Plant Growth Promoting Rhizobacteria Having 1-Aminocyclopropane-1-Carboxylic Acid Deaminase to Induce Salt Tolerance in Sunflower (<i>Helianthus annus L.</i>)

Soil salinity badly affects agriculture productivity through accumulation of salts in upper layers of soils. The harmful effects of salts in arable lands have influenced modern as well as ancient civilizations. A pot study was carried out to test the performance of two PGPR isolates (KS 8, KS 28) on sunflower (SMH-0917) under different salinity levels (8, 10 and 12 dS·m-1). These salinity levels were developed by adding calculated amount of salts (NaCl, Na2SO4, CaCl2 and MgSO4) with ratio of 3:4:2:1. The bacterial strains KS 8 and KS 28 were applied separately in two treatments while third treatment was co-inoculation (KS mix). Completely randomized experimental design (CRD) was used and data were collected at flowering stage about pre-decided plant growth parameters (plant height, shoot dry weight and root dry weight). The bacterial isolate KS 8 showed an increase of 26, 102% and 83% in plant height, shoot dry weight and root dry weight at EC 8 dS·m-1, while this improvement was 67%, 163% and 296% at EC 10 dS·m-1, however an increase of 100%, 74% and 382% was recorded over control respectively at EC 12 dS·m-1. Similarly isolate KS 28 exhibited an increase of 14%, 69% and 54% in plant height;shoot dry weight and root dry weight at EC 8 dS·m-1, whereas this improvement was 56%, 163% and 188% at EC 10 dS·m-1, while an increase of 60%, 41% and 282% was registered respectively over control at EC 12 dS·m-1. The increase due to mixture treatments was 4%, 41% and 16% in plant height, shoot dry weight and root dry weight at EC 8 dS·m-1, while an increase of 33%, 57% and 100% at EC 10 dS·m-1, whereas an improvement of 53%, 33% and 164% respectively was noted at EC 12 dS·m-1 over un-inoculated. The isolate KS 8 performed better than KS 28 and mixture treatment. These two PGPR strains could be used to mitigate the adverse impact caused by salinity stress on sunflower.

Optimizing Yield and Nutrients Content in Tomato by Vermicompost Application under Greenhouse Conditions

Optimizing yield and nutrients content in crop production is the high time demand of arable farming. Vermicompost furnishes one of the most promising alternatives to costly chemical fertilizer. Therefore, a greenhouse experiment was conducted at NARC, Islamabad, to investigate the effect of different levels of vermicompost [having C/N=15/1 and 14 Plant Growth Promoting Rhizobacteria (PGPR)] on growth, yield and nutrients content in hybrid tomato (National Tunnel Tomato-04-08) under greenhouse conditions during Rabi 2015. The data revealed that different rates (0.5 t·ha-1, 1 t·ha-1, 1.5 t·ha-1 and 2 t·ha-1) of vermicompost produced varied and significant (P -1) at the application of 2.0 t vermicompost ha-1 followed by 3.226 t·ha-1 where vermicompost was applied @ 1.5 t·ha-1. N, P and K content in tomato fruit and plant increased significantly with the application of increasing levels of vermicompost. The highest content of N (3.7%), P (0.67%), K (5.17%) in tomato fruit and N (3.4%), P (0.32%), K (3.2%) in tomato plant respectively were registered with soil application of vermicompost @ 2.0 t·ha-1. This study confirms that the vermicompost has a tremendous potential of plant nutrients supply for sustainable crop production.

Economics of Residues Incorporation and Phosphorus Application for Direct Seeded Rice and Wheat under Saline Soil

Two-year long field study was conducted using a permanent layout to investigate the economics of crop residues incorporation (2 t·ha-1) and P application (0, 40, 80 and 120 kg P2O5 ha-1) to directly sowing of rice and wheat crops gown under naturally salt-affected calcareous soil (ECe = 4.59 dS m-1;pHs = 8.38;SAR = 6.57 (mmolc L-1)1/2;CaCO3 = 3.21%;Extractable P = 4.07 mg·kg-1;sandy clay loam) at farmers field in district Hafizabad during the year 2012-13. Split plot design (crop residues in main plots and P application in sub plots) was followed with three replications. Agronomic data on growth and yield were collected at the time of each crop maturity. Maximum growth and yield of both the crops were harvested from the plots where P2O5 was applied @ 80 kg·ha-1 along with crop residues incorporation. On an average of two years, maximum paddy (3.26 t·ha-1) and wheat grain (3.56 t·ha-1) yield were produced with P application @ 80 kg P2O5 ha-1 along with crop residues incorporation. Although, the yield harvested with this treatment (80 kg P2O5 ha-1 + crop residues) performed statistically equal to 120 kg P2O5 ha-1 without crop residues incorporation during both the years, however, on an average of two years, grain yield of directly sowing rice and subsequent wheat was significantly superior (22% and 24% respectively) than that of higher P rate (120 kg·ha-1) without crop residues. Overall, continuous two-year crop residues incorporation further increased (17%) paddy yields during the follow up year of crop harvest. Economic analyses of both the crops were carried out to choose the best treatment with adequate economic benefits as compared to those without crop residue incorporation. Maximum net benefit of Rs = 108,680/- for direct seeded rice and Rs = 99,362/- for wheat grown with 80 kg P2O5 ha-1 application under crop residues incorporation was determined. Modelling of debris-flow susceptibility and propagation: a case study from Northwest Himalaya

