Roscovitine synergizes with conventional chemo-therapeutic drugs to induce efficient apoptosis of human colorectal cancer cells

AiM: To examine the ability of cyclin-dependent kinase inhibitor (CDKi) roscovitine (Rosco) to enhance the antitumor effects of conventional chemotherapeutic agents acting by different mechanisms against human colorectal cancer. METHODS: Human colorectal cancer cells were treat-ed, individually and in combination, with Rosco, taxol, 5-Fluorouracil (5-FU), doxorubicine or vinblastine. The antiproliferative effects and the type of interaction of Rosco with tested chemotherapeutic drugs were de-termined. Cell cycle alterations were investigated by fluorescence-activated cell sorter FACS analysis. Apop-tosis was determined by DNA fragmentation assay. RESULTS: Rosco inhibited the proliferation of tumor cells in a time-and dose-dependent manner. The ef-ficacies of all tested chemotherapeutic drugs were markedly enhanced 3.0-8.42 × 103 and 130-5.28 × 103 fold in combination with 5 and 10 μg/mL Rosco, re-spectively. The combination of Rosco and chemothera-peutic drugs inhibited the growth of human colorectal cancer cells in an additive or synergistic fashion, and in a time and dose dependent manner. Rosco induced apoptosis and synergized with tested chemothera-peutic drugs to induce efficient apoptosis in human colorectal cancer cells. Sequential, inverted sequential and simultaneous treatment of cancer cells with combi-nations of chemotherapeutic drugs and Rosco arrested the growth of human colorectal cancer cells at various phases of the cell cycle as follows: Taxol/Rosco (G2/M-and S-phases), 5-FU/Rosco (S-phase), Dox/Rosco (S-phase) and Vinb/Rosco (G2/M-and S-phases). CONCLUSiON: Since the eff icacy of many anticancer drugs depends on their ability to induce apoptotic cell death, modulation of this parameter by cell cycle inhibi-tors may provide a novel chemo-preventive and chemo-therapeutic strategy for human colorectal cancer.

Price prediction of goods is a vital point of research due to how common e-commerce platforms *** are several efforts conducted to forecast the price of items using classicmachine learning algorithms and *** models can predict prices of various financial instruments,e.g.,gold,oil,cryptocurrencies,stocks,and second-hand *** these efforts,the literature has no model for predicting the prices of seasonal goods(e.g.,Christmas gifts).in this context,we framed the task of seasonal goods price prediction as a regression ***,we utilized a real online trailer dataset of Christmas gifts and then we proposed several machine learningbased models and one statistical-based model to predict the prices of these seasonal ***,we utilized a real-life dataset of Christmas gifts for the prediction ***,we proposed support vector regressor(SVR),linear regression,random forest,and ridgemodels as machine learningmodels for price ***,we proposed an autoregressive-integrated-movingaverage(ARiMA)model for the same purpose as a statistical-based ***,we evaluated the performance of the proposed models;the comparison shows that the best performing model was the random forest model,followed by the ARiMA model.

Delayed Grafting as a Valid Technique in Burn Management: Experience from a Burn Unit in Oman

Background: Early excision and grafting has been the preferred method of managing major burns around the world since 1970. Considering the advances in health care and the development of new antibiotics over the past 50 years, delayed grafting as a technique for the management of burns over 15%-20% of total body surface area (TBSA) could have comparable results to that of early excision. This study aims to highlight the outcomes of practicing delayed grafting in burn patients. Methods: A case series analysis was performed of 51 patients who were admitted to the burns unit in Sultan Qaboos Hospital Salalah with over 20% TBSA between January 2014 and December 2019. The patients received prophylactic antibiotics and silver sulphadiazine dressing until the burn eschar had completely separated, followed by grafting. Results: Two patients were lost during the entire duration of the study. The mortality rate was comparable to that of early excision, while the rate of hypertrophic scarring was lower than the range reported by other studies. Conclusion: in the management of patients with over 20% TBSA, delayed grafting after complete separation of eschar is still a valid technique.

Analysis and Characterization of Pore System and Grain Sizes of Carbonate Rocks from Southern Lebanon

Carbonate rocks are common in many parts of the world, including the Eastern Mediterranean, where they host significant groundwater supplies and are widely used as building and ornamental stones. The porosity of carbonate rocks plays a critical role in fluid storage and retrieval. The pore structure connectivity, in particular, controls many properties of geological formations, as well as the relationships between the properties of individual minerals and the bulk properties of the rock. To study the relationships between porosity, rock properties, pore structure, pore size, and their impact on reservoir characteristics, 46 carbonate rock samples were collected from four stratigraphic sections exposed near Sidon, South Lebanon. The studied carbonate rocks are related to marine deposits of different ages(e.g., Upper Cretaceous, Eocene, and Upper Miocene). in order to understand the pore connectivity, the MiCP(mercury injection capillary pressure) technique was conducted on ten representative samples. Results from the SEM analysis indicate the dominance of very fine and fine pore sizes, with various categories ranging in diameter from 0.1 to10 μm. The MiCP data revealed that the pore throat radii vary widely from 0.001 to 1.4 μm, and that all samples are dominated by micropore throats. The grain size analysis indicated that the studied rocks have significant amounts of silt-and clay-size grains with respect to the coarser ‘sand-size’ particles, suggesting a high proportion of microporosity. Obtained results, such as the poorly-sorted nature of grains, high microporosity, and the high percentage of micropore throats, justify the observed low mean hydraulic radius, the high entry pressure, and the very low permeability of the studied samples. These results suggest that the carbonate rocks near Sidon(south of Lebanon) are possibly classified as non-reservoir facies.

The energy market in Libya is expected to face substantial changes in the next few years:electrical energy consumption will increase by 50% within the next 4 ***,there is a plan to gradually increase renewable energy sources in the power network by 2030 to 30%.Solar photovoltaic(PV)plants will play a significant role in the energy transition and the mix of energy sources in *** article is a study conducted to investigate the challenges of power-flow management and power protection from integrating PV power plants into the Libyan power *** particular,a simulation model is built for the Kufra PV power plant(10 MW)with eight buses to assess the power network performance in terms of power quality such as voltage profile,power losses and ***,the impact of the PV plant on the short-circuit level and the power-protection system is presented under different operating *** fault ride-through(FRT)is operated using standard ambient temperature and a wide range of irradiation intensity,and is verified by using the Libyan grid *** results show that the integration of the PV plant into the grid has a significant impact on the short-circuit level and the FRT over the different fault levels and locations on the *** initial period of the three-phase fault shows that the PV plant will be disconnected from the grid due to a decrease in the root mean square value voltages where the reactive and active power of the grid reach 938 MVAR and 261 MW,*** results show that increasing the fault level will lead to an increase in injecting the active *** work is considered the first extensive investigation into the challenges of modern power-flow management and power protection for the power network system in Libya.
