In vitro inhibitory analysis of consensus siRNAs against NS3 gene of hepatitis C virus 1a genotype

Objective: To explore inhibitory effects of genome-specific, chemically synthesized siRNAs(small interference RNA) against NS3 gene of hepatitis C virus(HCV) 1a genotype in stable Huh-7(human hepatoma) cells as well as against viral replication in serum-inoculated Huh-7 cells. Methods: Stable Huh-7 cells persistently expressing NS3 gene were produced under antibiotic gentamycin(G418) selection. The cell clones resistant to 1 000 μg antibiotic concentration(G418) were picked as stable cell clones. The NS3 gene expression in stable cell clone was confirmed by RT-PCR and Western blotting. siRNA cell cytotoxicity was determined by MTT cell proliferation assay. Stable cell lines were transfected with sequence specific siRNAs and their inhibitory effects were determined by RT-PCR, real-time PCR and Western blotting. The viral replication inhibition by siRNAs in serum inoculated Huh-7 cells was determined by real-time PCR. Results: RT-PCR and Western blot analysis confirmed NS3 gene and protein expression in stable cell lines on day 10, 20 and 30 post transfection. MTT cell proliferation assay revealed that at most concentrated dose tested(50 nmol/L), siRNA had no cytotoxic effects on Huh-7 cells and cell proliferation remained unaffected. As demonstrated by the siRNA time-dependent inhibitory analysis, siRNA NS3-is44 showed maximum inhibition of NS3 gene in stable Huh-7 cell clones at 24(80%, P=0.013) and 48 h(75%, P=0.002) post transfection. The impact of siRNAs on virus replication in serum inoculated Huh-7 cells also demonstrated significant decrease in viral copy number, where siRNA NS3-is44 exhibited 70%(Pal RNA reduction as compared to NS3-is33, which showed a 64%(Pal copy number. siRNA synergism(NS3-is33 + NS3-is44) decreased viral load by 84%(Pal inhibition by each siRNA(i.e., 64%–70%(P<0.05) in serum-inoculated cells. Textile dyes are dramatic sources of pollution and non-aesthetic disturbance of aquatic life and therefore represent a potential risk of bioaccumulation that can affect living *** is imperative to reduce or eliminate these dyes from liquid effluents with innovative biomaterials and ***,this research aims to highlight the performance of Capparis spinosa L waste-activated carbon(CSLW-AC)adsorbent to remove crystal violet(CV)from an aqueous *** mechanism of CV adsorption on CSLW-AC was evaluated based on the coupling of experimental data and different characterization *** efficiency of the CSLW-AC material reflected by the equilibrium adsorption capacity of CV could reach more than 195.671 mg·g^(–1) when 0.5 g·L^(–1) of CSLW-AC(Particle size≤250μm)is introduced into the CV of initial concentration of 100 mg·L^(–1) at pH 6 and temperature 65℃ and in the presence of potassium ions after 60 min of contact time according to the one parameter at a time *** adsorption behavior of CV on CSLW-AC was found to be consistent with the pseudo-second-order kinetic model and Frumkin's linear isothermal *** thermodynamic aspects indicate that the process is physical,spontaneous,and *** optimization of the process by the Box Behnken design of experiments resulted in an adsorption capacity approaching 183.544 mg·g^(–1)([CV]=100 mg·L^(–1) and[CSLW-AC]=0.5 g·L^(–1) at 35 min).The results of the Lactuca sativa seeds germination in treated CV(70%),adsorbent solvent and thermal regeneration(more than 5 cycles),and process cost analysis(1.0484 USD·L^(–1))tests are encouraging and promising for future exploitations of the CSLW-AC material in different industrial fields.

