Correlation of p53 over-expression and alteration in p53 gene detected by polymerase chain reaction-single strand conformation polymorphism in adenocarcinoma of gastric cancer patients from India

AIM： To study the alterations in p53 gene among Indian gastric cancer patients and to correlate them with the various clinicopathological parameters. METHODS： A total of 103 gastric cancer patients were included in this study. The p53 alterations were studied by both immunohistochemical method as well as polymerase chain reaction （PCR）-single strand conformation polymorphism （SSCP） analysis. We only studied four （exon 5, 6, 7, and 8） of the 11 ,p53 exons. The alterations in p53 were also correlated with respect to various clinicopathological parameters. RESULTS： Among 103 cases, p53 over-expression and alteration were detected in 37 （35.92%） and 19 （18.44%） cases, respectively. Most of the ,p53 alterations were found at exon 5 （31.54%）, followed by exon 6 （26.31%）, exon 7 （21.04%） and exon 8 （21.04%）. A significant correlation of p53 overexpression was found with p53 alteration （P = 0.000）. Concordance between ,p53 alteration （as detected by SSCP） and over-expression [as detected by immunohistochemistry （IHC）] was found in 75% cases. We found that IHC-positive/SSCP-negative cases accounted for 21% of cases and IHC-negative/SSCP- positive cases accounted for remaining 4% cases. CONCLUSION： Our results show that p53 gene mutations are significantly correlated with p53 protein over-expression, with 75% concordance in over-expression and alteration in the p53 gene, but 25% disconcordance also cautions against the assumption that p53 over-expression is always associated with a gene mutation. There may be other mechanisms responsible for stabilization and accumulation of p53 protein with no evidence of gene mutation that reflect an accumulation of a non-mutated protein, or a false negative SSCP result.

Value of multi-disciplinary input into laparoscopic management of rectal cancer-An observational study

AIMTo assess the impact of multi-disciplinary teams (MDTs) management in optimising the outcome for rectal cancers. METHODSWe undertook a retrospective review of a prospectively maintained database of patients with rectal cancers (defined as tumours ≤ 15 cm from anal verge) discussed at our MDT between Jan 2008 and Jan 2011. The data was validated against the national database to ensure completeness of dataset. The clinical course and follow-up data was validated using the institution’s electronic patient records. The data was analysed in terms of frequencies and percentages. Significance of any differences were analysed using χ2 test. A Kaplan-Meier analysis was performed for overall survival and disease free survival. RESULTSFollowing appropriate staging, one hundred and thirty-three patients were suitable for potentially curative resections. Seventy two (54%) were upper rectal cancer (URC) - tumour was > 6 cm from the anal verge and 61 (46%) were lower rectal cancers (LRC) - lower extent of the tumour was palpable ≤ 6 cm. Circumferential resection margin (CRM) appeared threatened on pre-operative MRI in 19/61 (31%) patients with LRC requiring neo-adjuvant therapy (NAT). Of the 133 resections, 118 (89%) were attempted laparoscopically (5% conversion rate). CRM was positive in 9 (6.7%) patients; Median lymph node harvest was 12 (2-37). Major complications occurred in 8 (6%) patients. Median follow-up was 53 mo (0-82). The 90-d mortality was 2 (1.5%). Over the follow-up period, disease related mortality was 11 (8.2%) and overall mortality was 39 (29.3%). Four (3%) patients had local recurrence and 22 (16.5%) patients had distant metastases. CONCLUSIONManagement of rectal cancers can be optimized with multi-disciplinary input to attain acceptable long-term oncological outcomes even when incorporating a laparoscopic approach to rectal cancer resection.

