SPEcT-computed tomography in rats with TNBS-induced colitis: A first step toward functional imaging

AIMTo assess the feasibility of SPEcT-computed tomography (cT) in rats with trinitrobenzene sulfonic acid (TNBS)-induced acute colitis and confront it with model inflammatory *** was induced in Sprague-Dawley rats by intrarectal injection of TNBS (n = 10) while controls received vehicle (n = 10). SPEcT-cT with intravenous injection of 10 MBq of 67Ga-citrate was performed at day 2. SPEcT-cT criteria were colon wall thickness and maximal wall signal intensity. Laboratory parameters were assessed: colon weight:length ratio, colon cyclooxygenase-2 expression by western blot and histological inflammatory *** weight/length ratio, colon cOX-2 expression and histological inflammatory score were significantly higher in the TNBS group than in the control group (P = 0.0296, P < 0.0001, P = 0.0007 respectively). Pixel max tend to be higher in the TNBS group than in the control group but did not reach statistical significance (P = 0.0662). Maximal thickness is significantly increased in the TNBS group compared to the control group (P = 0.0016) while colon diameter is not (P = 0.1904). Maximal thickness and colon diameter were correlated to colon cOX-2 expression (P = 0.0093, P = 0.009 respectively) while pixel max was not (P = 0.22). Maximal thickness was significantly increased when inflammation was histologically observed (P = 0.0043) while pixel max and colon diameter did not (P = 0.2452, P = 0.3541, respectively).cONcLUSIONSPEcT-cT is feasible and easily distinguished control from colitic rats.

Dissipation of chlorpyrifos, pendimethalin, and thiophanate-methyl pesticides in compost based-versus peat-moss based-biomixture of biobeds

Biobed is a smart bioremediation system used to treat point-source pesticide *** is the main component of biobeds,and pesticide dissipation is affected by its *** study aimed to compare the effectiveness of compost-based(c)versus peat-moss-based(P)biomixtures of biobeds on tested pesticide *** concentrations(25,50,and 75 mg/kg)of chlorpyrifos,pendimethalin,and thiophanate methyl were added separately and as a mix to both biomixtures c and *** data showed the effect of biomixture type on the dissipation of the tested *** chlorpyrifos,its dissipation rate in biomixture P was more rapid than that in biomixture *** result was confirmed by the mineralization kinetic experiment,since 25%of the initial ^(14)c-chlorpyrifos concentration accumulated in the form ^(14)cO_(2) in biomixture P compared to only 14%in biomixture *** addition,the chlorpyrifos dissipation rate was influenced by the initial concentration when applied individually in biomixture *** contrast,biomixture c was more effective at pendimethalin dissipation than biomixture P,since>76%of pendimethalin was dissipated in biomixture c versus 67%in biomixture P at the same incubation *** thiophanate-methyl,the abilities of both biomixtures c and P were similar and less efficient than those of the other tested *** addition of the three tested pesticides to biomixture c only had a positive effect on both chlorpyrifos and thiophanate-methyl dissipation,while pendimethalin dissipation was similar when applied separately or as a *** activity was stimulated by the addition of separately or mixed pesticides to biomixtures c and P as measured by dehydrogenase activity.

Improvement of quantitative testing of liver function in patients with chronic hepatitis c after installment of antiviral therapy

MM： To investigate if and to what extent antiviral therapy influenced a broad panel of quantitative testing of liver function （QTLF）. METHODS： Fifty patients with chronic hepatitis c were either treated with interferon （n = 8）, interferon/ribavirin （n = 19） or peg-interferon/ribavirin （n = 23）. Quantitative testing of liver function, including aminopyrine breath test （ABT）, galactose elimination capacity （GEc）, sorbitol clearance （ScI） and indocyanine green clearance （IcG） was performed before and 3 mo after initiation of antiviral therapy. RESULTS： After 3 mo of antiviral treatment, 36 patients showed normal transaminases and were negative for HcVRNA, 14 patients did not respond to therapy. ABT and GEc as parameters of microsomal and cytosolic liver function were reduced in all patients before therapy initiation and returned to normal values in the 36 therapy responders after 3 too. Parameters of liver perfusion （Scl and IcG） were not affected by antiviral therapy. In the 14 non-responders, no changes in QTLF values were observed during the treatment period. cONcLUSION： IcG and ScI remained unaffected in patients with chronic hepatitis c, while ABT and GEc were significantly compromised. ABT and GEc normalized in responders to antiviral therapy. Early determination of ABT and GEc may differentiate responders from non-responders to antiviral treatment in hepatitis c.

