Learning,memory and hippocampal LTP in genetically obese rodents

We have found that leptin, at physiological concentrations of 10-12 mol/L, facilitates learning and memory and LTP maintenance in Wistar rats. To explore the role of leptin recepors in learning, memory and synaptic plasticity, experiments were carried out using Zucker rats (Z), db/db mice (db), and ob/ob mice(ob). The former two have defects in leptin receptors and the latter cannot produce normal leptin. Unlike the effects observed in normal rats, high or low frequency stimulation of Schaffer collateral-CA1 synapses in hippocampal slices prepared from Z, db and ob animals failed to induce the learning and memory relevant long-term potentiation or depression in CA1 neurons. However, LTP in ob CA1 synapses was facilitated by leptin at 10-12 mol/L concentration. Moreover, the paired-pulse facilitation of CA1 synaptic potentials and intracellularly recorded postsynaptic responses to the neurotransmitters AMPA, nMDA and GABA, applied electrophoretically to the apical dendrites of CA1 neurons, were approximately the same compared to the control lean animals. In addition, unlike the second messenger responses observed in Wistar rats, calmodulin kinase Ⅱ activity in the CA1 area of Z and db animals was not activated after tetanic stimulation of the Schaffer collaterals. It has been shown that all three strains, Z, db and ob display impaired spatial learning and memory when tested in the Morris water maze. The results of these experiments indicate a close relationship between spatial learning and memory, facilitation of LTP, and calmodulin kinase Ⅱ　activity.

Cell surface protein engineering for high-performance whole-cell catalysts

Cell surface protein engineering facilitated by accumulation of information on genome and protein structure involves heterologous production and modifica- tion of cell surface proteins using genetic engineering, and is important for the development of high-performance whole-cell catalysts. In this field, cell surface display is a major technology by exposing target proteins, such as enzymes, on the cell surface using a cartier protein. The target proteins are fused to the carrier proteins that transport and tether them to the cell surface, as well as to a secretion signal. This paper reviews cell surface display systems for prokaryotic and eukaryotic cells from the perspective of carrier proteins, which determine the number of displayed molecules, and the localization, size, and direction （n- or C-terminal an.horing） of the passengers. We also discuss advanced methods for displaying multiple enzymes and a new method for the immobilization of whole-cell catalysts using adhesive surface proteins.

nitrifiers activity and community characteristics under stress conditions in partial nitrification systems treating ammonium-rich wastewater

Long-term exposure of nitrifiers to high concentrations of free ammonia （FA） and free nitrous add （FnA） may affect nitrifiers activity and nitrous oxide （n2O） emission. Two sequencing batch reactors （SBRs） were operated at influent ammonium nitrogen （nH4-n） concentrations of 800 mg/L （SBRH） and 33S mg/L （SBRL）, respectively. The nH4-n removal rates in SBRH and SBRL were around 2.4 and 1.0 g/L/day with the nitritation efficiencies of 99.3% and 95.7%, respectively. In the simulated SBR cycle, the n20 emission factors were 1.61% in SBRH and 2.30% in SBRL. n2O emission was affected slightly by FA with the emission factor of 0.22%-0.65%, while n2O emission increased with increasing FnA concentrations with the emission factor of 0.22%～3.96%. The dominant ammonia oxidizing bacteria （AOB） were nitrosomonas spp. in both reactors, and their relative proportions were 38.89% in SBRH and 13.36% in SBRL. Within the AOB genus, a species （i.e., operational taxonomic unit [OTU] 76） that was phylogenetically identical to nitrosomonas europaea accounted for 99.07% and 82.04% in SBRH and SBRL, respectively. Additionally, OTU 215, which was related to nitrosomonas stercoris, accounted for 16.77% of the AOB in SBRL.

Incorporating nitrogen(n)atom in graphene is considered a key technique for tuning its electrical ***,this is still a great challenge,and it is unclear how to build n-graphene with desired nitrogen *** is a lack of experimental evidence to explain the influence and mechanism of structural defects for nitrogen incorporation into graphene compared to the derived DFT ***,this gap is bridged through a systematic study of different nitrogen-containing gaseous plasma post-treatments on graphene nanowalls(CnWs)to produce n-CnWs with incorporated and substituted *** structural and morphological analyses describe a remarkable difference in the plasma–surface interaction,nitrogen concentration and nitrogen incorporation mechanism in CnWs by using different nitrogen-containing *** conductivity measurements revealed that the conductivity of the n-graphene is strongly influenced by the position and concentration of C–n bonding *** findings open up a new pathway for the synthesis of n-graphene using plasma post-treatment to control the concentration and configuration of incorporated nitrogen for application-specific properties.

