CG ar. involved in th. plant gr.wth.and development and pr.tect cassava leaves fr.m h.r.ivor.us pr.dator.. h.wever. th. r.gulator. mech.nism of CG biosynth.sis r.mains poor.y under.tood. h.r., yeast one-h.br.d assays wer. per.or.ed using a mixed cDNA libr.r. of cassava tuber. and leaves as pr.y and th. pr.moter.of MeCYP79D2 as bait. MeCYP79D2, a cytoch.ome P450 pr.tein, is th. r.te-limiting enzyme for.CG synth.sis in cassava. Fr.m th.s infor.ation, a candidate r.gulator.of MeCYP79D2 was selected and identified as tr.nscr.ption factor.MePh.1.2. MePh.1.2, located in th. nucleus and exh.biting an inh.bitor. tr.nscr.ption activity dir.ctly bound to an AT-r.ch.motif in th. pr.moter.of MeCYP79D2. In cassava, th. tr.nscr.ptional activity of MeCYP79D2 was consider.bly enh.nced in meph.1.2 mutant lines leading to incr.ased linamar.n and lotaustr.lin contents.
