Vitamin E supplementation may negatively affect preimplantation development and mitochondrial ultrastructure of vitrified murine embryos

Objective:To observe the effects of vitamin E on post-vitrification preimplantation development,gross morphology as well as mitochondrial distribution and ***:Twenty-four female C57BL/6NTac mice,aged 12-16 weeks,were randomly divided into four *** A did not receive any treatment and served as the control *** B was treated with corn oil stripped of tocopherols and served as the vehicle *** C was treated with 60 mg/kg body weight of tocotrienol-rich-fraction with corn oil stripped of *** D was treated with 60 mg/kg body weight of alpha-tocopherol with corn oil stripped of *** treatments were administered orally for 7 consecutive *** superovulation and mating with fertile males,2-cell stage embryos were harvested for *** vitrification development in vitro,gross morphology and ultrastructure were compared between ***:The number of 2 and 8-cell embryo,and blastocysts in the treatment and control groups were not significantly different(P>0.05).Following vitrification,all 2-cell embryos had equal-sized blastomeres and intact zona *** aggregation toward the perinuclear region was seen in all of the treatment *** groups C and D had vacuolated mitochondria,which was reflected in the trend of preimplantation development ***:Vitamin E supplementation of 60 mg/kg body weight does not improve the viability of healthy embryos according to this *** a result,the most effective dose of vitamin E supplementation may be determined by the initial quality of the embryos.

The study aims to afford a deeper knowledge on reproductive biology of Mystus bleekeri from Surma River,Bangladesh in a multidisciplinary ***,the species is in demand as food and ornamental fish in Bangladesh but still now comprehensive details on reproductive biology of this species is *** 600 fish samples were derived from commercial catches from July 2018 to June *** differences in sex ratio,size distribution,condition factor(Kn),fecundity,gonadosomatic index and gonadal maturation cycle were *** results showed 384(64%)female and 216(36%)male with an overall sex ratio of 1.77:1(female:male).Mean total length(TL)and body weight(BW)of all fish studied were 14.85±3.38 cm TL and 27.54±15.76 g,*** Kn varied with length groups,the highest(1.45±0.23)and lowest(0.89±0.18)values obtained in 16.0-16.9 and 10.0-10.9 cm TL groups,*** highest Kn was documented in October while the lowest was in *** GSI values showed two peaks in July and November for both the *** fecundity was found to vary from 1120 to 14790 eggs with average value of 4968±3047 with 14.4-18.3 cm in TL and 19.97-34.7g in *** relationship of fecundity with TL and BW was F=1.385TL13.405,r^(2)=0.885 and F=5.201BW^(4.827),r^(2)=*** behavior of spawning together with allochronic oocytes development has also been found through *** results of this study could be used for predicting the response of populations of fish in Surma River and/or other where to human interferences and environmental change.

The Evolution of Iron-Related Comorbidities and Hospitalization in Patients with Hemochromatosis:An Analysis of the Nationwide Inpatient Sample

Mental health disorders in child and adolescent survivors of post-war landmine explosions

Background: To describe the mental health status of 78 child and adolescent survivors of post-war landmine ***: Child and adolescent survivors of landmine explosions who were younger than 18 years old at the time of the study were identified and enrolled in this study. The mental health status of the participants was assessed by general health assessment and psychiatric examinations. Psychiatric assessment and diagnosis were undertaken using the Diagnostic and Statistical Manual for mental disorders(DSM-IV) criteria. A psychiatrist visited and interviewed each survivor and identified psychiatric ***: Seventy-eight child and adolescent survivors with a mean age of 16.11±2 years old were identified and agreed to participate in the study. The mean age of the victims at the time of injury was 8.2±3.12 years old(range 2–15). Thirty-seven(47.4%) of the adolescent survivors suffered from at least one psychiatric disorder. Twenty-nine survivors(37.1%) were newly diagnosed and needed to start medication and psychiatric treatment. The most common findings were anxiety disorders(34.6%), including posttraumatic stress disorder(PTSD) in 20(25.6%), and generalized anxiety disorder(GAD) in 7(9%) subjects. Mild-Moderate depression was found in 5(6.4%) subjects. No personality disorders were observed, and two patients suffered from mental retardation. The study results revealed a significant association between age of casualty, duration of injury and limb amputation, and types of psychological ***: Child and adolescent survivors of landmine explosions had a high prevalence of psychiatric disorders.