Evaluation of soil loss severity and ecological restoration approach for sustainable agriculture in the Hindu Kush, Karakoram and Himalaya region

Soil erosion has resulted in removal of the topsoils containing fine soil particles and plant nutrients, causing decrease in soil fertility in the Hindu Kush, Karakoram and Himalaya(HKH) region. The existing production of cereal crop grains has been reduced to one third of the potential crop grains production owing to land degradation and poor farming practices. It is necessary to assess risk of soil loss and identify appropriate controlling measures to address issues of low agriculture productivity and water insecurity in the region. In the present study, severity of soil loss was predicted using Revised Universal Loss Equation(RUSLE) and ecological measures were identified for sustainable mountain agriculture in the HKH region of Upper Indus Basin, Pakistan. Overall 62.6% area was found to have very low risk of soil loss, i.e., 100 t/(ha·yr) in about 6.8% and 7.4% areas respectively. The mean rate of soil loss was about 41.9 t/(ha·yr) in the Hindu Kush, 31.1 t/(ha·yr) in the Himalayas, 18.8 t/(ha·yr) in the Karakoram and overall 29.7 t/(ha·yr) in the three HKH ranges. As such no considerable measures have been adopted by the communities for restoration of the degraded areas except raising fruit/farm trees and supporting limited social forestry for their livelihoods. The slopes cleared for cultivation and susceptible to erosion may be stabilized through sowing/planting of multi-purpose plant species and formation of proper bench terraces. The conservation of forest ecosystem and pastures at higher elevations would help in reducing overland water flow, risk of flash flood hazard and minimizing sediment loads in the downstream. Structural behavior of intermediate length cold-formed steel rack columns with C-stitches

This article presents an experimental and numerical investigation on the strength and performance of intermediate length rack column sections with C-stitches under axial compression. The test program consisted of 10 axial concentric compression tests on columns with and without C-stitches under pin end conditions for two different geometric lengths. Finite element (FE) models were developed using commercial FE package ABAQUS considering material and geometric nonlinearities as well as initial geometric imperfections. The elastic buckling properties of the section were calculated using readily available linear elastic buckling analysis tools based on Generalized Beam Theory (GBT) and Finite Strip Method (FSM). Obtained FE results were compared with those obtained experimentally, and once verified the developed FE modeling technique was used to carry out a parametric study to examine changes in structural response due to variations in length, depth and spacing of C-stitches. Observed influences of C-stitches on the behavior and resistance of the considered columns were carefully analyzed, and key design aspects are presented herein.

The Study of Thermal Mass as a Passive Design Technique for Building Comfort and Energy Efficiency

The day/night （diurnal） changes in temperature and solar radiation pose challenges for maintaining human thermal comfort in buildings. Passive and energy-conserving buildings seek to manage the available thermal energy by lowering peaks and dampening the fluctuations in order to maintain conditions for human comfort. Appropriate use of thermal mass moderates the internal temperatures by averaging diurnal extremes. Thermal mass is one of the powerful tools which architects and designers can use to control temperature. It can be used to optimize the performance of energy-conserving buildings that rely primarily on mechanical heating and cooling strategies. Massive building envelopes-such as masonry, concrete, earth, and insulating concrete forms （ICFs） can be utilized as one of the simplest ways of reducing building heating and cooling loads. This article analyses the role and effectiveness of thermal mass as a strategy for providing indoor thermal comfort for passive solar and energy conserving buildings.

Presowing Seed Treatment with Cytokinins and Its Effect on Growth, Photosynthetic Rate, Ionic Levels and Yield of Two Wheat Cultivars Differing in Salt Tolerance

The effects of presowing seed treatment with different concentrations of cytokinins （kinetin and benzylaminopurine; 100, 150, and 200 mg/L） on growth, photosynthetic capacity, and ion homeostasis were investigated in two spring wheat （Triticum aestivum L.） cultivars, namely MH-97 （salt sensitive） and Inqlab- 91 （salt tolerant）. Primed and non-primed seeds were sown in a field in which NaC1 salinity of 15 dS/m was developed. Of the different concentrations of priming agents tested, the effect of a moderate concentration of kinetin （150 mg/L） was very pronounced, particularly in improving growth and grain yield, in both cultivars. In addition, priming with kinetin alleviated the adverse effect of salt stress on gaseous exchange characteristics （net CO2 assimilation rate and water use efficiency） in both cultivars. Seed priming with a moderate concentration of kinetin also altered the pattern of accumulation of Na^＋ and Clˉ in the shoots, irrespective of the wheat cultivar, under conditions of salt stress. However, all other priming agents at the different concentrations tested did not show any consistent effect on ion levels, except hydropriming, which increased K^＋ levels in the shoots of both cultivars under salt stress. In conclusion, a moderate concentration of kinetin showed a consistent effect in altering the growth and grain yield of both wheat cultivars, which was related to the beneficial effects of kinetin priming on water use efficiency and photosynthetic rate under conditions of salt stress.

