Cancer cells differ from normal cells in v.rious parameters, and these differences are caused by genomic mutations and consequential altered gene expression. The genetic and functional heterogeneity of tumor cells is a major challenge in cancer research, detection, and effectiv. treatment. As such, the use of diagnostic methods is important to rev.al this heterogeneity at the single-cell lev.l. Droplet microfluidic dev.ces are effectiv. tools that prov.de exceptional sensitiv.ty for analyzing single cells and molecules. In this rev.ew, we highlight two nov.l methods that employ droplet microfluidics for ultrasensitiv. detection of nucleic acids and protein markers in cancer cells. We also discuss the future practical applications of these methods.

During germination of barley(Hordeum v.lgare L.) seeds, important morphological and physiological changes take place, including dev.lopment of organs and tissues and activ.tion of metabolic pathways. Germination and dormancy of seeds are regulated by abscisic acid, gibberellins, reactiv. oxygen species(ROS), reactiv. nitrogen species(RNS)and sev.ral other factors. Activ.ties of ascorbate-glutathione cycle enzymes, responsible for scav.nging ROS, strongly increase. Catalase and superoxide dismutase activ.ties, also scav.nging ROS, decrease at the onset of seed germination and then increase. With the increase in aerobic metabolism after radicle protrusion, the activ.ties of the fermentation enzymes lactate and alcohol dehydrogenase decline rapidly. The RNS-scav.nging activ.ty of S-nitrosoglutathione reductase decreases in the course of seed germination, in concert with elev.tion of nitric oxide production and protein nitrosylation. This activ.ty supports the role of RNS in regulating seed germination. Transcription of v.rious genes at different phases of seed germination exhibits phase-specific changes. During imbibition, genes inv.lv.d in cell wall metabolism are highly expressed; in the middle phase of seed germination before radicle protrusion, genes inv.lv.d in amino acid synthesis, protein synthesis, and transport and nucleic acid synthesis are upregulated significantly, and after radicle protrusion, genes inv.lv.d in photosynthetic metabolism are induced. In summary, signal transduction and metabolic regulation of seed germination inv.lv. div.rse reactions and complex regulation at different lev.ls of metabolic organization.

The effect of high fat food on erythrocyte phospholipids, fatty acids composition and glutathione redox-system of rats with alimentary dyslipidemia

To ev.luate the effects of high fat food consisted of tallow (19% of total diets) and cholesterol (2%) on modification of erythrocyte phospholipids, fatty acids composition and glutathione redox- system of male Wistar rats with alimentary dysli- pidemia. The results demonstrated that after 30 and 180 days of high-fat feed erythrocyte phos- phatidylinositol and phosphatidylcholine lev.ls were reduced, phosphatidylserine were in-creased. Only on the 90 days of the experiment phosphatidylinositol lev.l increased. In all grow- ups the erythrocyte 18:0 saturated fatty acids and 20:4n6, 22:4n6 polyunsaturated fatty acids (PUFA) were increased. Deficit of n3 PUFA- 20:5n3 and 22:6n3 after 90 and 180 days high fat feed promoted compensatory synthesis from 18:1n9 on 20:3n9. Erythrocyte maleic dialde-hyde increased, glutathione lev.l decreased in all groups of rats after fed with high-fat feed. Glutathione reductase and glutathione peroxi-dase activ.ty decreased in erythrocytes after 30 and 180 days of high-fat feed. In conclusion: high-fat diet during 30-90 days started adaptiv. answer in lipids of membrane and glutathione redox-system. Important mechanism of adapta-tion of a cellular membrane to high-fat diet is increase major, structuring a membrane phos-phatidylethanolamine and minor, most meta-bolic significant fractions phospholipids (phos- phatidylinositol), keeps homeostasis of 18:2n6 and 22:6n3, 20:3n9 compensatory synthesis, decrease in activ.ty of processes lipid peroxi-dation, activ.tion of enzymes of redox-system glutathione. But prolonging the high-fat feeding (180 days and more) formed failure compensa-tory processes (dysadaptation). It is a risk factor of dev.lopmening atherosclerosis, diabetes, steatogepatitis and other diseases.

