Dehydroepiandrosterone (DHEA), the most abundant adrenal androgen in primates, is also synthesized from cholesterol in the brain. Like testosterone, DHEA induces spine synapse formation in the hippocampus. In female rats, this response is blocked by co-administration of an inhibitor of aromatase, the enzyme responsible for estrogen biosynthesis. In males, by contrast, the hippocampal synaptic response to DHEA is unaffected by treatment with an aromatase inhibitor. We hypothesized that this sex difference might reflect differential dependence of the hippocampal responses on subcortical afferents from the basal forebrain. To test this hypothesis, we examined the effects of unilateral fimbria/ fornix transection (FFX) on DHEA-induced synapse formation in the cornu ammonis 1 (CA1) hippocampal subfield of gonadectomized female and male rats. In ovariectomized females, CA1 spine synapse density after DHEA treatment was reduced by more than 60% ipsilateral to FFX. In males, however, unilateral FFX transection had no effect on spine synapse density after DHEA treatment. These results suggest that sex differences in the dependence on local estrogen biosynthesis of the CA1 synaptic response to androgen may at least in part be the result of sex differences in the relative contributions of afferents to the hippocampus from the basal forebrain.

The synthesis of fluorescent nanomaterials has received considerable attention due to the great potential of these materials for a wide range of applications, from chemical sensing through bioimaging to optoelectron- ics. Herein, we report a facile and scalable approach to prepare fluorescent carbon dots （FCDs） via a one-pot reaction of citric acid with ethylenediamine at 150 ℃ under ambient air pressure. The resultant FCDs pos- sess an optical bandgap of 3.4 eV and exhibit strong excitation-wavelength-independent blue emission （λEm = 450 nm） under either one- or two-photon excitation. Owing to their low cytotoxicity and long fluorescence lifetime, these FCDs were successfully used as internalized fluorescent probes in human cancer cell lines （HeLa cells） for two-photon excited imaging of cells by fluorescence lifetime imaging microscopy with a high-contrast resolution. They were also homogenously mixed with commercial inks and used to draw fluo- rescent patterns on normal papers and on many other substrates （e.g., certain flexible plastic films, textiles, and clothes）. Thus, these nanomaterials are promising for use in solid-state fluorescent sensing, security labeling, and wearable optoelectronics.

Hepatic lipogranulomas in patients with chronic liver disease: Association with hepatitis C and fatty liver disease

AIM: To study the significance and clinical implication of hepatic lipogranuloma in chronic liver diseases, including fatty liver disease and hepatitis C. METHODS: A total of 376 sequential, archival liver biopsy specimens were reviewed. Lipogranuloma, steatosis and steato-fibrosis were evaluated with combined hematoxylin and eosin and Masson’s trichrome staining. RESULTS: Fifty-eight (15.4%) patients had lipogranuloma, including 46 patients with hepatitis C, 14 patients with fatty liver disease, and 5 patients with other diseases. Hepatic lipogranuloma was more frequently seen in patients with hepatitis C (21%) and fatty liver disease (18%), and its incidence was significantly higher than that in control group (P < 0.0002 and P < 0.007, respectively). In addition, 39 out of the 58 patients with lipogranuloma were associated with steatosis and/or steato-fibrosis. Of the 18 lipogranuloma patients with clinical information available for review, 15 (83%) had risk factors associated with fatty liver disease, such as alcohol use, obesity, hyperlipidemia, and diabetes mellitus. Although the incidence of these risk factors was greater in patients with lipogranuloma than in control group (60%), it did not reach statistical significance. CONCLUSION: Hepatic lipogranuloma is not limited to mineral oil use and commonly associated with hepatic steatosis, hepatitis C and fatty liver disease. With additional histological features of steato-fibrosis, lipogranuloma can also be used as a marker of prior hepatic steatosis.

In cereals, the presence of soluble polysaccharides including（1,3;1,4）-b-glucan has downstream implications for human health, animal feed and biofuel applications. Sorghum bicolor（L.） Moench is a versatile crop, but there are limited reports regarding the content of such soluble ***, the amount of（1,3;1,4）-b-glucan present in sorghum tissues was measured using a Megazyme assay. Very low amounts were present in the grain, ranging from 0.16%–0.27%（w/w）, while there was a greater quantity in vegetative tissues at 0.12–1.71%（w/w）. The fine structure of（1,3;1,4）-b-glucan, as denoted by the ratio of cellotriosyl and cellotetraosyl residues,was assessed by high performance liquid chromatography（HPLC） and ranged from 2.6–3：1 in the grain, while ratios in vegetative tissues were lower at 2.1–2.6：1. The distribution of（1,3;1,4）-b-glucan was examined using a specific antibody and observed with fl uorescence and transmission electron microscopy. Micrographs showed a variable distribution of（1,3;1,4）-b-glucan in fl uenced by temporal and spatial factors. The sorghum orthologs of genes implicated in the synthesis of（1,3;1,4）-b-glucan in other cereals, such as the Cellulose synthase-like（Csl） F and H gene families were de *** pro filing of these genes across sorghum tissues was carried out using real-time quantitative polymerase chain reaction, indicating that, as in other cereals, Csl F6 transcripts dominated.

This is the first report on a polysaccharide(RCj2-Ib)isolated from Rosa Chinensis ***2-Ib was obtained through the extraction with water,precipitation with ethanol,separation with DEAE-52 column and purification with DEAE-Sepharose Fast Flow column and Sephadex G100 ***,FT-IR and NMR analyses revealed that RCj2-Ib(3.3 k Da)was a 1,4-linked polymannuronic acid containing substantialβ-Danomers *** anticoagulant effect of RCj2-Ib evaluated by using rabbit ear venous blood and an acute blood stasis rat model showed that RCj2-Ib had obvious anticoagulant activity in regulating endogenous and exogenous coagulation pathways and reducing serum thromboxane B2 and *** addition,RCj2-Ib could also increase the number of Lactobacillus and Escherichia *** a result,RCj2-Ib has the potential to inhibit thrombosis and maintain the intestinal environment.

Assessing Chemical Mixtures and Human Health: Use of Bayesian Belief Net Analysis

Background: Despite humans being exposed to complex chemical mixtures, much of the available research continues to focus on a single compound or metabolite or a select subgroup of compounds inconsistent with the nature of human exposure. Uncertainty regarding how best to model chemical mixtures coupled with few analytic approaches remains a formidable challenge and served as the impetus for the study. Objectives: To identify the polychlorinated biphenyl (PCB) congener(s) within a chemical mixture that was most associated with an endometriosis diagnosis using novel graphical modeling techniques. Methods: Bayesian Belief Network (BBN) models were developed and empirically assessed in a cohort comprising 84 women aged 18 - 40 years who underwent a laparoscopy or laparotomy between 1999 and 2000;79 (94%) women had serum concentrations for 68 PCB congeners quantified. Adjusted odds ratios (AOR) for endometriosis were estimated for individual PCB congeners using BBN models. Results: PCB congeners #114 (AOR = 3.01;95% CI = 2.25, 3.77) and #136 (AOR = 1.79;95% CI = 1.03, 2.55) were associated with an endometriosis diagnosis. Combinations of mixtures inclusive of PCB #114 were all associated with higher odds of endometriosis, underscoring its potential relation with endometriosis. Conclusions: BBN models identified PCB congener 114 as the most influential congener for the odds of an endometriosis diagnosis in the context of a 68 congener chemical mixture. BBN models offer investigators the opportunity to assess which compounds within a mixture may drive a human health effect.
