Modulation of human B lymphocyte differentiation by therapeutic immunoglobulins: from protein to mRNA levels

Several groups are investigating the mechanisms of action of therapeutic immunoglobulins (IVIg) in order to improve their use. In vitro models such as CD40-CD154 interaction are necessary to study the physiological response of human B lymphocytes to IVIg. Human B lymphocytes treated with IVIg triggers a rapid phospho-rylation (pro-liferation. However, the modulation of human lymphocyte physiology by IVIg is a gradual and cumulative process and requires long-term experimentation. Differentiation of human B lymphocytes into Ig-secreting cells can be evaluated both at the transcription and translation levels. The secretion of immunoglobulins can be assessed using ELISA or ELISpOTS whereas expression of immunoglobulin genes can be measured using semi-quantitative or quantitative pCR methods. We hereby report a comparison of these methods to explain how contradictory observations towards IVIg effects could result from their use. Our results indicate that ELISA and ELISpOTS will provide consistent observations by opposition to real-time pCR quantification. Besides, the reliability of each of these me-thods remained dependent on the stimulation period as well as the preparation of cellular extracts or cell samples following IVIg-treatment.

Human metapneumovirus (hMpV), a newly discovered paramyxovirus, has been associated with acute respiratory tract infections (ARI) ranging from upper ARI to severe bronchiolitis and pneumonia. Important questions remain on the contribution of hMpV to ARI, and it’s impact on public health. During the 2001-2002 season, we conducted a collaborative study with four provincial public health laboratories to study the prevalence of this new virus in the Canadian population. A total of 445 specimens were collected from patients of all age groups suffering from ARI and tested for the presence of hMpV by RT-pER. Of these, 66(14.8%) tested positive for hMpV positive(?)

Background: Neurofibromatosis (NF) is a genetic disorder of the nervous system that affects the development and growth of neuronal tissues. It is characterized by the development of malignant and benign tumors of the peripheral nerves and meningiomas, and the clinical manifestations can vary, including cerebral vascular compromise that precipitates symptoms characteristic of Moyamoya syndrome. The behavior of a posttransplant child with type 1 NF who presents with characteristics of Moyamoya syndrome and is a carrier of a WT1 genetic mutation is unknown, which is why the report was made. Case Report: A child with type I NF with a history of cryptorchidism, hypospadias, bilateral sensorineural hearing loss, and chronic renal failure was arising from a congenital nephrotic syndrome due to a WT1 gene mutation at 3 years of age began renal replacement therapy initially with chronic ambulatory peritoneal dialysis and hemodialysis for refractory peritonitis. He underwent two episodes of transient ischemic attack (TIA) characterized by right hemiparesis during hemodialysis therapy and during six months post-kidney transplantation present a similar episode. Studies for other causes were negative, and imaging studies show characteristics of NF-related vasculitis, cortical lesions, and focal lesions in the left frontal subcortical white matter with annular enhancement and subcortical edema in the parasagittal region of the left parietal lobe, as well as a decrease in flow in the cortical branches of the middle cerebral artery were described. After four weeks, the clinical manifestations of hemiparesis completely disappeared without leaving any sequels continuing with the regular immunosuppression of the everolimus, mycophenolate and low dose steroids. Conclusion: These findings are suggestive of small and mediumsized vessel vasculitis compatible with Moyamoya syndrome in child post-transplantation associated with arteriopathy

＜正＞ In recent times, the development of privacy technologies has promoted the speed of research on privacy-preserving collaborative data mining. people borrowed the ideas of secure multi-party computation and developed secure multi-party protocols to deal with privacy-preserving collaborative data mining problems. Random perturbation was also identified to be an efficient estimation technique to solve the problems. Both secure multi-party protocol and random perturbation technique have their advantages and shortcomings. In this paper, we develop a new approach that combines existing techniques in such a way that the new approach gains the advantages from both of them.

AIM： To examine the relationships between pre-diag- nostic biomarkers and colorectal cancer risk and assess their relevance in predictive ***： A nested case-control study was designed to include all first primary incident colorectal cancer cases diagnosed between inclusion in the SUpplemen- tation en VItamines et Min^raux AntioXydants cohort in 1994 and the end of follow-up in 2007. Cases （n = 50） were matched with two randomly selected con- trols （n = 100）. Conditional logistic regression models were used to investigate the associations between pre- diagnostic levels of hs-CRp, adiponectin, leptin, soluble vascular cell adhesion molecule-1 （sVCAM-1）, soluble intercellular adhesion molecule-I, E-selectin, monocyte chemoattractant protein-1 and colorectal cancer risk. Area under the receiver operating curves （AUC） and relative integrated discrimination improvement （RIDI） statistics were used to assess the discriminatory poten- tial of the models. RESULTS： plasma adiponectin level was associated with decreased colorectal cancer risk （p for linear trend -- 0.03）. Quartiles of sVCAM-1 were associated with increased colorectal cancer risk （p for linear trend = 0.02）. No association was observed with any of the other biomarkers. Compared to standard models with known risk factors, those including both adiponectin and sVCAM-1 had substantially improved performance for colorectal cancer risk prediction （p for AUC improve- ment = 0.01, RIDI = 26.5%）. CONCLUSION： These results suggest that pre-diag- nostic plasma adiponectin and sVCAM-1 levels are as- sociated with decreased and increased colorectal cancer risk, respectively. These relationships must be confirmed in large validation studies.

Late steps of HIV-1 life cycle are determinant for optimal dissemination of the virus to new target cells. These steps include assembly of structural precursors, budding of the new particle and maturation into fully infectious virions. Each step is finely tuned and timely regulated to allow the appropriate assembly of structural components, the efficient recruitment of viral and cell partners and the timely regulated proteolytic processing of the protein precursors. Despite the huge number of studies devoted to the definition of molecular mechanisms regulating these steps, a number of question remains to be answered before they are clearly apprehended. The elucidation of the role played by each viral proteins, nucleic acids as well as host-encoded factors will provide new clues in the understanding of the retroviral assembly/maturation process and will allow further development of new antiviral compounds. This review reports the most recent progress as well as the questions that remain to be answered in the field of HIV-1 assembly, release and maturation. Finally, we also describe the data available on the design and use of new antiretroviral drugs targeting these specific steps of the retroviral replication.