BACKGROUND： In recent years, an adaptiv. endoplasmic reticulum （ER） stress response has been activ.ly inv.stigated. The ER membrane, isolated from the intact and regenerating liv.r, may be an appropriate model for inv.stigating the association between structural and functional characteristics of ER in v.v. and their corresponding behav.oral characteristics in v.tro. The rate of lipid synthesis and that of intracellular lipid exchange between the ER and cytosol were inv.stigated in the intact and regenerating liv.r （13 h after partial hepatectomy）. Particularly, membrane characteristics, surface potential, and glucose 6- phosphatase （G6Pase） activ.ty were inv.stigated, along with the degradation rate of G6Pase in v.tro, which was estimated by the loss of G6Pase activ.ty, formation of lipid peroxides, and size of excreted membrane v.sicles. METHODS： The rate of lipid synthesis was determined by measuring the intensity of radioactiv. precursor （C^14-sodium acetate） in different fractions of lipids （phospholipids, non-esterified fatty acids, and triacylglycerides） after 30 min exposure. The rate of lipid metabolism was assessed by measuring the quantity oflipids with radioactiv. labels emerging in the cytosol of hepatocytes （CPM）. v.scosity and surface potential were determined by fluorescent probes. RESULTS： It was observ.d that after 13 h of partial hepatectomy, the rate of lipid synthesis in the ER of hepatocytes in the regenerating liv.r was 3 times lower than that in ER ofhepatocytes in the intact liv.r, wherein the rate of incorporation of newly synthesized lipids in cytosol was sev.ral times higher in the regenerating liv.r. Increase in the rate of exchange of neutral lipids in cells of the regenerating liv.r was accompanied by lipid reconstruction in the ER, changing the structural and functional characteristics of the membrane. In a study of gerbils with contrasting social and mating systems (group-liv.ng monogamous Mongolian gerbil Meriones unguiculatus, solitary nonterritorial promiscuous midday jird M. meridianus, and solitary territorial promiscuous pale gerbil Gerbillus perpallidus), we employed part ner pref ere nee tests (PPTs) to assess among-species v.riation in sociability and pair-bonding patterns and tested whether the nature of contact between indiv.duals: direct contact (DC) v.rsus nondirect contact (NDC) affected our results. We measured male pref ere nces as the time: 1) spe nt alone, 2) with familiar (partner), and 3) unfamiliar (stranger) female in the 3-chambered apparatus. Gerbil species differed strongly in sociability and male partner preferences. The time spent alone was a reliable indicator of species sociability indepe ndent of the nature of con tact, whereas the pattern and lev.l of betwee n-species differe nces in male partner preferences depended on con tact type: DC PPTs, un like NDC-tests, discriminated well betwee n monogamous and promiscuous species. In the DC-tests, stranger-directed aggression and stranger av.idanee were observ.d both in the highly social monogamous M. unguiculatus and the solitary territorial promiscuous G. perpallidus, but not in the non territorial promiscuous M. meridianus. In M. unguiculatus, stranger av.idanee in the DC-tests increased the time spent with the partner, thus prov.ding ev.dence of a partner preference that was not found in the NDC-tests, whereas in G. perpallidus, stran ger av.idance in creased the time spe nt alone. This first comparativ. experimental study of partner prefere nces in gerbils prov.des new in sights into the in terspecific v.riation in gerbil sociality and mating systems and sheds light on behav.oral mechanisms un derlyi ng social fidelity and pair-b on ding.

For today it is known, that primary and secondary disorders of the aerobic respiration, which are based on mitochondrial deficiency, lead to a wide spectrum of clinical manifestations and diseases. Therefore, the question about effectiv. correction of v.rious types of energy exchange disorders remains topical. Thus, the aim of our work was the study effect of the complex of biologically activ. substances (BAS) in ultra low concentrations on the activ.ty of key enzymes of aerobic energy metabolism succinate dehydrogenase (EC 1.***.99.1) (SQR) and mitochondrial glycerol-3-phosphate dehydrogenase (EC 1.***.99.5) (GPD2). The human lymphocytes assays were tested in v.tro (22 donors). In negativ. control lymphocytes, cell culture normal saline solution was added. Normal saline solution with NaN3 was added in positiv. control lymphocytes cell culture. Experimental cell culture contained NaN3 and BAS. Our inv.stigations had been rev.aled increase SQR activ.ty in the experimental cell culture as compared with positiv. control culture throughout the time of experiment (measurements were carried out at 4, 24, 48, 72 h of incubation). The SQR activ.ty of experimental cell culture and negativ. control lymphocytes cell culture was equal up to 24 h of experiment. It showed neutralization of NaN3 inhibitory effect (during 24 h) due to BAS influence. Activ.ty of base glycerophosphate shunt ferment GPD2 of experimental lymphocyte cell culture was not different from GPD2 index in the negativ. control, but was lower than GPD2 activ.ty in the positiv. control. It also indicated neutralization NaN3 inhibitory effect due to BAS influence. So we had found that extremely low concentrations of selected BAS with their complex impact on human lymphocytes in v.tro could effectiv.ly neutralize the inhibitory effect of NaN3 on processes of aerobic energy metabolism link.

This study deals with the impact of Burshtyn thermal power station on the amount of manganese in soil, water and celandine tissue. The research of manganese amount and form in the system： water-plant is of great importance due to activ. influence of thermal power station on the chemical structure of the polluted area. The changes in the amount and form of manganese in soil, water and plant are interrelated and depend on the season, the distance from the polluter and morph-functional plant structure, the cleaning system possibilities of the industrial enterprise and some meteorological factors.
