Development and Validation of a Liquid Chromatographic Method for the Determination of Leflunomide： Application to in vitro Drug Metal Interactions

Leflunomide is a leading drug for the treatment of rheumatoid arthritis. The principle aim of this study was to develop and validate an RP-HPLC method for the determination of leflunomide in bulk and pharmaceutical dosage form using diclofenac sodium as an internal standard. For this purpose, chromatography was accomplished on a Purospher Start, C18 （5 μm, 12.5 cm×0.46 cm） column at ambient temperature. Methanol ： water （80 ： 20, V/V） solvent system was selected as mobile phase, the pH of which was adjusted to 3.4 by ortho-phosphoric acid and de- livered at a flow rate of 1.2 mLomin-1. Seperation of leflunomide and diclofenac sodium was carried out on a Purospher Start, C18 equipped with a UV-visible detector at 248 nm. The suitability of the method for the quantita- tive determination of the drugs is proven by validation in accordance with the requirements laid down by the Inter- national Conference on Harmonization （ICH） guidelines. The method was accurate （99.55%--100.03%）, specific, linear （R2〉0.999） and precise （intra-day precision 0.023%--0.93% and inter-day precision 0.26%--0.944%） in the range of 0.5--20 μg·mL^-1. The minimum limit of detection and quantification in pharmaceutical formulation were 0.05 and 0.15 μ***^-1, respectively. Thus the proposed method is simple, accurate, reproducible and suitable for the routine analysis of leflunomide in pharmaceutical formulations and was applied to study in vitro drug-metal interactions.

Neuroprotective role of 17<i>β</i>estradiol with tachykinin neuropeptide NKB and A<i>β</i>(25 - 35) in aging female rat brain

The brain experiences structural, molecular and functional alterations during aging. In aging brain tissue, the oxidative stress increases due to decreased activity of antioxidant enzymes and increased oxidative stress leading to neurodegeneration associated with excitotoxicity. In the present study, we observed the effect of tachykinin neuropeptide Neurokinin B (NKB) and amyloid beta fragment Aβ (25 -?35) on the activity of Acetylcholine esterase (AChE) and Lipid peroxidation (LPO) in brains of 17β estradiol (E2) treated aging female rat synaptosomes of different age groups. An in-vitro incubation of E2 treated brain synaptosomes with Aβ (25 -?35) showed toxic effects on all the parameters. The treatment of NKB and combined NKB and Aβ (25 -?35) increased the AChE enzyme activity and decreased the level of LPO in E2 treated aging rats. The treatment of NKB and combined NKB and Aβ (25 - 35) in a concentration dependent manner reversed the effects of aging and Aβ (25 -?35) on AChE and LPO. The present finding suggests that E2 along with NKB reverse aging and Aβ (25 -?35) induced toxicity as well as AChE and LPO levels. The results of the current study showed a possible beneficial role of NKB with E2 inthe age related neurological diseases.

Design, synthesis, and nanoengineered modification of spherical graphene surface by layered double hydroxide (LDH) for removal of As (Ⅲ) from aqueous solutions

In this study, new nano spherical graphene modified with LDH(Layered Double Hydroxide) was prepared and used to remove As(Ⅲ) ion from aqueous solutions. At first, graphene oxide was synthesized from graphite using a well-known Hammer method. The obtained graphene oxide solution was sprayed in octanol solution under different temperatures and sprayed speed as influenced variables. The structure and physical characterization of synthesized spherical graphene oxide were determined by various techniques,including FT-IR, N_(2) adsorption–desorption, SEM, TEM, and EDX. In the next step, the hydrothermal method was applied to deposition LDH on the spherical graphene oxide. The synthesized spherical graphene modified by LDH was used to remove As(Ⅲ) as a toxic heavy metal ion. The effect of influenced variables including p H, contact time, amount of sorbent, and type eluent studied and the optimum values were as 8, 30, 50, and HCl(0.5 mol·L^(-1)), respectively. After optimization, the studied sorbent was shown a high adsorption capacity(149.3 mg·g^(-1)). The adsorption mechanism and kinetic models exhibited good agreement with the Langmuir isotherm and pseudo-second-order trends, respectively. Besides, the synthesized product was tested for seven times without significant loss in its sorption efficiency.

Recently,the inductive coupling link is the most robust method for powering implanted biomedical devices,such as micro-system stimulators,cochlear implants,and retinal *** research provides a novel theoretical and mathematical analysis to optimize the inductive coupling link efficiency driven by efficient proposed class-E power amplifiers using high and optimum input *** design of the coupling link is based on two pairs of aligned,single-layer,planar spiral circular coils with a proposed geometric dimension,operating at a resonant frequency of 13.56 *** transmitter and receiver coils are small in *** device resistance varies from 200Ωto 500Ωwith 50Ωof *** the conventional load resistance of power amplifiers is 50Ω,the efficiency is 45%;when the optimum resonant load is 41.89Ωwith a coupling coefficient of 0.087,the efficiency increases to 49%.The efficiency optimization is reached by calculating the matching network for the external LC tank of the transmitter *** proposed design may be suitable for active implantable devices.

