Outbreaks of Monkeypox(mpox)in over 100 non-endemic countries in 2022 represented a serious global health *** a neglected disease,mpox has become a global public health issue.A42R profilinlike protein from mpox(PDB ID:4QWO)represents a potential new lead for drug development and may interact with various synthetic and natural *** this report,the interaction of A42R profilinlike protein with six phytochemicals found in the medicinal plant Ficus religiosa(abundant in India)was *** on the predicted and compared protein–ligand binding energies,biological properties,IC_(50) values and toxicity,two compounds,kaempferol(C-1)and piperine(C-4),were *** characteristics and quantitative structure–activity relationship(QSAR)of these two compounds were determined,and molecular dynamics(MD)simulations were *** silico examination of the kaempferol(C-1)and piperine(C-4)interactions with A42R profilin-like protein gave best-pose ligandbinding energies of–6.98 and–5.57 kcal/mol,*** predicted IC_(50) of C-1 was 7.63 lM and 82 lM for *** data indicated that kaempferol and piperine are non-mutagenic,and the QSAR data revealed that piperlongumine(5.92)and piperine(5.25)had higher log P values than the other compounds *** simulations of A42R profilin-like protein in complex with C-1 and C-4 were performed to examine the stability of the ligand–protein ***/C and C-4 showed the highest affinity and activities,they may be suitable lead candidates for developing mpox therapeutic *** study should facilitate discovering and synthesizing innovative therapeutics to address other infectious diseases.

Background: Nitrogen(N) is a required macronutrient for cotton growth and productivity. Excessive N fertilizers are applied in agriculture for crop yield maximization, which also generates environmental pollution. Improving crop N use efficiency(NUE) is the most economical and desirable way of reducing fertilizer application and environmental pollution. NUE has been an important issue in cotton. So far there is no report on cotton NUE improvement via transgenic approach. Nin-like proteins(NLP) are transcription factors regulating NUE. We previously demonstrated that At NLP7 improved NUE and biomass when overexpressed in Arabidopsis. However, it is not known whether At NLP7 can be used to improve NUE in ***: To test the feasibility, we expressed At NLP7 in cotton and evaluated NUE and yield of the transgenic cotton in the field. Transgenic cotton showed improved NUE and yield under both low and high N conditions. In addition, plant biomass, amount of absorbed N, N contents, activities of N-assimilating enzymes, and the expression of N-related marker genes were significantly increased in transgenic cotton compared with the wild type control, suggesting that At NLP7 enhances NUE in ***: Together, our results demonstrate that At NLP7 is a promising candidate to improve NUE and yield in cotton.

Glutaredoxins(Grxs) constitute a superfamily of proteins executing diverse biological *** cerevisiae glutaredoxin Grx6 not only serves as a glutathione(GSH)-dependent oxidoreductase and a GSH S-transferase,but also an essential [2Fe-2S]-binding *** we report the dimeric structure of Grx6 C-terminal domain(holo-Grx6C),bridged by one [2Fe-2S] cluster coordinated by the active-site Cys136 and two external GSH *** comparison combined with multiple-sequence alignment demonstrated that holo-Grx6 C is similar to the [2Fe-2S] cluster incorporated dithiol Grxs,which share a highly conserved [2Fe-2S] cluster binding pattern and dimeric conformation,distinct from the previously identified [2Fe-2S] cluster ligated monothiol Grxs.

Cytochrome P450 monooxygenase are synthetically useful enzymes and promising versatile oxidative biocatalysts,which catalyze the monooxygenation of various organic ***,the practical use of P450 is limited by its co-enzyme NAD(P)H and complex electron transport *** HO can simple the catalytic cycle but suffer from poor activity of most P450 in the presence of *** overcome the problem,by mimicking the catalytic mechanism of natural fungal peroxygenase,we have constructed a unique strategy for the modification of P450 monooxygenase into its peroxygenase mode,resulting in versatile oxidation of non-native substrates in assistance of dual-functional small molecule(DFSM). This system afforded the best peroxygenase activity for epoxidation and sulfoxidation among those P450-HO system previously *** approach expanded the potential use of P450 s in organic *** combination with protein engineering,the DFSM-facilitated P450 peroxygenase has been used for the hydroxylation of inert aromatic and aliphatic C-H bond.
