Effects of Some Thiol Chelators on Enzymatic Activities in Blood, Liver and Kidneys ofAcute Arsenic (Ⅲ) Exposed Mice

The effects of meso 2, 3-dimercaptosuccinic acid (DMSA), sodium 2, 3-dimercaptopropane 1-sulfonate (DMPS) and S-adenosyl L-methionine (SAM) on the enzymatic activities of mice were studied. The mice were given intraperitoneal (i.p.) injections of these chelating agents (1 mmol/kg) and 3 h later the activity of delta-aminolevulinic acid dehydratase (ALAD) in the blood, and aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma-glutamyltranspeptidase (gamma-GT), alkaline phosphatase (ALP) in the liver and kidney were determined. The activity of blood ALAD was significantly increased by the administration of DMSA and SAM while DMPS had only a moderate effect. The activities of other hepatic enzymes changed little when the mice were treated with these chelating agents, except for a significant reduction in hepatic ALP activity following DMPS administration. Arsenic (III) administration markedly increased the activities of ALT and ALP in the liver and kidneys. The changes in the enzymatic activities by treatment with arsenic were prevented by injection of DMSA, DMPS and SAM, DMSA being the most effective. These results indicate that DMSA, DMPS and SAM were not toxic to the liver or kidneys of mice and that treatment with DMSA is more effective than DMPS or SAM in protecting mice from acute hepatic or renal toxicity caused by arsenic.

Thalassemia is associated with low antioxidant enzyme deficiency especially glutathione peroxidase. GPX exists in 6 isomeric forms out of which GPX1 Single Nucleotide Polymorphism is found to be associated with Thalassemia major. In our study, the determination of the allelic frequency and phenotype of a common polymorphism in Se-dependent glutathione peroxidase 1 (GPX1) was observed in Thalassemic populations. A proline/leucine variant occurs at position 197 close to the C-terminus of the protein. The genotypes encoding Pro/Pro, Pro/Leu, and Leu/ Leu are distributed according to the Hardy-Weinberg relationship. The study has been carried out in 40 Thalassemic cases and 40 control subjects. No significant association between allele frequency and risk to get fatal was evident. Erythrocyte GPX activity was determined and no significant differences were obtained between the genotypes. It can be concluded that the Pro/Leu genetic variation does not appear to compromise the defense against oxidative stress in red blood cells or to be associated with significant pathology.

Efficacy of Haemocoagulase as a Topical Haemostatic Agent after Minor Oral Surgical Procedures—A Prospective Study

Purpose: Haemocoagulase is a topical haemostatic agent which provides the adequate haemostasis after minor oral surgical procedures and it has also been proved to be beneficial in promoting wound healing. The aim of this study was to check the efficacy of haemocoagulase in stopping the bleeding and its effect over wound healing after the minor oral surgical procedure. Material & Method: This study is comprised of 150 surgical sites in 75 patients. The subjects were divided into 2 groups in which Group 1 consists of 50 surgical sites in 25 patients and Group II consists of 100 surgical sites in 50 patients. Group I comprised of the group of simple extraction. In these patients one tooth socket was selected as haemocoagulase site and the other socket was the control group in which no drug was used to control haemorrhage. Group II comprised of the group of patients with bilateral impactions. 50 sockets and surgical sites were sprinkled with Haemocoagulase, and 50 sockets and surgical sites were used as control side in which no drug was used to control haemorrhage. Results: In Group I bleeding was stopped with the average time of 1.35 minutes, while at control side bleeding was stopped with the average time of 2.25 minutes. In Group II bleeding was stopped with average time for haemostasis being 1.46 minutes, while at control side the bleeding was stopped in an average time of 2.43 minutes. Conclusion: Haemocoagulase after minor oral surgery not only provides faster haemostasis but also enhances healing.

Clinical evaluation of HPV DNA test combined with liquid-based cytology in the diagnosis of cervical disease

Objectives: To evaluate the sensitivity and accuracy of the HPV DNA test in conjunction with thin prep cytology test as a screening method of human papillomavirus (HPV) infection. To study either the cervical erosion is related to high risk HPV infection or to determine the mean age distribution that is more prone to HPV infection. Material and Methods: The study is a retrospective cohort implemented to determine the real performance of liquid based medium and HPV DNA testing combined in second clinical hospital of Jilin University Changchun, China. The study group included total 150 patients from January 1, 2011 to December 30, 2012. A computerized search identified patients with thin prep test results and high risk HPV DNA testing during a 2-year period was recruited. The patients were chosen after proper speculum examination followed by thin prep cytology (TCT) and HPV DNA test. Cytologic specimens were obtained with endocervical brush, which was rinsed into the vial of Cytyc. The residual samples after the cytology report were taken for reflex HPV DNA test. The manufacture protocol was followed for HPV DNA testing using Hybrid Capture II. Colposcopic biopsy was performed for the diagnosis purpose, in patients who had atypical squamous cells of undeter-mined significance (AUS-US), low grade intraepithelial lesion (LSIL) or high-grade intraepithelial lesion (HSIL) in cytology and with positive results of highrisk HPV DNA. The diagnostic criteria were based on the Bethesda System (TBS). Findings: The high risk HPV positive women with abnormal cytology had a CIN I risk of 73 (86%), whereas 35 (23.3%) high-risk HPV positive women out of 109 (72.7%) normal cytology who underwent histological biopsy had CIN I 16 (10.7%). The risk for cervical intraepithelial neoplasia (CIN) in women with high-risk HPV positive with normal cytology was higher among women invited for the first time 31 - 40 years of age 12 (8%) than among older women 1 (0.7%). Chondrosarcoma is a malignant neoplasm that results in abnormal bone and cartilage growth. Although Chondrosarcoma is rare, it is the second most common primary bone malignancy. Mesenchymal Chondrosarcoma (MCS) is a rare histological variant of Chondrosarcoma (CS) that accounts for only 1% of all CS and has high predilection for the head and neck region with a peak incidence in 2nd and 3rd decade of life. There is comparable male to female ratio with two thirds of the cases arising from bone and one third from soft tissue. The tumor is unique because of its aggressive growth with a high tendency for late recurrence and delayed metastasis. We present a case of this highly malignant lesion arising from the infratemporal fossa and rapidly involving maxillary tuberosity and mandibular ramus.

