Induction of CXC chemokines in human mesenchymal stem cells by stimulation with secreted frizzled-related proteins through non-canonical Wnt signaling

AIM: To investigate the effect of secreted frizzledrelated proteins(s FRPs) on CXC chemokine expression in human mesenchymal stem cells(h MsCs).METHODs: CXC chemokines such as CXCL5 and CXCL8 are induced in h MsCs during differentiation with osteogenic differentiation medium(OGM) and may be involved in angiogenic stimulation during bone repair. h MsCs were treated with conditioned medium(CM) from L-cells expressing non-canonical Wnt5 a protein, or with control CM from wild type L-cells, or directly with s FRPs for up to 10 d in culture. m RNA expression levels of both CXCL5 and CXCL8 were quantitated by real-time reverse transcriptase-polymerase chain reaction and secreted protein levels of these proteins determined by ELIsA. Dose-(0-500 ng/m L) and time-response curves were generated for treatment with s FRP1. signal transduction pathways were explored by western blot analysis with pan- or phosphorylation-specific antibodies, through use of specific pathway inhibitors, and through use of si RNAs targeting specific frizzled receptors(Fzd)-2 and 5 or thereceptor tyrosine kinase-like orphan receptor-2(Ro R2) prior to treatment with s FRPs. REsULTs: CM from L-cells expressing Wnt5 a, a noncanonical Wnt, stimulated an increase in CXCL5 m RNA expression and protein secretion in comparison to control L-cell CM. s FRP1, which should inhibit both canonical and non-canonical Wnt signaling, surprisingly enhanced the expression of CXCL5 at 7 and 10 d. Dickkopf1, an inhibitor of canonical Wnt signaling prevented the s FRPstimulated induction of CXCL5 and actually inhibited basal levels of CXCL5 expression at 7 but not at 10 d post treatment. In addition, all four s FRPs isoforms induced CXCL8 expression in a dose- and time-dependent manner with maximum expression at 7 d with treatment at 150 ng/m L. Cellulose based polymers in development of amorphous solid dispersions

Cellulose derivatives have gained immense popularity as stabilizers for amorphous solid dispersion owing to their diverse physicochemical properties. More than 20 amorphous solid dispersion-based products that have been approved for marketing consist of cellulose derivatives as stabilizers, thus highlighting their importance in generation of amorphous solid dispersions. These polymers offer numerous advantages like drug solubilization, crystallization inhibition and improvement in release patterns of drugs. Exploring their potential and exploiting their chemistry and p H responsive behaviour have led to the synthesis of new derivatives that has broadened the scope of the use of cellulose derivatives in amorphous formulation development. The present review aims to provide an overview of different mechanisms by which these cellulose derivatives inhibit the crystallization of drugs in the solid state and from supersaturated solution. A summary of different categories of cellulose derivatives along with the newly explored polymers has been provided. A special segment on strengths, weaknesses, opportunities, and threats(sWOT) analysis and critical quality attributes(CQAs) which affect the performance of the cellulose based amorphous solid dispersion will aid the researchers in identifying the major challenges in the development of cellulose based solid dispersion and serve as a guide for further formulation development.

Antioxidative properties of phenolic compounds isolated from the fungal endophytes of Zingiber nimmonii （J. Graham） Dalzell.

BACKGROUND： The microbes living in planta termed ＇endophytes＇ is bestowed with the potential to produce bioactive substances. The aim of this investigation was focused on the isolation and molecular identification of the fungal endophytes from Zingiber nimmonii （J. Graham） Dalzell., an endemic medicinal plant species of the ＇Western ghats＇, a hotspot location in southern India and characterization of the secondary metabolites responsible for the ant^oxidant and DNA protective capacity using chromatography and mass spectrometry techniques. METHODs： Endophytic fungi were isolated and identified by sequencing the Internal Transcribed spacer （ITs）. The secondary metabolites were extracted with ethyl acetate and evaluated for the total phenolic, flavonoid and antioxidant capacities. The isolates with potential antioxidative property were further analyzed for the DNA protection ability and the presence ofbioactive phenolic compounds by High Performance Liquid Chromatography （HPLC） and Electrospray Ionization- Mass spectroscopy/Mass spectroscopy （EsI-Ms/Ms） techniques. REsULTs： Endophytic fungi belonging to 11 different taxa were identified. The total phenolic content of the extracts ranged from 10.8±0.7 to 81.6±6.0 mg gallic acid equivalent/g dry extract. F lavonoid was present in eight extracts in the range of 5.2 ±0.5 to 24.3±0.9 mg catechin equivalents/g dry extract. Bipolaris specifera, Alternaria tenuissima, Aspergillus terreus, Nectria haematococca and Fusarium chlamydosporum extracts exhibited a potentially high ant^oxidant capacity. Characterization of the extracts revealed an array of phenolic acids and flavonoids. N. haematococca and E chlamydosporum extracts contained quercetin and showed DNA protection ability. CONCLUsION： This study is the first comprehensive report on the fungal endophytes from Z. nimmonii, as potential sources of antioxidative and DNA protective compounds. The study indicates that Z. nimmonii endophytes are potential sources of antioxidants

