amino acid substitutions, and, for the remaining mutant, a substitution in the 5＇ untranslated region of the mRNA is proposed to interfere with translation initiation. There was perfect linkage in segregating populations between new mutant alleles and the gsel phenotype, leading to the conclusion that the putative GID1 GA receptor sequence in barley corresponds to the Gsel locus. Determination of endogenous GA contents in one of the mutants revealed enhanced accumulation of bioactive GA1, and a deficit of C20 GA precursors. All of the gsel mutants had reduced sensitivity to exogenous GA3, and to AC94377 （a GA analogue） at concentrations that are normally ＇saturating＇, but, at much higher concentrations, there was often a considerable response. The comparison between barley and rice mutants reveals interesting differences between these two cereal species in GA hormonal physiology.

environmental sampling of volatile organic compounds during the 2018 Camp Fire in Northern California

Trace analysis of volatile organic compounds(VOCs) during wildfires is imperative for environmental and health risk assessment. The use of gas sampling devices mounted on unmanned aerial vehicles(UAVs) to chemically sample air during wildfires is of great interest because these devices move freely about their environment, allowing for more representative air samples and the ability to sample areas dangerous or unreachable by humans. This work presents chemical data from air samples obtained in Davis, CA during the most destructive wildfire in California's history-the 2018 Camp Fire – as well as the deployment of our sampling device during a controlled experimental fire while fixed to a UAV. The sampling mechanism was an in-house manufactured micro-gas preconcentrator(μPC) embedded onto a compact battery-operated sampler that was returned to the laboratory for chemical analysis. Compounds commonly observed in wildfires were detected during the Camp Fire using gas chromatography mass spectrometry(GC–MS), including BTeX(benzene, toluene, ethylbenzene, m + p-xylene, and o-xylene), benzaldehyde, 1,4-dichlorobenzene, naphthalene, 1,2,3-trimethylbenzene and 1-ethyl-3-methylbenzene. Concentrations of BTeX were calculated and we observed that benzene and toluene were highest with average concentrations of 4.7 and 15.1 μg/m^(3), respectively. Numerous fire-related compounds including BTeX and aldehydes such as octanal and nonanal were detected upon experimental fire ignition, even at a much smaller sampling time compared to samples taken during the Camp Fire. Analysis of the air samples taken both stationary during the Camp Fire and mobile during an experimental fire show the successful operation of our sampler in a fire environment.

The mechanisms of color production in black skin versus red skin on the heads of New World vultures

A determination of how the color of animal integument is produced is a starting point for investigations into the function and evolution of *** mechanisms that give rise to the color of bare skin of New World vultures are largely ***,we investigate the source of color production in the bare skin of Turkey Vultures(Cathartes aura)and Black Vultures(Coragyps atratus).Using UV-vis reflectance spectroscopy,we found evidence that hemoglobin is the primary pigment responsible for the red coloration of the bare skin on the heads of Turkey Vultures,and that eumelanin is responsible for the black coloration of the bare skin on the heads of Black *** microscopy of incisional skin samples further supported these mechanisms of color production by revealing the presence of numerous blood vessels near the surface of the Turkey Vulture skin,and a high concentration of melanosomes in the skin of Black *** high performance liquid chromatography(HPLC),we detected carotenoids within the skin of both species with significantly higher total concentrations of carotenoids in the skin of Turkey Vultures compared to the skin of Black *** carotenoids detected were dietary carotenoids that typically produce yellow coloration when accumulated in integument and were present in low *** hypothesize that the dietary carotenoids present do not contribute to the color of the skin,but rather help to compensate for the lack of melanosomes found in Turkey Vulture *** presence of additional carotenoids may act as an antioxidant to minimize UV damage when the bare Turkey Vulture head skin is exposed to direct sunlight for prolonged periods of time when soaring and scavenging for food.

Genome editing is a transformational technology—by precisely altering the coding or regulatory sequence of a specific gene,it is now possible to change specific traits within a wide variety of *** a consequence,plant breeding can be taken to a new level of precision,and many countries are moving rapidly to adopt new legislation to permit genome editing(Greenwood et al.,2023).However,genome editing has so far seldom been used to develop disease-resistant *** is because plant immunity often depends on single dominant resistance genes,which encode immune receptors that recognize secreted effectors deployed by plant pathogens(Jones and Dangl,2006).In this context,genetic modification has proven to be a much more powerful technology so far to introduce these genes from distinct varieties,or even different species,to develop disease resistant crop species(Greenwood et al.,2023),especially when multiple disease-resistance loci can be introduced together to provide more durable resistance(Luo et al.,2021).Genome editing can be used in an innovative way to resurrect previously effective resistance genes(Contreras et al.,2023),but it has been less effective at introducing new forms of disease resistance to crops.

Governance in the exploration of global and regional determinants of ICT development

The present study assesses how governance affects information and communication technology(IcT)at the global level contingent on macroeconomic policy factors such as trade,foreign direct investment(FDI),manufacturing value added,and agricultural value *** study focuses on 183 countries from 2003 to 2021,and the empirical evidence is based on the generalized method of moments(GMM).The following main findings are *** the full sample,governance unconditionally promotes ICT development,while trade openness(industrial added value)moderates governance to promote(dampen)ICT *** sub-Saharan Africa,only trade openness effectively moderates governance to induce an overall positive effect on ICT,while in the Middle east and North Africa(MeNA)region,all policy variables moderate governance for an overall positive incidence on ICT sector *** findings for the MeNA region are confirmed in the europe and Central Asia(eCA)region,with the exception of the moderating role of industrial added value,which engenders an overall negative *** the east and South Asia and the Pacific(eSAP)countries,one overall positive incidence is apparent in the role of trade openness,while net negative effects are established from the moderating roles of industrial added value and agricultural added *** the American sub-sample,a positive(negative)net effect is apparent from the role of industrial added value(trade)in moderating the incidence of gover-nance on ICT sector ***,policy implications are discussed.

Separations of Metal Ions Using Ionic Liquids: The Challenges of Multiple Mechanisms

Ionic liquids are a distinct sub-set of liquids, comprising only of cations and anions, often with negligible vapor pressure. As a result of the low or non-volatility of these fluids, ionic liquids are often considered in liquid/liquid separation schemes where the goal is to replace volatile organic solvents. Unfortunately, it is often not yet recognized that the ionic nature of these solvents can result in a variety of extraction mechanisms, including solvent ion-pair extraction, ion exchange, and simultaneous combinations of these. This paper discusses current ionic liquid-based separations research where the effects of the nature of the solvent ions, ligands, and metal ion species were studied in order to be able to understand the nature of the challenges in utilizing ionic liquids for practical applications.