Return to Dialysis after Kidney Graft Failure

Introduction: The transition period from renal transplantation to dialysis is associated with high morbidity and mortality. The aim of this study is to describe the clinical and paraclinical characteristics, therapeutic management and evolutionary profile of patients returning to dialysis after kidney graft failure. Material and Methods: This was a retrospective, descriptive study conducted in the Nephrology-Dialysis-Renal Transplant Department at university hospital IbnSina between January 1998 and December 2021. We included all renal transplant recipients who had experienced kidney graft dysfunction and returned to dialysis. Patients with a follow-up after return to dialysis of less than 1 year were excluded. Results: Among 166 renal transplant recipients, 20 returned to dialysis after a median renal graft life of 85.5 months [42 - 186], corresponding to a prevalence of 12%. The mean age of our patients was 38.7 ± 11.9 years, with a M/F sex ratio of 2.3. Dialysis was initiated urgently in 10 patients (50%). Hemodialysis was the most commonly used modality (75%). Central venous catheterization was used in 35% of cases, including tunneled catheters. General condition is impaired in all patients, with persistent hypertension in 70% of cases. Mean uremia was 2.35 ± 0.8 g/l, mean creatinine 116 ± 48.3 mg/l, giving a mean GFR of 5.1 ± 2.2 ml/min. Mean albuminemia was 32.9 ± 6 g/l and mean hemoglobinemia 8.6 ± 1.9 g/dl. During the first year of follow-up, none of the patients died. However, 13 patients required hospitalization, with a mean length of stay of 15 days. Eight patients were hospitalized for infections and 5 for renal graft intolerance syndrome. After a mean follow-up of 22 months, 6 patients were detransplanted following graft necrosis. Conclusion: Return to dialysis after RT is fraught with a high rate of complications. The management of these patients must be optimized to improve their vital prognosis and quality of life.

Parkinson's disease is a neurodegenerative disorder caused by loss of dopamine neurons in the substantia nigra pars compacta. Tremor, rigidity, and bradykinesia are the major symptoms of the disease. These motor impairments are often accompanied by affective and emotional dysfunctions which have been largely studied over the last decade. The aim of this study was to investigate emotional processing organization in the brain of patients with Parkinson's disease and to explore whether there are differences between recognition of different types of emotions in Parkinson's disease. We examined 18 patients with Parkinson's disease(8 men, 10 women) with no history of neurological or psychiatric comorbidities. all these patients underwent identical brain blood oxygenation level-dependent functional magnetic resonance imaging for emotion evaluation. Blood oxygenation level-dependent functional magnetic resonance imaging results revealed that the occipito-temporal cortices, insula, orbitofrontal cortex, basal ganglia, and parietal cortex which are involved in emotion processing, were activated during the functional control. Additionally, positive emotions activate larger volumes of the same anatomical entities than neutral and negative emotions. Results also revealed that Parkinson's disease associated with emotional disorders are increasingly recognized as disabling as classic motor symptoms. These findings help clinical physicians to recognize the emotional dysfunction of patients with Parkinson's disease.

大多数研究者认为,自十九世纪初期以来,世界人口增长率的迅速增加导致了生活在地球上的人口数量增大。尤其是在发展中国家,为了经济社会的全面发展,实现高层次的社会服务,必须积极促进教育发展,同时控制人口增长是必要的。在此背景下,自20世纪60年代中期以来,坦桑尼亚教育和人口之间的联系已经引起了许多学者和政策制定者们的重视。其中,初等教育日益被看作是大众教育的基础,尤其是在学前教育曾经不作为义务教育的坦桑尼亚。近年来,在坦桑尼亚,通过不断完善初等教育体系,发展初等教育提升国民文化素质,以提高生存技能的作用日益受到关注。本文研究首先系统分析了坦桑尼亚教育体系发展状况,初等教育发展状况;以及坦桑尼亚初等教育与中国的比较情况。在这些研究的基础上,特别分析了从哪些方面来完善坦桑尼亚的小学教育,以及其必要性。研究认为,坦桑尼亚是一个高生育率的国家,人口构成趋于年轻化,这使坦桑尼亚成为一个人口迅速增长的国家。尽管人口增长已经使很多家庭的生活捉襟见肘,然而,人们依然很少采用避孕措施。大家庭规模的决定因素之一是两性关系。虽然艾滋病流行,但是坦桑尼亚的人口还将继续快速增长。更重要的是,高生育率将成为未来人口规模的一个决定因素。人口继续快速增长将使国家实现社会、经济目标变得更加艰难。本论文最后提出关于坦桑尼亚如何控制人口数量、提高人口素质,和借鉴中国等国际经验,促进初等教育发展的对策建议。

An Assessment of Aquifer Potential in and around a Proposed Well Field Area near Madunaghat, Chattogram Using Isotopic Techniques