While wing form is known to differ between males and females of the genus culicoides,detailed studies of sexual dimorphism are *** this study,we analyze sex-specific differences in the wing form of 5 species of the subgenus Avaritia,using geometric morphometrics and comparative phylogenetic *** results confirm the existence of marked sexual dimorphism in the wing form of the studied species and reveal for the first time that while there is a shared general pattern of sexual shape dimorphism within the subgenus,sexual size dimorphism,and particular features of sexual shape dimorphism differ among *** shape dimorphism was found to be poorly associated to size and the evolutionary history of the *** tight association of sexual shape dimorphism with aspect ratio suggests that the shape of the wing is optimized for the type of flight of each sex,that is,dispersal flight in females versus aerobatic flight in ***,the fact that interspecific shape differences are greater and more strongly associated to aspect ratio in males than in females might be indicating that in males the selective pressures affecting flight performance characteristics are more heterogeneous and/or stronger than in females among the studied species.

A 3D Human Liver Model of Nonalcoholic Steatohepatitis

Background and Aims: To better understand nonalcoholic steatohepatitis (NASH) disease progression and to evaluate drug targets and compound activity, we undertook the devel-opment of an in vitro 3D model to mimic liver architecture and the NASH environment. Methods:We have developed an in vitro preclinical 3D NASH model by coculturing primary hu-man hepatocytes, human stellate cells, liver endothelial cells and Kupffer cells embedded in a hydrogel of rat collagen on a 96-well plate. A NASH-like environment was induced by ad-dition of medium containing free fatty acids and tumor ne-crosis factor-α. This model was then characterized by biochemical, imaging and transcriptomics analyses. Results:We succeeded in defining suitable culture conditions to main-tain the 3D coculture for up to 10 days in vitro, with the lowest level of steatosis and reproducible low level of inflammation and fibrosis. NASH disease was induced with a custom me-dium mimicking NASH features. The cell model exhibited the key NASH disease phenotypes of hepatocyte injury, steatosis, inflammation, and fibrosis. Hepatocyte injury was highlighted by a decrease of cYP3A4 expression and activity, without loss of viability up to day 10. Moreover, the model was able to stimulate a stable inflammatory and early fibrotic environ-ment, with expression and secretion of several cytokines. A global gene expression analysis confirmed the NASH induc-tion. conclusions:This is a new in vitro model of NASH dis-ease consisting of four human primary cell-types that exhibits most features of the disease. The 10-day cell viability and cost effectiveness of the model make it suitable for medium throughput drug screening and provide attractive avenues to better understand disease physiology and to identify and characterize new drug targets.

Added value of molecular karyotype in childhood acute lymphoblastic leukemia

Background:Thanks to an improved therapeutic regimen in childhood B-cell precursor acute lymphoblastic leukemia(BcP-ALL),5 year-overall survival now exceeds 90%.Unfortunately,the 25%of children who relapse have an initial poor prognosis,potentially driven by pre-existing or emerging molecular *** latter are initially and essentially identified by ***,some subtle alterations are not visible through ***:Single nucleotide polymorphisms(SNP)array is an alternative way of chromosomal analysis allowing for a more in-depth evaluation of chromosomal modifications such as the assessment of copy number alterations(cNA)and loss of heterozygosity(LOH).This method was applied here in retrospective diagnosis/relapse paired samples from seven children with BcP-ALL and in a prospective cohort of 38 newly diagnosed childhood ***:In the matched study,compared to the initial karyotype,SNP array analysis reclassified two patients as poor prognosis *** during relapse was seen for 4 cNA and 0.9 *** the prospective study,SNP reclassified the 10 patients with intermediate karyotype as 7 good prognosis and 3 poor ***,in all the children tested,SNP array allowed to identify additional anomalies compared to conventional karyotype,refine its prognostic value and identify some druggable anomalies that could be used for precision ***,the anomalies detected could be segregated in four groups respectively involved in B-cell development,cell proliferation,transcription and molecular ***:SNP therefore appears to be a method of choice in the integrated diagnosis of BcP ALL,especially for patients initially classified as intermediate *** complementary method of both cytogenetics and high throughput sequencing allows to obtain further classified information and can be useful in case of failure of these techniques.