Objective: To evaluate the impact of Olea (O.) europaea extract on markers of oxidative stress and apoptosis of ovarian tissues in a rat model of torsion/detorsion-induced ovarian damage. Methods: A total of 28 Wistar female rats were randomly assigned into 4 groups, with 7 rats in each group. The sham group received a 2.5 cm longitudinal incision in the midline part of the abdomen which was then sutured with 5-0 nylon thread;the torsion/detorsion group underwent torsion induction for 3 h followed by reperfusion for 10 days;the torsion/detorsion+O. europaea group received 300 mg/kg hydro-alcoholic extract of O. europaea 30 min before detorsion, followed by reperfusion for 10 days;and the O. europaea group only received 300 mg/kg hydro-alcoholic extract of O. europaea for 10 days. After the treatment period, blood samples were taken;the levels of estrogen, glutathione peroxidase, superoxide dismutase, and malondialdehyde were assayed. The histological changes, as well as the rate of apoptosis in ovarian tissues, were also carried out by histomorphometric analysis at day 10 post-procedure. Results: Histological comparisons demonstrated a significant detrimental change in the torsion/detorsion group as compared with other groups. The number of pre-antral and antral follicles and corpus luteum was significantly decreased in the torsion/detorsion group compared with the sham group, while treatment with O. europaea could enhance their numbers (P<0.05). The index of apoptosis and the number of atretic body in the ovarian tissue were significantly higher in the torsion/detorsion group compared with the sham group (P<0.05). The concentrations of glutathione peroxidase, estrogen, and superoxide dismutase as well as the mRNA expression of Bcl-2 were considerably diminished in the torsion/detorsion group while they were elevated in the torsion/detorsion+O. europaea group (P<0.05) compared with the torsion/detorsion group. Co-encapsulation of curcumin and doxorubicin in albumin nanoparticles blocks the adaptive treatment tolerance of cancer cells

The adaptive treatment tolerance(ATT) of cancer cells is the main encumbrance to cancer chemotherapy.A potential solution to this problem is to treat cancer cells with multiple drugs using nanoparticles(NPs).In this study, we tested the co-administration of curcumin(Cur) and doxorubicin(Dox) to MCF-7 resistant breast cancer cells to block the ATT and elicit efficient cell killing. Drugs were co-administered to cells both sequentially and simultaneously. Sequential drug co-administration was carried out by pre-treating the cells with albumin nanoparticles(ANPs) loaded with Cur(Cur@ANPs) followed by treatment with Dox-loaded ANPs(Dox@ANPs). Simultaneous drug co-administration was carried out by treating the cells with ANPs loaded with both the drugs(Cur/Dox@ANPs). We found that the simultaneous drug co-administration led to a greater intra-cellular accumulation of Dox and cell killing with respect to the sequential drug co-administration. However, the simultaneous drug co-administration led to a lower intracellular accumulation of Cur with respect to the sequential drug co-administration. We showed that this result was due to the aggregation and entrapment of Cur in the lysosomes as soon as it was released from Cur@ANPs, a phenomenon called lysosomotropism. In contrast, the simultaneous release of Dox and Cur from Cur/Dox@ANPs into the lysosomes led to lysosomal p H elevation, which, in turn, avoided Cur aggregation, led to lysosome swelling and drug release in the cytosol, and finally provoked efficient cell killing. Our study shed the light on the molecular processes driving the therapeutic effects of anti-cancer drugs co-administered to cancer cells in different manners.
