Detection of tumor stem cell markers in pancreatic carcinoma cell lines

BACKGROUND: Cancer of the pancreas is the fourth leading cause of cancer death in industrialized countries. In malignancy, actively proliferating cells may be effectively targeted and killed by anti-cancer therapies, but stem cells may survive and support re-growth of the tumor. Thus, new strategies for the treatment of cancer clearly will also have to target cancer stem cells. The goal of the present study was to determine whether pancreatic carcinoma cell growth may be driven by a subpopulation of cancer stem cells. Because previous data implicated ABCG2 and CD133 as stem cell markers in hematopoietic and neural stem/progenitor cells, we analyzed the expression of these two proteins in pancreatic carcinoma cell lines. METHODS: Five established pancreatic adenocarcinoma cell lines were analyzed. Total RNA was isolated and real- time RT-PCR was performed to determine the expression of ABCG2 and CD133. Surface expression of ABCG2 and CD133 was analyzed by flow cytometric analysis. RESULTS: All pancreatic carcinoma cell lines tested expressed significantly higher levels of ABCG2 than non-malignant fibroblasts or two other malignant non- pancreatic cell lines, i.e., SaOS2 osteosarcoma and SKOV3 ovarian cancer. Elevated CD133 expression was found in two out of five pancreatic carcinoma cell lines tested. Using flow cytometric analysis we confirmed surface expression of ABCG2 in all five lines. Yet, CD133 surface expression was detectable in the two cell lines, A818-6 and PancTu1, which exhibited higher mRNA ***: Two stem cell markers, ABCG2 and CD133 are expressed in pancreatic carcinoma cell lines. ABCG2 and/or CD133 positive cells may represent subpopulation of putative cancer stem cells also in this malignancy. Because cancer stem cells are thought to be responsible for tumor initiation and its recurrence after an initial response to chemotherapy, they may be a very promising target for new drug developments.

The residual effect of tobacco biochar(TB≥500°C)mono and co-application with Cahydroxide(CH),Ca-bentonite(CB)and natural zeolite(NZ)on the bio-availability of trace elements TE(s)in alkaline soils has not been deeply studied yet.A pot study that had earlier been investigated TB mono and blended with CH,CB and NZ on the immobilization of Pb,Cu Cd,and Zn by Chinese *** crop in the rotation was selected as test plant to assess the residual impact of amendments on stabilization of Pb,Cu Cd,and Zn in mine polluted(M-P),smelter heavily and low polluted(S-HP and S-LP,respectively)*** obtained results showed that stabilization of Pb,Cd,Cu and Zn reached 63.84% with TB+CB,61.19% with TB+CH,83.31% with TB+CH and 35.27% with TB+CH for M-P soil,36.46% with TB+NZ,38.46% with TB+NZ,19.40% with TB+CH and 62.43% with TB+CH for S-LP soil,52.94%TB+NZ,57.65% with TB+NZ,52.94% with TB+NZ,and 28.44% with TB+CH for S-LP ***,TB+CH and TB alone had mobilized Pb and Zn up to19.29% and 34.96% in M-P *** mobility of Zn reached 8.38% with TB+CB and 66.03% with TB for S-HP and S-LP *** uptake and accumulation of Pb,Cd,Cu and Zn in shoot and root were reduced in three polluted ***,the combination of TB along with CH,CB and NZ has been proven to be effective in Pb,Cd,Cu and Zn polluted mine/smelter soils restoration.

Convergence Curve for Non-Blind Adaptive Equalizers

In this paper a closed-form approximated expression is proposed for the Intersymbol Interference (ISI) as a function of time valid during the entire stages of the non-blind adaptive deconvolution process and is suitable for the noisy, real and two independent quadrature carrier input case. The obtained expression is applicable for type of channels where the resulting ISI as a function of time can be described with an exponential model having a single time constant. Based on this new expression for the ISI as a function of time, the convergence time (or number of iteration number required for convergence) of the non-blind adaptive equalizer can be calculated. Up to now, the equalizer’s performance (convergence time and ISI as a function of time) could be obtained only via simulation when the channel coefficients were known. The new proposed expression for the ISI as a function of time is based on the knowledge of the initial ISI and channel power (which is measurable) and eliminates the need to carry out any more the above mentioned simulation. Simulation results indicate a high correlation between the simulated and calculated ISI (based on our proposed expression for the ISI as a function of time) during the whole deconvolution process for the high as well as for the low signal to noise ratio (SNR) condition.

