Molecular epidemiology, characterization of virulence factors and antibiotic resistance profile of Streptococcus agalactiae isolated from dairy farms in China and Pakistan

Streptococcus agalactiae is one of the most common pathogens that cause bovine mastitis worldwide. Identifying pathogen prevalence and virulence factors is critical for developing prevention and control approaches. Herein, 1 161 milk samples from various dairy farms in China(n=558) and Pakistan(n=603) were collected between 2019–2021 and were subjected to S. agalactiae isolation. Prevalence, serotyping, virulence genes, and antibiotic-resistant genes of S. agalactiae were evaluated by PCR assay. All isolates were characterized for haemolysis, biofilm production, cytotoxicity, adhesion, and invasion on bovine mammary epithelial cells. The prevalence of S. agalactiae-induced mastitis in cattle was found to be considerably higher in Pakistan than in China. Jiangsu and Sindh provinces had the highest area-wise prevalence in China and Pakistan, respectively. Serotypes Ia and II were prevalent in both countries, whereas serotype III was found only in Pakistan. Moreover, all isolates tested positive for PI-2b gene but negative for PI-1 and PI-2a genes. All isolates harboured cfb, cylE, hylB, and fbsB virulent genes, whereas many of them lacked bibA, rib and bca. However, the absence of bac and scp genes in Chinese isolates and cspA in Pakistani isolates was noted, while spb1 and lmb were not detected in isolates of both countries. Pakistani isolates, particularly serotype Iapositive, had a considerably higher ability to produce biofilm, haemolysis, cytotoxicity, adhesion, and invasion than Chinese isolates. Most of the isolates were phenotypically resistant to tetracycline, erythromycin, and clindamycin and genotypic resistance was confirmed by the presence of ermA, ermB, tetM and tetO genes. Our study highlights the antimicrobial resistance profile and virulence-related factors contributing to the epidemiological spread of mastitis-causing S. agalactiae in China and Pakistan. Zika virus(ZIKV)is the causative agent of a viral infection that causes neurological complications in newborns and adults *** wide transmission route and alarming spread rates are of great concern to the scientific *** trials have been conducted to develop treatment options for ZIKV *** review highlights the latest developments in the fields of vaccinology and pharmaceuticals developments for ZIKV infection.A systematic and comprehensive approach was used to gather relevant and up-to-date data so that inferences could be made about the gaps in therapeutic *** results indicate that several therapeutic interventions are being tested against ZIKV infection,such as DNA vaccines,subunit vaccines,live-attenuated vaccines,virus-vector-based vaccines,inactivated vaccines,virus-like particles,and mRNA-based *** addition,approved anti-ZIKV drugs that can reduce the global burden are *** many vaccine candidates for ZIKV are at different stages of development,none of them have received Food and Drug Authority approval for use up to *** issue of side effects associated with these drugs in vulnerable newborns and pregnant women is a major obstacle in the therapeutic pathway.

Developing a simple scalable method to fabricate electrodes with high capacity and wide voltage range is desired for the real use of electrochemical ***,we synthesized amorphous NiCo-LDH nanosheets vertically aligned on activated carbon cloth substrate,which was in situ transformed from Co-metal-organic framework materials nano-columns by a simple ion exchange process at room *** to the amorphous and vertically aligned ultrathin structure of NiCo-LDH,the NiCo-LDH/activated carbon cloth composites present high areal capacities of 3770 and 1480 mF cm^(-2)as cathode and anode at 2 mA cm^(-2),and 79.5%and 80%capacity have been preserved at 50 mA cm^(-2).In the meantime,they all showed excellent cycling performance with negligible change after>10000 *** fabricating them into an asymmetric supercapacitor,the device achieves high energy densities(5.61 mWh cm^(-2)and 0.352 mW cm^(-3)).This work provides an innovative strategy for simplifying the design of supercapacitors as well as providing a new understanding of improving the rate capabilities/cycling stability of NiCo-LDH materials.

For microelectronic devices,the on-chip microsupercapacitors with facile construction and high performance,are attracting researchers'prior consideration due to their high compatibility with modern ***,we proposed interchanging interdigital Au-/MnO_(2)/polyethylene dioxythiophene stacked microsupercapacitor based on a microfabrication process followed by successive electrochemical *** stacked configuration of two pseudocapacitive active microelectrodes meritoriously leads to an enhanced contact area between MnO_(2)and the conductive and electroactive layer of polyethylene dioxythiophene,hence providing excellent electron transport and diffusion pathways of electrolyte ions,resulting in increased pseudocapacitance of MnO_(2)and polyethylene *** stacked quasi-solid-state microsupercapacitors delivered the maximum specific capacitance of 43 mF cm^(-2)(211.9 F cm^(-3)),an energy density of 3.8μWh cm^(-2)(at a voltage window of 0.8 V)and 5.1μWh cm^(-2)(at a voltage window of 1.0 V)with excellent rate capability(96.6%at 2 mA cm^(-2))and cycling performance of 85.3%retention of initial capacitance after 10000 consecutive cycles at a current density of 5 mA cm^(-2),higher than those of ever reported polyethylene dioxythiophene and MnO_(2)-based planar *** from the favorable morphology,bilayer microsupercapacitor is utilized as a flexible humidity sensor with a response/relaxation time superior to those of some commercially available integrated *** strategy will be of significance in developing high-performance on-chip integrated microsupercapacitors/microsensors at low cost and environment-friendly routes.