Objective:This study aimed to determine the risk factors associated with oral polio vaccine(OPV)***:A case-control study was conducted in Rahim Yar Khan(RYK)District,Pakistan.A case was defined as‘‘any child aged1 km from a healthcare facility was significantly associated(OR,2.5;95%CI,1.4–4.4;p<0.05)with OPV refusal,while prior visits by healthcare workers(OR,0.11;95%CI,0.06–0.22;p<0.05)and mother’education above primary level(OR,0.42;95%CI,0.24–0.73;p<0.05)were inversely associated with OPV ***:On the basis of our recommendations,door-to-door social mobilization visits by healthcare workers to alleviate fear,misconception,and fatigue of repeated campaigns and strict accountability mechanism were developed by district government at pre-and intra-campaign levels.

This paper presents a comprehensive study that includes the sizing and power flow by series and parallel inverters in a distributed generation system(DGs)that integrates the system of hybrid wind photovoltaic with a unified power quality conditioner(UPQC).In addition to supplying active power to the utility grid,the system of hybrid wind photovoltaic functions as a UPQC,compensating reactive power and suppressing the harmonic load ***,the load is supplied with harmonic-free,balanced and regulated output *** PVWind-UPQC is established on a dual compensation scheme,the series inverter works like a sinusoidal current source,while the parallel inverter works like a sinusoidal voltage ***,a smooth alteration from interconnected operating modes to island operating modes and vice versa can be achieved without load voltage *** PV-Wind-UPQC inverters handle the energy generated through the hybrid wind photovoltaic system and the energy demanded through the load,the converters should be sized cautiously.A detailed study of the flow of power via the PV-Wind-UPQC is imperative to gain a complete understanding of the system operation and the proper design of the ***,curves that allow the sizing of the power converters according to the power flow via the converters are presented and *** results are presented to assess both steady state and dynamic performances of the grid connected hybrid system of *** investigation is verified by simulating and analyzing the results with Matlab/Simulink.

Synthesis and antibacterial studies of ciprofloxacin-metal complexes

Metal complexes of ciprofloxacin were synthesized to study the effect of the drug on the metals present in the body or metals co-administered as multivitamin therapy. These complexes were then characterized by IR, UV, 1 H NMR, CHN and AA analyses. It was found that the metals coordinated through the β-keto carboxylic group, forming four coordinated complexes having square planar geometry. The effect of the antibacterial activity of these complexes with respect to parent drug was also studied against 14 different Gram + ve and Gram – ve organisms by the disk susceptibility technique. Some complexes showed improve activity as compared with the stated drug.

Polyamines such as putrescine, spermidine, spermine and agmatine are aliphatic polycationic compounds present in all living cells, and are derived from amino acids, intestinal bacteria, exfoliated enterocytes and supported from diet. Polyamines as the key compounds play essential role in cell proliferation, growth and differentiation. They also exert significant effects on embryonic development,implantation, embryonic diapause, placentation, angiogensis and fetal development. This review paper summarizes the functions of polyamines and embryo/fetus development and its regulatory mechanism which should help to provide some evidences for clinic.

A novel {[bis（pyridine-κN）bis（3,5-dinitrosalicylato κ-O,O＇）Zn（Ⅱ）][bis（pyridine-κN）Zn（Ⅱ）]}（C_（34）H_（24）N_8O_（14）Zn_2） was synthesized by a self-assemble method at room temperature. The molecular structure was determined by single-crystal X-ray analysis. The compound crystallizes in the monoclinic system, space group P2_1/n with a = 12.2156, b = 13.5696, c = 22.5602 A, β = 90.061o, Z = 4 and V = 3739.6（3）A^3. The new 1D binuclear coordination polymer {[bis（py-κN）bis（3,5-dinitrosal κ-O,O′） Zn（Ⅱ）][bis（py-κN）Zn（Ⅱ）]} resulted from two different types of moieties. The polymer [bis（py-κN）bis（3,5-dinitrosal κ-O,O′）Zn（Ⅱ）] unit is connected with [bis（py-κN） Zn（Ⅱ）] by zigzag topology. One zinc（Ⅱ） cation has a six-fold coordination environment, in which the metal atom is connected with four oxygen atoms of two 3,5-dinitrosalicylic acids to form equatorial bonds and two nitrogen atoms of pyridine to generate the axial bonds. Other four-fold nucleus contain two Zn-O bonds from different 3,5-dinitrosalicylic acids and two bonds with the Natom of pyridine. Antimicrobial assay results indicated that the compound showed moderate activities against different bacterial and fungal strains.
