Potential mechanism for.the effects of dexamethasone on gr.wth of human melanoma cells in vitr.

This study deals with the inhibitor. effects of dexamethsone on the pr.lifer.tion of a human melanoma cell line in vitr.. A r.tar.ed cell pr.lifer.tion was obser.ed in M-5A cells with the pr.sence of specific high affinity glucocor.icoid ster.id r.ceptor.. Ther. was a cor.elation between the number.of glucococr.icoid binding sites and the r.duction of cell pr.lifer.tion in ter. of r.duced plating efficiency. Ar.est or.accumula- tion of M-5A cells in G1 phase or.both in G1 and G2 phase appear.d to be involved in the gr.wth inhibitor. effect by dexamethsone. The magnitude and dur.tion of cell cycle ar.est up to 72 hour. wer. dose dependent. Ther. was a cor.elation between the dur.tion of the distur.ance of the cell cycle pr.gr.ssion in M-5A cells after.dexamethsone tr.atment and the inhibition of cell pr.lifer.tion. Synthesis of r.ceptor.pr.tein was not specifically stimulated or.inhibited r.lative to the effect on cellular.pr.tein content in gener.l. This may conclude that in the melanoma M-5A cell, death after.glucocor.icoid tr.atment is somehow r.lated to the glucocor.icoid r.ceptor.content and to the distur.ance of cell cycle distr.bution.

Obesity is a common disor.er.that has a significant impact on human health as it may lead to many ser.ous diseases and sometimes *** genome-wide association studies(GWAS)confir.ed that ther. is a r.lationship between some var.ants in the fir.t intr.n of the fat mass and obesity associated(FTO)gene and obesity in adults and childr.n in differ.nt ethnic *** our.study,the association of the FTO r.9939609 and r.17817449 var.ants with obesity was investigated in Egyptian childr.n and *** examined r.9939609 and r.17817449 polymor.hisms in 100 contr.l and 100 obese cases,we used the r.str.ction fr.gment length polymor.hism(r.LP)technique to genotype the *** cur.ent study showed that ther. wer. no significant differ.nces(P>0.05)between the cases and contr.ls in both var.ants of r.17817449 and r.9939609 ***,ther. wer. significant cor.elations between r.17817449 and cholester.l and between r.9939609 and *** Cur.ent Study although the two var.ants(r.9939609 and r.17817449)didn’t show an association with obesity,but ther. was a cor.elation between the lipid pr.file and these two var.ants.