The study was undertaken for understanding the potential of deep aquifers as a source of safe drinking water and for assessing the status of groundwater aquifer near Madunaghat area, Chattogram. The specific issues are salinity, interconnectivity of the shallow and deep aquifers, mixing of groundwater with the adjacent Halda river water, recharge condition and groundwater age. The isotopic data suggest that most of the groundwater results from a mixture between recent recharge and an older component recharge under climatic conditions cooler than at present. The interconnectivity between shallow aquifers and river waters are mostly found in the line wells installed particularly in shallow depth (16 m) close to Halda river as evidenced from the similar tritium values of sampled line well water with that of the Halda river water. The groundwater in Madunaghat well field area is not affected by salinity, as it is evidenced by higher values of Na/Cl ratio of the groundwater samples compared to the sea water fresh water mixing line. The geochemistry of intermediate and deep groundwater is dominated by Na-Mg-HCO3 and Na-HCO3 type waters. Even the relationship between chloride and oxygen-18 (Cl-δ18O) depicts that the waters from the deep, intermediate and shallow wells do not fall on the seawater mixing line. The Carbon-14 contents of intermediate to deep groundwater samples vary from 16.2 to 59.3 pMC indicating the residence time in the range of 4300 to 15,000 years BP, i.e., the sourced water recharged the aquifers a long time ago. The intermediate and deep wells have water with arsenic concentrations less than the detection limit of 3.0 μg/L. Only a few shallow wells have arsenic concentrations greater than the detection limit varying from 13.7 - 47.4 μg/L, which is less than the DoE permissible limit (50 μg/L) implying that the groundwater at Madunaghat area is not affected by Arsenic contamination.

Infective Endocarditis in Hemodialysis: Descriptive Study

Introduction: Infective endocarditis (IE) is a complication associated with high mortality in chronic hemodialysis patients. The aim of our study is to describe the clinical, radiological, microbiological characteristics, and risk factors of mortality in hemodialysis patients with infective endocarditis. Methods: A retrospective study from November 2019 to November 2021 including hemodialysis patients with IE hospitalized in Ibn Sina hospital center in Rabat. Results: Eighteen hemodialysis patients were included. Mean age was 49.5 ± 18 years, sex ratio was 0.8. Catheters were the access sites most commonly used (72.3%). Median duration of hemodialysis was 12 [6;24] months. Staphylococcus species is major causative bacteria in 60%. The mitral valve was the most commonly affected (44%). Frequent complications including valve insufficiency (50%), septic embolism (66.7%). Four patients underwent surgery and eight died during hospitalization (44.5%). Mitral localization of vegetation was the only factor significantly associated with mortality in our study (P Conclusion: The catheter was the most commonly used access site compared to the arteriovenous fistula. This confirms that it’s the vascular access of choice for his low rate of infectious complication compared to the catheter.

Pleuroperitoneal Leak in Peritoneal Dialysis: A Series of 10 Cases

Pleuroperitoneal Leak (PPL) is a rare, but serious complication of Peritoneal Dialysis (PD). It is the result of congenital or acquired communication between the 2 peritoneal and pleural cavities, and is often linked to high Intra-Abdominal Pressures (IAP). Its diagnosis is clinical, radiological and biological. In this article, we report 10 cases of PPL identified over a period of 13 years (2006-2019), that is to say, a prevalence of 5%. We suspected the diagnosis of FPP in front of dyspnea alone in 5 cases (50%), dyspnea associated with cough in 4 cases (40%). Catheter migration was reported in 5 patients (50% of cases). None of our patients presented with peritonitis before FPP. The pleurisy was right in 7 patients (70%). The exploratory puncture of the Pleural Fluid (PF) was performed in 3 patients, and which showed results consistent with the PD fluid. The drop in infusion volumes was required in 4 patients (40%) with an increase in the number of daily exchanges of CAPD with IVs ranges from 1200 - 1500 ml. PD was suspended in 6 patients (60%) for an average of 40 days (21 days - 60 days), 2 withdrawals from the technique were reported after the 2nd recurrence (20%). PD was stopped straight away in 1 patient with a hypo-permeable peritoneum with unsatisfactory residual renal function and a second who freely chose to stay in HD. The clinical cases presented show the great variability of FPP in terms of clinical and radiological presentation and therapeutic management, and focus on the significant risk of a transient or permanent withdrawal from the technique. According to literature data, Video-assisted thoracoscopic surgery appears to be promising in the event of failure of conservative treatment, and will thus prolong the survival of the technique.