目的左心房容积(LAV)是定量评价左心房大小的准确方法。常用的二维超声心动图(2DE)测量方法包括双平面面积长度(A-L)法和双平面Simpson’s法。而A-L法公式中所采用的长度L在临床上不尽一致:心尖四腔心和两腔心切面中较短的长度(SL)、较长的长度(LL)或二者的平均长度(AL)。本研究以实时三维超声心动图(3DE)测量的LAV为对比标准,对比不同2DE方法测量LAV的准确性。方法对100例行常规超声心动图检查的门诊病人进行前瞻性研究。年龄(59.0±15.2)岁,男性占44.0%。所有病人均为窦性心律,分别记录标准的心尖四腔心和两腔心2DE图像,清晰显示左心房轮廓,尽量避免左心房的短切,记录全容积3DE图像。分别应用双平面A-L法、双平面Simpson’s法和3DE法进行脱机测量LAV。采用配对t检验,线性回归和Bland-Altman方法进行对比分析。结果 3DELAV测值为(61.3±22.2)ml,双平面Simpson’s法和双平面A-L法测量的LAV均与3DE-LAV高度相关(r=0.93～0.94,P均<0.01)。双平面Simpson’s法与3DE的一致性[平均差=(1.1±8.2)ml,P=0.20)]明显优于A-L法。与3DE-LAV相比较,所有三种长度的双平面A-L法均显著高估LAV[SL:(7.2±9.1)ml,LL:(3.4±8.7)ml,AL:(5.2±8.8)ml,P均<0.01],其中以美国超声心动图学会(ASE)推荐的SL法高估LAV最为明显。无论是四腔心还是两腔心切面,A-L法公式中的长度L均显著低于Simpson’s法中的长度[四腔心:(51.2±7.4)ml vs(55.1±6.8)ml,两腔心:(55.2±6.3)ml vs(55.2±6.3)ml,P均<0.001]。双平面Simpson’s法测量LAV的观察者间和观察者内的变异最佳(4.5%和2.8%),其次是3DE(5.4%和4.8%),而双平面A-L法测量的变异最大(11.0%～12.0%和6.8%～7.5%)。结论双平面Simpson’s法和所有双平面A-L法测量的LAV与3DE-LAV均高度相关。采用不同长度L的A-L法均显著高估LAV,尤其是临床上最为常用的SL法的高估程度最为明显。而双平面Simpson’s法具有比A-L法更高的可重复性和与3DE-LAV更好的一致性,应作为临床上测量LAV的首选方法。

For the first time we report quantitative structure activity relationship (QSAR) studies based on Kier-Hall Electrotopological State (E-State) Indices for Dihydroalkoxybenzyloxopyrimidines (DABO) derivatives acting as NNRTIs of HIV-1. A dataset of 74 compounds was compiled from published studies and randomly subdivided into training and test sets. To understand the pharmacophoric effect, Kier-Hall Electrotopological State descriptors namely SN1, SN3, SF, SAr, SS, SO, SNO2, SCl, SY (Y = S-alkyl and NH-alkyl), SX (X = Me) and biological activity were used as independent and dependent variable respectively. Statistical results were highly encouraging for the training set multiple linear regression [(MLR): r2 = 0.961, F = 100.41 and q2 = 0.926, neural networks (NN): r2 = 0.966, F = 115.594, degrees of freedom = 40 and k-nearest neighbour (k-NN): r2 = 0.770, q2 = 0.757, degrees of freedom = 40]. Results of validation using a test set showed the same trend as training set (NN > MLR > kNN). The above results suggest that of various functional groups present in DABO such as SN3, SO, SCl, SAr and SNO2 contribute more significantly towards activity. On the other hand SN1, SS, and SF do not play any role in enhancing the activity. The substitution of S-alkyl and NH-alkyl at C2 position is essential though it does not contribute much towards the activity. The substitution of methyl group at C5 position is unfavorable and exhibit negative impact on inhibitory activity. Therefore, it seems reasonable to choose E-state indices as suitable and significant descriptors for exploring the relationship between the pIC50 and the pharmacological properties of the compounds.

Background:To evaluate efficacy and safety of combined pars plana vitrectomy(PPV)and scleral fixated intraocular lens(SFIOL)surgery as a single ***:Retrospective interventional case series done at a tertiary eye care center in Northern *** patients who underwent combined PPV and SFIOL surgery were included and analyzed ***:Mean age of the patients was 43.36±15.12 years(range,22-64 years).Eight were *** baseline best corrected visual acuity(BCVA)was 0.78±0.63 logMAR units while the mean post-operative BCVA at 6 months follow-up was 0.37±0.29 logMAR units,the visual gain being statistically significant(P=0.021).None of the patients had a drop in BCVA with nine patients having final BCVA better than 0.48 logMAR *** detachment(CD)was the only notable complication,seen in three *** complications included two cases of intraoperative retinal breaks,a case each of reversible corneal edema,ocular hypertension and cystoid macular ***:Combined PPV and SFIOL is an efficacious procedure for managing IOL/lens dislocation and aphakia in a single *** may be short-term reversible complications with no impact on final visual gain.
