Suppression of monosodium urate crystal-induced inflammation by inhibiting TGF-β-activated kinase 1-dependent signaling:role of the ubiquitin proteasome system

Monosodium urate(MSU)crystals activate inflammatory pathways that overlap with interleukin-1β(IL-1β)***,the post-translational mechanisms involved and the role of signaling proteins in this activation are *** the present study,we investigated the intracellular signaling mechanisms involved in MSU-induced activation of THP-1 macrophages and human nondiseased synovial fibroblasts(NLSFs)and the in vivo efficacy of an inhibitor of tumor grovArth factor-β(TGF-β)-activated kinase 1(TAK1),5Z-7-oxozeaenoI,in MSU-induced paw inflammation in C57BL76 ***-1 macrophage activation with MSU crystals(25-200 ng/ml)resulted in the rapid and sustained phosphorylation of interleukin-1 receptor-activated kinase 1(IRAK1 Thr^(209))and TAK1(Thr^(184/187))and their association with the E3 ubiquitin ligase *** the cellular level,MSU inhibited the deubiquitinases A20 and UCHL2 and increased 20s proteasomal activity,leading to a global decrease in K^(63)-linked ubiquitination and increase in K^(48)-linked ubiquitination in THP-1 *** MSU did not stimulate cytokine produaion in NLSFs,it significantly amplified IL-1β-induced IL-6,IL-8,and ENA-78/CXCL5 *** studies and MD simulations followed by TAK1 in vitro kinase assays revealed that uric acid molecules are capable of arresting TAK1 in an active-state conformation,resulting in sustained TAK1 kinase ***,MSU-induced proinflammatory cytokine production was completely inhibited by 5Z-7-oxozeaenol but not IRAK1/4 or TRAF6 *** of 5Z-7-oxozeaenol(5 or 15 mg/kg;orally)significantly inhibited MSU-induced paw inflammation in C57BL/6 *** study identifies a novel post-translational mechanism of TAK1 activation by MSU and suggests the therapeutic potential of TAK1 in regulating MSU-induced Inflammation.

Phytosomes(phytophospholipid complex) are dosage forms that have recently been introduced to increase the stability and therapeutic effect of herbal medicine. Currently, bioactive herbs and the phytochemicals they contain are considered to be the best remedies for chronic diseases. One promising approach to increase the efficacy of plant-based therapies is to improve the stability and bioavailability of their bio-active ingredients. Phytosomes employ phospholipids as their active ingredients, and use their amphiphilic properties to solubilize and protect herbal extracts. The unique properties of phospholipids in drug delivery and their use in herbal medicines to improve bioavailability results in significantly enhanced health benefits. The introduction of phytosome nanotechnology can alter and revolutionize the current state of drug delivery. The goal of this review is to explain the application of phytosomes, their future prospects in drug delivery, and their advantages over conventional formulations.

Effects of various parameters on the attrition of bed material in a recirculating fluidized bed with a draft tube

We performed an experimental study to investigate the effects of various parameters on the attrition of bed material and its size distribution with increasing operation time in a recirculating fluidized bed （RCFB）. The studied parameters included superficial velocity of fluidizing air, bed inventory, and spacing between the jet top and draft tube bottom （spacer height）. The bed material was prepared from Indian Standard （IS） Grade I sand from sieves with a size range of 2.20-1.00 mm. Experiments were performed at ambient conditions, with the superficial air velocity ranging from 7.13-9.16 m/s, a bed inventory of 7-10 kg, spacing of 0.085 and 0.045 m between the jet top and draft tube bottom, and an operating time of 40 h. We investigated the influence of these parameters in terms of changes in the size distribution of particles, changes in the %-weight of particles of different size ranges, generation of particles with smaller diameters, the decrease of the downcomer bed height, variations in the coefficient of uniformity and coefficient of curvature, and material loss from entrainment of fines with increasing operation time. The mode of attrition was abrasion in all experiments. We found that with increasing operation time and other parameters （bed inventory, superficial air velocity, and spacer height） attrition of the bed material also increased. Generation and elutriation of fines were more pronounced at higher superficial air velocity, bed inventory, and spacer height.

Seismic response of precast reinforced concrete wall subjected to cyclic in-plane and constant out-of-plane loading

This paper provides insight into the seismic behavior of a full-scale precast reinforced concrete wall under in-plane cyclic loading combined with out-of-plane loading replicated by sand backfill to simulate the actual condition of basement *** tested wall exhibited flexural cracks,owing to the high aspect ratio and considerable out-of-plane movement due to lateral pressure from the *** wall performed satisfactorily by exhibiting competent seismic parameters and deformation characteristics governed by its ductile response in the nonlinear phase during the test with smaller residual *** analysis was conducted to validate experimental findings,which complied with each *** numerical model was used to conduct parametric studies to study the effect of backfill density and aspect ratio on seismic response of the proposed precast wall *** in-plane capacity of walls reduced,while deformation characteristics were unaffected by the increase in backfill *** increase in aspect ratio leads to a reduction in in-plane capacity and an increase in *** between the ratio of in-plane yield capacity and design shear load of walls are proposed for the backfill density,which may be adopted to determine the in-plane yield capacity of the basement walls based on their design shear.