Coccidiosis is an endemic problem in broilers inflicting disastrous losses to worldwide poultry *** are generally effective in controlling the infectious diseases;the subunit vaccines effectiveness can hopefully be improved with concurrent use of probiotics as assessed in this experiment using an Eimeria tenella *** mapped protein-1(IMP 1)is a novel immunogenic protein of Apicomplexans,including in Eimeria *** performance of the cumulative effect of probiotics and EtIMP1 C(Part of Eimeria tenella immune mapped protein1) was evaluated based on various parameters such as an intestinal lesion,oocyst scores,feed conversion ratio,and organ *** were also analyzed on both immunological and hematological *** EtIMP1 C and probiotics administered group showed less intestinal lesion,decreased oocyst shedding,satisfactory feed conversion ratio(FCR) and improved hematological parameters as compared to EtIMP1 C emulsified with FCA ***,there was no statistically significant difference between the two groups(P>0.05),in aspect of lesion and oocyst scores,as well as immunological and hematological *** experimental work showed that probiotics could be a good hope due to its antioxidant,immunomodulatory and growth promoting effect against poultry coccidiosis alone or in combination with vaccines,including IMP1 C based vaccine,but further studies are required to formulate its dose with the vaccine as well as a different strain of probiotics effect against coccidiosis.

Dendritic cells play a crucial role in inducing antigen-specific immunity to *** host-parasite interaction,host immune response to the parasite molecules is considered essential for recognizing novel antigens for control ***,in the present study,chicken dendritic cells(DCs)(ChDCs),derived from spleens were used to evaluate their capacity to proliferate and differentiate autologous T lymphocytes in response to actin-depolymerizing factor from Eimeria tenella(EtADF).Immunoblot analysis showed that recombinant EtADF protein(rEtADF) was able to interact with rat anti-rEtADF *** immunofluorescence test confirmed rEtADF binding on ChDCs *** *** revealed that phenotypes for MHCII,CD1.1,CD11 c,CD80,and CD86 were increased in ChDCs after rEtADF ***-PCR results indicated that ChDCs triggered TLR signaling in response to rEtADF,and suppressed Wnt *** levels of CD83,CCL5,and CCR7 in ChDCs were improved following rEtADF *** addition,rEtADF promoted DC-directed T cell proliferation and differentiation of naive T cells into CD3/CD4T cells in DC/T cell co-incubation *** analysis of rEtADF-pulsed ChDCs showed increased levels of IL-12 and IFN-γ,while IL-10 and TGF-β remained ***,rEtADF-treated ChDCs enhanced production of IFN-γ when incubated with T cells,and IL-4 secretion remained *** findings indicted that rEtADF could facilitate the polarization of Th1 immune cells by triggering both host DCs and T *** findings provide useful insights into future work aimed at anticoccidial vaccine strategies.

Dendritic cells(DCs) play a pivotal role to amplify antigen-specific immune *** that sensitize T cells via antigen-presentation by DCs could enhance the capacity of host immunity to fight *** this study,we tested the immunogenic profiles of chicken DCs towards Glyceraldehyde-3-phosphate dehydrogenase from Eimeria acervulina(EaGAPDH).Immunoblot analysis showed that recombinant EaGAPDH(rEaGAPDH) protein was successfully recognized by rat sera generated against *** and internalisation of rEaGAPDH by chicken splenic-derived DCs(chSPDCs) was confirmed by immunofluorescence *** cytometry revealed that chSPDCs upregulated MHCII,CD 1.1,CD11 c,CD80,and CD86 cell-surface ***,mRNA expressions of DC maturation biomarkers(CCL5,CCR7,and CD83) and TLR signalling genes(TLR15 and MyD88) were also upregulated whereas those of Wnt signalling were non-significant compared to negative *** treatment induced IL-12 and IFN-γ secretion in chSPDCs but had no effect on IL-10 and TGF-β.Likewise,DC-T cell co-culture promoted IFN-γ secretion and the level of IL-4 was *** of T cells and their differentiation into CD3+/CD4+T cells were triggered in chSPDCs-T cells co-culture *** together,rEaGAPDH could promote Th1 polarization by activating both host DCs and T cells and sheds new light on the role of this important molecule which might contribute to the development of new DCs-based immunotherapeutic strategies against coccidiosis.