The geological and geographical position of the Northwest Himalayas makes it a vulnerable area for mass movements particularly landslides and debris flows. Mass movements have had a substantial impact on the study area which is extending along Karakorum Highway(KKH) from Besham to Chilas. Intense seismicity, deep gorges, steep terrain and extreme climatic events trigger multiple mountain hazards along the KKH, among which debris flow is recognized as the most destructive geohazard. This study aims to prepare a field-based debris flow inventory map at a regional scale along a 200 km stretch from Besham to Chilas. A total of 117 debris flows were identified in the field, and subsequently, a point-based debris-flow inventory and catchment delineation were performed through Arc GIS analysis. Regional scale debris flow susceptibility and propagation maps were prepared using Weighted Overlay Method(WOM) and Flow-R technique sequentially. Predisposing factors include slope, slope aspect, elevation, Topographic Roughness Index(TRI), Topographic Wetness Index(TWI), stream buffer, distance to faults, lithology rainfall, curvature, and collapsed material layer. The dataset was randomly divided into training data(75%) and validation data(25%). Results were validated through the Receiver Operator Characteristics(ROC) curve. Results show that Area Under the Curve(AUC) using WOM model is 79.2%. Flow-R propagation of debris flow shows that the 13.15%, 22.94%, and 63.91% areas are very high, high, and low susceptible to debris flow respectively. The propagation predicated by Flow-R validates the naturally occurring debris flow propagation as observed in the field surveys. The output of this research will provide valuable input to the decision makers for the site selection, designing of the prevention system, and for the protection of current infrastructure.

Face recognition (FR) technology has numerous applications in artificial intelligence including biometrics, security,authentication, law enforcement, and surveillance. Deep learning (DL) models, notably convolutional neuralnetworks (CNNs), have shown promising results in the field of FR. However CNNs are easily fooled since theydo not encode position and orientation correlations between features. Hinton et al. envisioned Capsule Networksas a more robust design capable of retaining pose information and spatial correlations to recognize objects morelike the brain does. Lower-level capsules hold 8-dimensional vectors of attributes like position, hue, texture, andso on, which are routed to higher-level capsules via a new routing by agreement algorithm. This provides capsulenetworks with viewpoint invariance, which has previously evaded CNNs. This research presents a FR model basedon capsule networks that was tested using the LFW dataset, COMSATS face dataset, and own acquired photos usingcameras measuring 128 × 128 pixels, 40 × 40 pixels, and 30 × 30 pixels. The trained model outperforms state-ofthe-art algorithms, achieving 95.82% test accuracy and performing well on unseen faces that have been blurred orrotated. Additionally, the suggested model outperformed the recently released approaches on the COMSATS facedataset, achieving a high accuracy of 92.47%. Based on the results of this research as well as previous results, capsulenetworks perform better than deeper CNNs on unobserved altered data because of their special equivarianceproperties.

A New Approach for the Determination of Metronidazole Using Switchable Solvent Coupled with Au-MWCNTs-Modified Electrochemical Sensor

In this work,a new liquid-phase microextraction method coupled with an electrochemical technique using a switchable solvent was proposed for the determination of *** extraction solvent used was dipropylamine(DPA),which exhibited switchable *** means that it can become miscible or immiscible when in contact with carboxylic acids in the aqueous *** metronidazole was then measured using a glassy carbon electrode that was modified with Au-multiwalled carbon nanotubes(MWCNTs).The eff ect of diff erent parameters such as the type and amount of trigger,the addition of salt,and the volume of extraction solvent on the e fficiency of switchable hydrophilic solvent-based liquid-phase microextraction(SHS-LPME)was investigated using the one-factor-at-a-time *** optimizing the conditions,the linear calibration curve was obtained in the concentration range of 0.005–250μmol/***,the limit of quantification(LOQ)of 0.005μmol/L and the limit of detection(LOD)of 0.0015μmol/L were obtained,respectively.

Analysis of Genetic Alterations in TP53 Gene in Breast Cancer - A Secondary Publication

Tumor protein p53 (TP53) mediates DNA repair and cell proliferation in growing cells. The TP53 gene is a tumor suppressor that regulates the expression of target genes in response to multiple cellular stress factors. Key target genes are involved in crucial cellular events such as DNA repair, cell cycle regulation, apoptosis, metabolism, and senescence. TP53 genetic variants and the activity of the wild-type p53 protein (WT-p53) have been linked to a wide range of tumorigenesis. Various genetic and epigenetic alterations, including germline and somatic mutations, loss of heterozygosity, and DNA methylation, can alter TP53 activity, potentially resulting in cancer initiation and progression. This study was designed to screen three reported mutations in the DNA-binding domain of the p53 protein in breast cancer, to evaluate the relative susceptibility and risk associated with breast cancer in the local population. Genomic DNA was isolated from 30 breast tumor tissues along with controls. Tetra and Tri ARMS PCR were performed to detect mutations in the TP53 coding region. For SNPs c.637C>T and c.733C>T, all analyzed cases were homozygous for the wild-type allele ‘C,’ while for SNP c.745A>G, all cases were homozygous for the wild-type allele ‘A.’ These results indicate no relevance of these three SNPs to cancer progression in our study cohort. Additionally, the findings from whole exon sequencing will help to predict more precise outcomes and assess the importance of TP53 gene mutations in breast cancer patients.