Wetlands are those landscapes which are saturated with water or covered by water either perennially or for a major part of the year. Due to transforming nature of the wetlands from aquatic to terrestrial, the related physical features are not easy to be monitored. With the recent advancement in Remote sensing technique, the feature extraction of wetland with the help of different satellite derived band algebras including Normalized Difference Vegetation Index (NDVI), Normalized Difference Water Index (NDWI), Modified NDWI (MNDWI), Normalized Difference Moisture Index (NDMI) is being used by experts. The common diagnostic features of wetlands are surface water, swamps and aquatic vegetation. The present study is based on a comparison between these four indices. Baraila wetland of Vaishali district Bihar is selected as a site for the study because the surface water of this “BAT” shaped wetland has decreased rapidly in last half decade which is alarming for the related ecology and biodiversity.

Seed Treatment with Auxins Modulates Growth and Ion Partitioning in Salt-stressed Wheat Plants

Experiments were performed to determine whether seed priming with different concentrations （100, 150, and 200 mg/L） of auxins （indoleacetic acid （IAA）, indolebutyric acid （IBA）, or their precursor tryptophane （Trp）） could alter salinity induced perturbances in salicylic acid and ion concentrations and, hence, growth in wheat （Triticum aestivum L.） cultivars, namely M.H.-97 （salt intolerant） and tnqtab-91 （salt tolerant）. Primed and non-primed seeds were sown in Petri dishes in a growth room, as well as in a field treated with 15 dS/m NaCl salinity. All priming agents, except IBA, increased the final germination percentage in both cultivars. The seedlings of either cultivar raised from Trp-treated seeds had greater dry biomass when under salt stress. In field experiments, Trp priming was much more effective in mediating the increase in grain yield, irrespective of the cultivar, under salt stress. The alleviatory effect of Trp was found to be associated with reduced uptake of Na^＋ in the roots and subsequent translocation to the shoots, as well as increased partitioning of Ca^＋ in the roots of salt-stressed wheat plants. Plants of both cultivars raised from Trp-and IAA-treated seeds accumulated free salicylic acid in their leaves when under salt stress. Overall, the Trp priming-induced improvement in germination and the subsequent growth of wheat plants could be related to ion homeostasis when under salt stress. The possible involvement of salicylic acid in the Trp priming-induced better growth under Conditions of salt stress is discussed.

Evaluation of glacial resource potential for sustaining kuhl irrigation system under changing climate in the Himalayan region

A large numbers of mountain communities in the Himalayan region depend on glacier-fed kuhl system – gravity flow irrigation, for their sustenance. The depletion and retreat of glaciers often result in disruption of melt flows to kuhl system, which ultimately impacts the agriculture as well as livelihood of the local communities. Sustainability of glacial resource was assessed for establishing kuhl irrigation system based on indices of glacier decrease by number(GDN) and glacier depletion by volume(GDV) during the 2001-2013 period. The selected factors are important in terms of understanding point source and reserve potential of glaciers for sustaining glacier-fed kuhl irrigation in this region. The sustainability of glaciers was found good(i.e. a least decrease or rather increase in glacier number and volume) in about 23.6% glaciers(ice reserves about 705.9 km^3) under >10 km^2 area class of glaciers mainly in Gilgit and Shigar river basins of the Karakoram range. In the 5–10 km^2 class of glaciers, sustainability was good in 49 glaciers(ice reserve 26.59 km^3) and under 1–5 km^2 class, in 558 glaciers(ice reserve 60.189 km^3). Overall sustainability of glaciers appears to prevail in the Karakoram basins. The lesser sustainability of glaciers in the Himalaya basins like Astore and Jhelum would apparently be compensated by monsoon and change in rainfall pattern. A regular monitoring of the glacier resource and utilizing advance water conservation techniques would help in coping with negative impacts of downwasting of glaciers and provide long-term support to kuhl irrigation system in the Himalayan region in future.
