Activating Silent Glycolysis Bypasses in Escherichia coli

all living organisms share similar reactions within their central metabolism to provide precursors for all essential building blocksand reducing power. To identify whether alternative metabolic routes of glycolysis can operate in E. coli, we complementarilyemployed in silico design, rational engineering, and adaptive laboratory evolution. First, we used a genome-scale model andidentified two potential pathways within the metabolic network of this organism replacing canonical Embden-Meyerhof-Parnas(EMP) glycolysis to convert phosphosugars into organic acids. One of these glycolytic routes proceeds via methylglyoxal andthe other via serine biosynthesis and degradation. Then, we implemented both pathways in E. coli strains harboring defectiveEMP glycolysis. Surprisingly, the pathway via methylglyoxal seemed to immediately operate in a triosephosphate isomerasedeletion strain cultivated on glycerol. By contrast, in a phosphoglycerate kinase deletion strain, the overexpression ofmethylglyoxal synthase was necessary to restore growth of the strain. Furthermore, we engineered the “serine shunt” whichconverts 3-phosphoglycerate via serine biosynthesis and degradation to pyruvate, bypassing an enolase deletion. Finally, toexplore which of these alternatives would emerge by natural selection, we performed an adaptive laboratory evolution studyusing an enolase deletion strain. Our experiments suggest that the evolved mutants use the serine shunt. Our study reveals theflexible repurposing of metabolic pathways to create new metabolite links and rewire central metabolism.

Relationship of Pre-Existing Maternal/Caregiver Acute Respiratory Infection in the Pattern and Risk of Acute Respiratory Infection among Infants in Rivers State, nigeria

History of upper respiratory tract infection in the mother or siblings was associated with higher risk of acute lower respiratory tract infection in cases. Most upper respiratory tract infections were caused by viral pathogens and likely to occur in many members of the family. The study aimed to determine the existence and pattern of relationship between risk of acute respiratory infection (ari) among infants and exposure to pre-existing maternal/caregiver acute respiratory tract infection. The study was designed as a community-based nested case-control study of 1100 infants randomly selected from 12 communities out of 6 Local Government Areas of the 3 senatorial districts of Rivers State. A multistage random sampling technique was used in selecting the subjects up to the community level. Descriptive method was used to represent the characteristics of the subjects and the differences in ari between exposed and unexposed infants were tested in a bivariate logistics regression at 5% level of significance. Odds ratio (OR) was used to interpret the size effect measures of ari on exposure to pre-existing maternal/caregiver ari differences. A total of 275 Cases of ari and 825 controls were included in the study. Among exposed infants (n = 104), ari cases were found to be higher n = 80 (76.9%) than in control n = 24 (23.1%). Whereas, among unexposed infants n = 991, ari cases were found to be lower n = 195 (19.7%) than in control n = 796 (80.3%). For the exposed infants, the odds for ari were 13.5 times significantly higher compared to those of their unexposed counterparts (OR-Unadjusted = 13.52, (p ndings will widen the horizon in the etiological consideration of ari among infants vis-à-vis exposure potential to pre-existing maternal/caregiver ari via nursing care. Therefore, community-based sensitization programme on barrier nursing care techniques and personal hygiene practices should be on focus.

目的探讨间歇性导尿结合蒙医温针用于脊髓损伤后神经源性膀胱患者的临床效果。方法将60例脊髓损伤后神经源性膀胱患者随机分为间歇导尿组和联合组各30例。间歇导尿组采用间歇性导尿并辅以排尿训练,联合组在此基础上结合蒙医温针治疗。结果干预4周及8周后,联合组自主排尿量显著多于间歇导尿组,膀胱残余尿量显著少于间歇导尿组,膀胱功能评定分级及恢复自主排尿情况显著优于间歇导尿组(P<0.05,P<0.01)。结论间歇性导尿联合蒙医温针治疗可有效改善脊髓损伤后神经源性膀胱患者的膀胱功能。

A total of 1 312 kindergarten children in Beijing city were surveyed for acute respiratory infections (ari) during a one-year period. The average number of ari episodes was 3.1/child-year and the pneumonia incidence in children under 5 years of age was 59.2/1 000 ***. The viral infection distribution was analysed. Influenza viruses played an important role in ari epidemics in these children while respiratory syncytial virus was the major agent in acute lower respiratory infections.

Background: The term urinary lithiasis (UL) from the Greek “lithos” refers to the disease characterised by the result of abnormal precipitation of normal constituents of the urine within the urinary tract. The aim of this work was to determine the frequency of obstructive renal failure (ORF) of lithiasis origin and to describe the therapeutic indications. Methods: This was a retrospective prospective study in patients hospitalised in the nephrology department of Point G University Hospital for ORF of lithiasis origin over a 26-month period from 1 January 2018 to 1 February 2020 inclusive. Results: Among 1898 hospitalized patients, 32 met the inclusion criteria, i.e. a frequency of 1.7%. The male sex was 68.75% with a sex ratio of 2.2. The mean age was 48.38 ± 13.423 years with extremes of 20 and 65 years. Dysuria and urinary bilharzia were the main uro-nephrological antecedents, accounting for 25% of the cases each. Pain syndrome was the main functional sign, accounting for 100%. Ultrasound of the urinary tract (n = 28) showed stones in 92.85%. These stones were bilateral in 22.22% of cases. The dilatation was pyelocalic in 14 cases (51.8%). The uroscanner showed a pyelic location of the stones in both kidneys;42.1% on the right and 33.3% on the left. Hydronephrosis was the most common dilatation: 37.8% on the right and 29.7% on the left. Renal lithiasis was complicated by acute kidney injury (ari) in 17 cases (53.1%) versus 15 cases (46.9%) of chronic kidney disease (CKD). Urethral catheter was the means of drainage in 24 (75%) followed by nephrostomy in 8 cases (25%). nephrolithotomy accounted for 9.4% of cases. The case fatality rate was 28.12% (9 cases). Deaths occurred in the context of uraemic coma 6 cases and cardiorespiratory arrest 3 cases. Africa’s fertilizer use averages only 8 kg per hectare per year. Available fertilizer recommenda-tions in Africa are high, and vary between countries. The recommendations are generally out dated, and/or “blanket” recommendations that are not site, or crop specific. Recommendation developed for one crop is often used for another and possibly unrelated crop. CABI and AGRA designed a project whose goal is “to help improve the capacity of national Research Institutions in developing fertilizer recommendations for efficient and profitable fertilizer use in 13 sub-Saharan African countries within the framework of ISFM practices under smallholder farming by the end of March 2016”. Baseline survey conducted in May 2014 targeted scientists, policy makers, extension workers, agro-dealers, fertilizer companies and farmers/farmer organization. Snowballing and convenience sampling techniques were used to get the sample and emails were used to reach the respondents. A total of 416 respondents were targeted but only 219 responded from 12 countries. Out of the 219, 148 were extension workers and researchers. About half (51%) of the scientists are aware of fertilizer optimization tool, and a lesser proportion have used the tool across all countries (31%). Fewer extension workers (31%) were aware of the fertilizer optimization tool and none reported to have used the tool. On average about 5% of the study participants reported that other stakeholders were involved in the development of fertilizer recommendations including farmers, farmer organizations, policy makers, researchers, extension workers, agro dealers, fertilizer companies and donor organizations. Stakeholder involvement was highest in nigeria at 15%, followed by Ethiopia (13%) and Malawi (10%) but most of the countries reported neral understanding that development of fertilizer recommendations is a multi-stakeholder initiative and therefore need for involving many stakeholders in the development of fertili