seroprevalence of varicella zoster virus in Colombo district, sri Lanka

Objective: To determine the seroprevalence of varicella zoster virus(VZV) antibodies among the population residing in the Colombo district of sri Lanka. Methods: A cross-sectional population-based study was conducted which included 1 258 participants. Blood samples were collected and questionnaires administered to obtain sociodemographic information and history of varicella and/or herpes zoster. serum samples were assayed for VZV IgG antibodies using a commercial enzyme-linked immunosorbent assay kit. Results: Overall, the seroprevalence was 54.2%(95% CI= 51.5% 57.0%). Children below 1 year of age were seronegative, and only about 20.0% of children between 1 and 10 years of age were seropositive. seropositivitiy increased with age and by the age of 40 years 74.3% were seropositive. Among women of childbearing age, the overall seroprevalence was about 62.0%(95% CI = 57.7%-66.1%) but was low 37.0% in the 15-19 age group. Conclusion: In this population, 45.8% lacked natural immunity against varicella. Of women of childbearing age, 39.9% lacked immunity and in the subgroup of women 15-19 years of age, 63.0% women lacked immunity. In light of the country's success with the control and high coverage of other vaccine preventable diseases and that the vaccine is available in the private sector, the inclusion of varicella vaccine in the national immunization program may be considered.

目的:建立C57BL/6小鼠脉络膜新生血管(Choroidal neovascularization,CNV)动物模型的评估方法。方法:①应用氪红激光对小鼠实验性视网膜进行光凝,激光的功率、光凝斑直径和曝光时间分别为250 mW、50μm及0.05 s诱导CNV动物模型。②分别于光凝后1、3、5、7、10d,用2×106FITC-dextran荧光素灌注小鼠心脏。③mAb特定抗Elastin抗体标记Bruch膜。④视网膜色素上皮(Retinal pigmentepithelium,RPE)-脉络膜-巩膜复合体铺片。⑤激光共聚焦显微镜对RPE-脉络膜-巩膜复合体进行CNV形态进行观察和测量。结果:光凝后3 d可见新生血管及走向清晰可见,光凝后7 d CNV达到高峰。结论:用抗体标记Bruch膜。FITC-dextran荧光素灌注小鼠心脏,激光共聚焦显微镜对RPE-脉络膜-巩膜复合体进行氪红激光诱导C57BL/6小鼠的CNV模型进行评估,直观,可靠。

Effect of dietary incorporation of n-3 polyunsaturated fatty acids rich oil sources on fatty acid profile, keeping quality and sensory attributes of broiler chicken meat

The present study was undertaken to investigate the effect of dietary replacement of commonly used vegetable oil(sunflower oil, sFO) with n-3 polyunsaturated fatty acids(PUFA) rich oil sources on broiler chicken performance, carcass yield, meat fatty acid composition, keeping quality and sensory attributes of meat. In the current experiment, 300 day-old Krishibro broiler chicks were randomly distributed to 5 dietary groups(50 replicates with 6 chicks in each) prepared by replacing sFO(2% and 3% of diet during starter and finisher periods, respectively) with n-3 PUFA rich soybean oil(sO), mustard oil(MO), linseed oil(LO) or fish oil(FO) on weight basis. Variation in oil sources had no influence(P > 0.05) on performance and carcass yield. supplementation of MO, LO or FO significantly(P sed the n-3 PUFA, lowered the n-6 PUFA deposition and n-6:n-3 ratio in breast and thigh without affecting the organoleptic characters(appearance, flavour, juiciness, tenderness and overall acceptability) of ***, thiobarbituric acid reacting substances concentration in meat was increased(P supplementation compared with sFO. It is concluded that, dietary incorporation of MO, LO or FO at 2% and 3% levels during starter and finisher phase can enrich broiler chicken meat with n-3 PUFA without affecting the bird's performance and sensory characters of meat.