High sensitizer and activator concentrations have been increasingly examined to improve the performance of multi-color emissive upconversion(UC)nanocrystals(UCNC)like NaYF_(4):Yb,Er and first strategies were reported to reduce concentration quenching in highly doped *** luminescence(UCL)is,however,controlled not only by dopant concentration,yet by an interplay of different parameters including size,crystal and shell quality,and excitation power density(P).Thus,identifying optimum dopant concentrations requires systematic studies of UCNC designed to minimize additional quenching pathways and quantitative ***,we quantify the dopant concentration dependence of the UCL quantum yield(ΦUC)of solid NaYF_(4):Yb,Er/NaYF_(4):Lu upconversion core/shell nanocrystals of varying Yb3+and Er3+concentrations(Yb3+series:20%‒98%Yb3+;2%Er3+;Er3+series:60%Yb3+;2%‒40%Er3+).To circumvent other luminescence quenching processes,an elaborate synthesis yielding OH-free UCNC with recordΦUC of~9%and~25 nm core particles with a thick surface shell were *** Yb3+concentrations barely reduceΦUC from~9%(20%Yb3+)to~7%(98%Yb3+)for an Er3+concentration of 2%,thereby allowing to strongly increase the particle absorption cross section and UCNC *** an increased Er3+concentration reducesΦUC from~7%(2%Er3+)to 1%(40%)for 60%Yb3+.Nevertheless,at very high P(>1 MW/cm^(2))used for microscopic studies,highly Er3+-doped UCNC display a high brightness because of reduced *** findings underline the importance of synthesis control and will pave the road to many fundamental studies of UC materials.

Inspection of the Output of a Convolution and Deconvolution Process from the Leading Digit Point of View—Benford’s Law

In the communication field, during transmission, a source signal undergoes a convolutive distortion between its symbols and the channel impulse response. This distortion is referred to as Intersymbol Interference (ISI) and can be reduced significantly by applying a blind adaptive deconvolution process (blind adaptive equalizer) on the distorted received symbols. But, since the entire blind deconvolution process is carried out with no training symbols and the channel’s coefficients are obviously unknown to the receiver, no actual indication can be given (via the mean square error (MSE) or ISI expression) during the deconvolution process whether the blind adaptive equalizer succeeded to remove the heavy ISI from the transmitted symbols or not. Up to now, the output of a convolution and deconvolution process was mainly investigated from the ISI point of view. In this paper, the output of a convolution and deconvolution process is inspected from the leading digit point of view. Simulation results indicate that for the 4PAM (Pulse Amplitude Modulation) and 16QAM (Quadrature Amplitude Modulation) input case, the number “1” is the leading digit at the output of a convolution and deconvolution process respectively as long as heavy ISI exists. However, this leading digit does not follow exactly Benford’s Law but follows approximately the leading digit (digit 1) of a Gaussian process for independent identically distributed input symbols and a channel with many coefficients.

Morphology and glucosinolate profiles of chimeric Brassica and the responses of Bemisia tabaci in host selection,oviposition and development

Plant structures and chemicals, which are developed from the shoot apical meristem（SAM）, form the main barriers to insect feeding. A plant chimera containing cells of different genetic origins in the SAM will be morphologically and chemically different compared with the parents and thus may result in differential resistance to herbivores. In this study, we explore if particular elements of plant resistance are localized in one of the layers of SAM; the replacement of one cell layer in a chimera may be linked to change of a single resistance trait to herbivores. The morphology and glucosinolate profiles of two periclinal chimeras（labeled as TTC and TCC, respectively） and grafted parents tuber mustard（labeled as TTT） and red cabbage（labeled as CCC） were compared and the performance of whitefly（Bemisia tabaci） in host selection, oviposition preference and development were assessed under controlled conditions. Both chimeras possessed leaf trichomes as parent tuber mustard TTT, however, TTC had significantly more trichomes than TCC and parent TTT. Leaf wax content of both chimeras was intermediate between the two parents. Five aliphatic and two indole glucosinolates were detected in both chimeras, whereas three aliphatic glucosinolates（3-methyl-sulfinylpropyl, 4-methyl-sulfinylbutyl and 2-hydroxy-3-butenyl） were not detected in tuber mustard, and one aliphatic glucosinolate（3-butenyl） was not detected in red cabbage. Unexpectedly for a chimera, the quantities of two aliphatic glucosinolates（3-methyl-sulfinylpropyl and 4-methyl-sulfinylbutyl） in both TTC and TCC were 3-to 5-fold higher than parents. In olfactory preference assays, B. tabaci showed preference to CCC, followed by TCC, TTC and TTT, and number of eggs laid showed the same pattern： CCC〉TCC〉TTC〉TTT. Interestingly, more whiteflies landed on TTT plants than the other three types in a free choice experiment and the developmental duration from egg to adult was the shortest on TTT and increased in the order TTT