Spontaneous necrosis of solid gallbladder adenocarcinoma accompanied with pancreaticobiliary maljunction

A 71-year-old Japanese man with acute cholecystitis and an incarcerated gallbladder （GB） stone was admitted. Plain ultrasonography （US） incidentally detected a mass-like lesion in the fundus. Doppler US revealed that this elevated lesion had no blood flow. Computed tomography showed a relatively low-density mass, measuring 5 cm x 4 cm in diameter, with no positive enhancement. Magnetic resonance imaging showed a mass in the fundus with a slightly low intensity on Tl-weighted images and a slightly high intensity on T2-weighted images. We were agonized in making the qualitative diagnosis of mass-like lesions of the fundus, such as a benign tumor, cancer, or debris. We performed laparoscopic cholecystectomy, because the incarcerated GB stone clearly caused acute cholecystitis. Intra-operative cholangiography clearly revealed pancreaticobiliary maljunction. Amylase levels in the common bile duct and gallbladder were quite high. The elevated lesion in the fundus dearly showed severe necrosis. Although this necrotic nodule included non-viable adenocarcinoma cells, viable cancer cell nests were located in the muscularis propria and subcutaneous layer. Histopathological examination confirmed a solid adenocarcinoma. Thus, we diagnosed it as a gallbladder cancer, based on histopathological analysis of the resected specimen. We therefore undertook radical surgery, including wedge resection of the liver, radical dissection of regional lymph nodes, and resection of the extrahepatic bile duct. Histopathological findings revealed no cancer, hyperplasia or dysplasia in the additionally resected specimens. The patient was finally staged as T2, n0, H0, P0, M（-）, stage Ⅱ. We present the first case of spontaneous necrosis of solid gallbladder adenocarcinoma, with a review of previous studies.

Various soft actuators have been investigated to overcome the drawbacks of conventional solid machines and explore the applications of soft *** particular,and because they are expected to be applicable in minimally invasive medicine because of their safety,soft inflatable microactuators using an actuation conversion mechanism from balloon inflation to bending motion have been proposed for high-output bending *** microactuators could be applied to create an operation space by safely moving organs and tissues;however,the conversion efficiency could be further *** study aimed to improve conversion efficiency by investigating the design of the conversion *** contact conditions between the inflated balloon and conversion film were examined to improve the contact area for force transmission,with the contact area dependent on the length of the contact arc between the balloon and force conversion mechanism and on the amount of balloon *** addition,surface contact friction between the balloon and film,which affects actuator performance,was also *** generated force of the improved device is 1.21 n at 80 kPa when it bends 10 mm,which is 2.2 times the generated force of the previous *** improved soft inflatable microactuator is expected to assist in performing operations in a limited space,such as in endoscopic or laparoscopic operations.

Management of gastric mucosa-associated lymphoid tissue lymphoma in patients with extra copies of the MALT1 gene

AIM To identify the clinical features of gastric mucosaassociated lymphoid tissue(MALT) lymphoma with extra copies of *** This is a multi-centered,retrospective study. We reviewed 146 patients with MALT lymphoma in the stomach who underwent fluorescence in situ hybridization analysis for t(11;18) translocation. Patients were subdivided into patients without t(11;18) translocation or extra copies of MALT1(Group A,n = 88),patients with t(11;18) translocation(Group B,n = 27),and patients with extra copies of MALT1(Group C,n = 31). The clinical background,treatment,and outcomes of each group were *** Groups A and C showed slight female predominance,whereas Group B showed slight male predominance. Mean ages and clinical stages at lymphoma diagnosis were not different between groups. Complete response was obtained in 61 patients in Group A(69.3%),22 in Group B(81.5%),and 21 in Group C(67.7%). Helicobacter pylori(H. pylori) eradication alone resulted in complete remission in 44 patients in Group A and 13 in Group C. In Group B,14 patients underwent radiotherapy alone,which resulted in lymphoma disappearance. Although the difference was not statistically significant,event-free survival in Group C tended to be inferior to that in Group A(P = 0.10).COnCLUSIOn Patients with t(11;18) translocation should be treated differently from others. Patients with extra copies of MALT1 could be initially treated with H. pylori eradication,similar to patients without t(11;18) translocation or extra copies of MALT1.
