Investigation of HER-2 codon 655 single nucleotide polymorphism frequency and c-ErbB-2 protein expression alterations in gastric cancer patients

AIM： To investigate both whether the risk of gastric cancer is associated with the Ile/Val single nucleotide polymorphism （SnP） of human epidermal growth factor receptor-2 （HER-2） transmembrane domain-coding region at codon 655 and the suggested existence of HER-2 expression in gastric cancer cases in a Turkish patient group. METHODS： Polymerase chain reaction （PCR） followed by restriction fragment length polymorphism （RFLP） strategy was used to analyze the presence of HER-2 SnP at codon 655. c-erbB-2 expression pattern was analyzed by immunohistochemistry. The results were compared between gastric carcinoma group and chronic gastritis group, as well as between clinicopathological parameters and carcinoma. RESULTS： Results showed that Ile/Val genotype accounted for 20% within the Turkish gastric carcinoma group, and none in chronic gastritis group, and this genotyping was associated with stage Ⅳ gastric cancers （P = 0.04）. Positive membranous HER-2 immunoreactivity, on the other hand, accounted for 24% within the Turkish gastric carcinoma group and none from chronic gastritis cases; further, it was correlated with intestinal type carcinomas （P = 0.007）, and stage Ⅲ-Ⅳ carcinomas （P = 0.004）. COnCLUSIOn： These observations imply that the tested HER-2 SnP may participate in the development and progression of gastric cancer. Thus, after confirming these results with large sample groups, HER-2 codon 655 SnP and/or c-erbB-2 overexpression may also be used as a poor prognostic indicator for gastric carcinomas.

Apoptotic cell death and its relationship to gastric carcinogenesis

AIM: To investigate the apoptotic process of cells with in the intestinal metaplasia areas co-localizing with chronic gastritis and gastric carcinomas and to analyze the involvement of proteins regulating apoptosis in the process of intestinal metaplasia related gastric carcinogenesis. METHODS: Forty-two gastric carcinoma and seventeen chronic gastritis cases were included in this study. All cases were examined for the existence of intestinal metaplasia. Ten cases randomly selected from each group were processed for TUnEL assay. TUnEL positive cells within the intestinal metaplasia areas, co-localizing either to gastric carcinoma or chronic gastritis, were counted and converted to apoptotic indices. In addition, p53, bcl-2 and bax expression patterns within these tissues were analyzed on the basis of immunohistochemistry. RESULTS: Twenty-eight of the cases were intestinal and 14 of the cases were diffuse type adenocarcinomas. 64% (27/42) of the gastric carcinoma cases had intestinal metaplasia. Intestinal metaplasia co-localized more with intestinal type carcinomas compared with diffuse type carcinomas [75% (21/28) vs 42% (6/14), respectively; P ≤0.05]. The mean apoptotic index in tumor cells was 0.70±0.08. The mean apoptotic index in intestinal metaplasias co-localizing to tumors was significantly higher than that of intestinal metaplasias co-localizing to chronic gastritis (0.70±0.03 vs 0.09±0.01, respectively; P≤0.05). p53 positivity was not observed in areas of intestinal metaplasia adjacent to tumors or chronic gastritis. Intestinal metaplasia areas adjacent to tumors showed lower cytoplasmic bcl-2 positivity compared to intestinal metaplasia areas adjacent to chronic gastritis [55.5% (15/27) vs 70.5% (12/17), respectively]. Equatorial Plasma Bubbles(EPBs)are ionospheric irregularities that take place near the magnetic *** most commonly occur after sunset during the equinox months,although they can also be observed during other *** phenomenon significantly disrupts radio wave signals essential to communication and navigation *** national network of Global navigation Satellite System(GnSS)receivers in Indonesia(>30°longitudinal range)provides an opportunity for detailed EPB *** explore this,we conducted preliminary 3D tomography of total electron content(TEC)data captured by GnSS receivers following a geomagnetic storm on December 3,2023,when at least four EPB clusters occurred in the Southeast Asian *** and extracted TEC depletion with a 120-minute running average were then used as inputs for a 3D tomography *** 2D spatial distribution consistently captured the four EPB clusters over *** tomography results were validated through a classical checkerboard test and comparisons with other ionospheric data sources,such as the Global Ionospheric Map(GIM)and International Reference Ionosphere(IRI)*** of the results demonstrates the capability of the Indonesian GnSS network to measure peak ionospheric *** findings highlight the potential for future three-dimensional research of plasma bubbles in low-latitude regions using existing GnSS networks,with extensive longitudinal coverage.

Statistical and Economic Issues Concerning the Rural Poor; Retrospect and Prospects

Decent living based on regular and secured earning is one of the fundamentals of prosperous life. For well-being/upliftment of rural poor some of the basic components required are basic education, productive and decent employment and health care. The amenities of common people＇s life do rotate around these. Due to geophysical and cultural diversities prevailing in our country, the nature and dynamics of development in different States has notable variations. Our rural areas have also progressed and changed but they still need a lot to be provided. To ensure a balanced development in our country we need to lay emphasis on upliftment of the rural and marginalized poor. To accomplish this task government is working in this direction. However, it is still felt that concerted efforts are required on the basis of micro level background information. It could be done by addressing some of statistical and economic issues. The notable among them are percentage of surpluses mobilized from labour saving technological change utilized for essential services such as health, sanitation and education to entertainment and cultural activities; variation in acreage from traditional food crop to cash crop; changing trend, of the prices of agricultural inputs vis-a-vis the cost of agricultural output and use of ground water in cultivation; trend of informal credits in relation to rising cost of cultivation; analysis of the dynamics of welfare Schemes regarding employment, healthcare and basic education etc. Inclusive growth was the cardinal principle of 11 th Five Year Plan of our country. It was further strengthened in 12th Five Year Plan by prefixing faster and sustainable features to it. Therefore, in order to ensure that this basic objective is being fulfilled; it is necessary that along with policy making there should be in-built components of data collection, monitoring and evaluation.

There are few evidences on the effect of biochar on vegetable yield,nitrogen(n)and phosphorus(P)leaching losses under intensive vegetable production *** current field plot scale study evaluated responses of Chinese cabbage(Brassica oleracea L.)yield,n and P leaching losses using five n treatments of common n application rate according to local farmers’practice(n100%),reducing 20%or 40%n fertilizer(n80%and n60%),and reducing 40%n fertilizer but incorporating 10 or 20 t/ha biochar(n60%+BC10 and n60%+BC20).Results showed that n80%and n60%decreased both the cabbage economic and leaf yields by 6.8%-36.3%and 27.4%-37.7%,*** of biochar with reduced n fertilizer rates improved the cabbage yield,in particular the n60%+BC20 matched the yield that observed in n100%*** n and P uptake capacities of cabbage shoot probably contributed the higher vegetable production under both biochar amendment *** application mitigated the nH_(4)^(+)-n and total P leaching losses by 20%-30%and 29%-32%,respectively,compared with their counterpart treatment n60%.nevertheless,biochar exerted no influence on the nO_(3)^(-)-n *** addition,soil organic matter content was recorded with 7.4%-28.7%higher following 10-20 t/ha biochar *** conclusion,biochar application can increase economic yield of cabbage via increasing n and P use efficiency,decrease n and P leaching losses,and improve soil quality in an intensive vegetable production system.