characterization of a Novel <i>Pseudomonas stutzeri</i>Lipase/Esterase with Potential Application in the Production of chiral Secondary Alcohols

The search for new enzymes is facilitated by the rapidly growing number of genome sequences from different organisms. However, the discovery of functional proteins is still time intensive and complex alignments have to be performed. Herein, a genome database search identified a new, until now undescribed, putative lipase from Pseudomonas stutzeri strain A1501. The gene was cloned and expressed as functional protein in E. coli. A biochemical characterization provided an indication that the enzyme could be classified as an esterase with an alkaline pH optimum and a temperature optimum at 50°c. The enzyme was able to perform the kinetic resolution of racemic esters and could therefore be an interesting candidate for chiral synthesis.

The photorespiratory pathway or photorespiration is an essential process in oxygenic photosynthetic or- ganisms, which can reduce the efficiency of photosynthetic carbon assimilation and is hence frequently considered as a wasteful process. By comparing the response of the wild-type plants and mutants impaired in photorespiration to a shift in ambient c02 concentrations, we demonstrate that photorespiration also plays a beneficial role during short-term acclimation to reduced c02 availability. The wild-type plants re- sponded with few differentially expressed genes, mostly involved in drought stress, which is likely a conse- quence of enhanced opening of stomata and concomitant water loss upon a shift toward low c02. In contrast, mutants with impaired activity of photorespiratory enzymes were highly stressed and not able to adjust stomatal conductance to reduced external c02 availability. The transcriptional response of mutant plants was congruent, indicating a general reprogramming to deal with the consequences of reduced c02 availability, signaled by enhanced oxygenation of ribulose-l,5-bisphosphate and amplified by the artificially impaired photorespiratory metabolism. central in this reprogramming was the pro- nounced reallocation of resources from growth processes to stress responses. Taken together, our results indicate that unrestricted photorespiratory metabolism is a prerequisite for rapid physiological acclimation to a reduction in c02 availability.

The Deep Thioredoxome in chlamydomonas reinhardtii： New Insights into Redox Regulation

Thiol-based redox post-translational modifications have emerged as important mechanisms of signaling and regulation in all organisms, and thioredoxin plays a key role by controlling the thiol-disulfide status of target proteins. Recent redox proteomic studies revealed hundreds of proteins regulated by glutathio- nylation and nitrosylation in the unicellular green alga chlamydomonas reinhardtii, while much less is known about the thioredoxin interactome in this organism. By combining qualitative and quantitative proteomic analyses, we have comprehensively investigated the chlamydomonas thioredoxome and 1188 targets have been identified. They participate in a wide range of metabolic pathways and cellular pro- cesses. This study broadens not only the redox regulation to new enzymes involved in well-known thiore- doxin-regulated metabolic pathways but also sheds light on cellular processes for which data supporting redox regulation are scarce （aromatic amino acid biosynthesis, nuclear transport, etc）. Moreover, we char- acterized 1052 thioredoxin-dependent regulatory sites and showed that these data constitute a valuable resource for future functional studies in chlamydomonas. By comparing this thioredoxome with proteomic data for glutathionylation and nitrosylation at the protein and cysteine levels, this work confirms the existence of a complex redox regulation network in chlamydomonas and provides evidence of a tremendous selectivity of redox post-translational modifications for specific cysteine residues.