The synthesis of bioactive steroids is catalyzed by an array of enzymes of diverse properties and *** the present study,seasonal dynamics and kinetics of key steroidogenic enzymes,17α-hydroxylase(Cyp17a),3β-hydroxysteroid dehydrogenase(Hsd3b),20α-hydroxysteroid dehydrogenase(Hsd20a)and 20β-hydroxysteroid dehydrogenase(Hsd20b)were investigated in the female catfish Heteropneustes ***,the effects of the estrogen metabolite 2-hydroxyestradiol-17β(2-OHE2)and human chorionic gonadotropin(hCG)on activities of the above enzymes,cytochrome P450 aromatase(Cyp19a1),and steroid products including testosterone and cortisol were *** enzymes under investigation showed significant seasonal variations across the annual ovarian cycle with low activity in the gonad resting *** enzymes Hsd3b and Cyp17a showed high activity during early oogenesis but the activities of Hsd20a and Hsd20b were higher towards late oogenesis in the spawning ***3b and Cyp17a elicited high apparent Km values(low substrate affinity)and high apparent Vmax in the vitellogenic phase compared to the postvitellogenic ***20a did not elicit any significant differences in the kinetic parameters between the two ***20b showed high apparent Km values(low substrate affinity)and high Vmax in the postvitellogenic *** incubation of ovarian slices with 2-OHE2 for 24 h increased dose-dependently Hsd3b,Cyp17a,Hsd20a and Hsd20b activities,similar to *** levels of the corresponding C21 steroid products,progesterone(P4),17α-hydroxyprogesterone(17-OHP4),17,20α-dihydroxy-4-pregnen-3-one(17,20α-DP)and 17,20β-dihydoxy-4-pregnen-3-one(17,20β-DP),and cortisol were ***,2-OHE2 decreased significantly the C19 and C18 steroids,testosterone and E2 levels,and Cyp19a *** co-incubation with hCG and 2-OHE2 produced a synergistic effect on the enzyme activities except that of *** co-incubation reversed the inhibitory effect of *** data show that

Multiple Impact of Integrated Watershed Management in Low Rainfall Semi-Arid Region: A Case Study from Eastern Rajasthan, India

The agriculture in low rainfall areas of eastern Rajasthan, India is characterized by high risks from drought, degraded natural resources and pervasive poverty, food insecurity and malnutrition. In this region, water is the main limiting factor for upgrading rainfed agriculture. For such areas integrated watershed management is recognized as a potential approach for agriculture growth and rehabilitation of fragile and degraded lands. At Gokulpura-Goverdhanpura village in Bundi eastern Rajasthan, India an integrated watershed project was implemented using the holistic systems approach. This paper discusses the impacts of this watershed program on bio-physical, socio-economic, environmental and ecological parameters. Results indicate that due to watershed interventions the groundwater availability has substantially increased which brought changes in cropping patterns with high value crops. Significant increases in irrigated area, cropping intensity along with diversification of crops from traditional to commercial cash crops were recorded. The watershed program also significantly improved the socio-economic status of the watershed community. It has increased the income and reduced poverty of the people in the watershed. The watershed interventions generated good employment opportunities and significantly reduced the migration of both skilled and unskilled labor from the watershed village to urban areas. It has also improved the environmental quality and ecological status in the watershed. The watershed interventions increased the vegetative index or greenery, reduced runoff, soil loss, and land degradations and improved the bio-diversity in fragile ecosystems. Overall, the integrated watershed program at Gokulpura-Goverdhanpura provided resilience by ensuring continued and sustainable multiple outputs, besides soil and water conservation and other positive environmental effects.

Influential aspects on seismic performance of confined masonry construction

Recent earthquakes around the world have resulted in loss of human lives and high economic losses due to poor performance of unreinforced masonry constructions as well as poorly-built reinforced concrete framed buildings. This has necessitated alternative building technologies with improved seismic performance. Confined masonry (CM) construction, has shown excellent behavior during past earthquakes across the world and requires similar skill at a marginally higher cost than that of unreinforced masonry. This paper summarizes the main features of generic construction and gains insight into the behavior of CM elements under earthquake excitations, representing a viable alternative for safe and economical construction in seismic areas.?The paper discusses various influential aspects like sequence of construction, properties and type of masonry material, structural configuration, reinforcement detailing in tie column/beam and masonry, panel aspect ratio, interface between concrete and masonry, axial stress, multiple confining column, opening in wall panels and damage pattern etc. along with solution to overcome the limitations.