Poland has a unique history of traditional Chinese medicine(TCM)dating back to the 17th century when Polish missionary Michael(Michal)Boym was a pioneer in the *** the 20th century,his successor,Professor Zbigniew Garnuszewski,reintroduced acupuncture to medical practice in ***,other methods of TCM and its holistic approach to patient care have not found their place in modern medicine in *** present,the legal status of TCM in Poland remains unregulated,with TCM included in the broad spectrum of complementary and alternative medicine(CAM)*** reports are available on the use of TCM methods among the Polish *** medicine combines conventional medicine with evidence-based CAM interventions and considers all aspects of a patient's health,including physical,emotional,mental,social,and environmental *** integrative healthcare model that incorporates TCM modalities and lifestyle recommendations as well as a whole person approach may provide a more sustainable solution for the constantly underfinanced Polish healthcare system,which faces challenges of multimorbidity in an aging society and limited access to *** coronavirus disease 2019 pandemic,war in Ukraine,and ongoing climate crisis have underscored the need to strengthen the resilience of the Polish healthcare system and search for new solutions.A model of care that blends the best of biomedicine and TCM healing approaches may be a better option for both patients and the healthcare system in Poland.

This paper describes a formation of hybrid coatings on a Ti-2Ta-3Zr-36Nb *** is accomplished by plasma electrolytic oxidation and a dip-coating technique with poly(adipic anhydride)((C6H8O3)n)that is loaded with drugs:amoxicillin(C16H19N3O5S),cefazolin(C14H14N8O4S3)or vancomycin(C66H75Cl2N9O24·xHCl).The characteristic microstructure of the polymer was evaluated using scanning electron microscopy and confocal *** on the surface treatment,the surface roughness varied(between 1.53μm and 2.06μm),and the wettability was change with the over of time.X-ray photoelectron spectroscopy analysis showed that the oxide layer did not affect the polymer layer or loaded ***,the drugs lose their stability in a phosphate-buffered saline solution after 6.5 h of exposure,and its decrease was greater than 7%(HPLC analysis).The stability,drug release and concentration of the drug loaded into the material were precisely analyzed by high-performance liquid *** results correlated with the degradation of the polymer in which the addition of drugs caused the percent of degraded polymer to be between 35.5%and 49.4%after 1 h of material immersion,depending on the mass of the loaded drug and various biological responses that were ***,all of the coatings were cytocompatible with MG-63 osteoblast-like *** drug concentrations released from the coatings were sufficient to inhibit adhesion of reference and clinical bacterial strains(***).The coatings with amoxicillin showed the best results in the bacterial inhibition zone,whereas coatings with cefazolin inhibited adhesion of the above bacteria on the surface.

Viewpoints of the target population regarding barriers and facilitators of colorectal cancer screening in the Czech Republic

BACKGROUND Public awareness of colorectal cancer(CRC) and uptake of CRC screening remain challenges. The viewpoints of the target population(asymptomatic individuals older than 50) regarding CRC screening information sources and the reasons for and against participation in CRC screening are not well known in the Czech Republic. This study aimed to acquire independent opinions from the target population independently on the health *** To investigate the viewpoints of the target population regarding the source of information for and barriers and facilitators of CRC *** A survey among relatives(aged 50 and older) of university students was conducted. Participants answered a questionnaire about sources of awareness regarding CRC screening, reasons for and against participation, and suggestions for improvements in CRC screening. The effect of certain variables on participation in CRC screening was *** Of 498 participants, 478(96%) respondents had some information about CRC screening and 375(75.3%) had participated in a CRC screening test. General practitioners(GPs)(n = 319, 64.1%) and traditional media(n = 166, 33.3%) were the most common information sources regarding CRC screening. A lack of interest or time and a fear of colonoscopy or positive results were reported as reasons for non-participation. Individuals aged > 60 years [adjusted odds ratio(aOR) = 2.30, 95% confidence interval(CI)(1.42-3.71), P = 0.001], females(aOR =1.95, 95%CI(1.26-3.01) P = 0.003), and relatives of CRC patients(aOR = 4.17,95%CI(1.82-9.58) P = 0.001) were more likely to participate in *** regarding screening provided by physicians-GPs:(aOR = 8.11,95%CI(4.90-13.41), P < 0.001) and other specialists(aOR = 4.19, 95%CI(1.87-9.38),P = 0.001) increased participation in screening. Respondents suggested that providing better explanations regarding screening procedures and equipment for stool capturing could improve CRC screening *** GPs
