Growth performance,oxidative stress and immune status of newly weaned pigs fed peroxidized lipids with or without supplemental vitamin E or polyphenols

Background:This study evaluated the use of dietary vitamin E and polyphenols on growth,immune and oxidative status of weaned pigs fed peroxidized lipids.A total of 192 piglets(21 days of age and body weight of 6.62±1.04 kg)were assigned within sex and weight blocks to a 2×3 factorial arrangement using 48 pens with 4 pigs per *** treatments consisted of lipid peroxidation(6%edible soybean oil or 6%peroxidized soybean oil),and antioxidant supplementation(control diet containing 33 IU/kg DL-α-tocopheryl-acetate;control with 200 IU/kg additional dl-α-tocopheryl-acetate;or control with 400 mg/kg polyphenols).Pigs were fed in 2 phases for 14 and 21 days,***:Peroxidation of oil for 12 days at 80°C with exposure to 50 L/min of air substantially increased peroxide values,anisidine value,hexanal,and 2,4-decadienal *** peroxidized lipids decreased(Pvs.18.74 kg),daily gain(473 vs.346 g/d),daily feed intake(658 vs.535 g/d)and gain:feed ratio(719 vs.647 g/kg).Lipid peroxidation decreased serum vitamin E(Pnd this decrease was larger on day 35(1.82 vs.0.81 mg/kg)than day 14(1.95 vs.1.38 mg/kg).Supplemental vitamin E,but not polyphenols,increased(P≤0.002)serum vitamin E by 84%and 22%for control and peroxidized diets,respectively(interaction,P=0.001).Serum malondialdehyde decreased(Pn on day 14,but not day 35 and protein carbonyl increased(Pn on day 35,but not day *** 8-hydroxydeoxyguanosine was not affected(P>0.05).Total antioxidant capacity decreased with peroxidation(Pnd increased with vitamin E(P=0.065)and polyphenols(P=0.046)for the control oil diet *** cytokine concentrations increased with feeding peroxidized lipids on day 35,but were not affected by antioxidant supplementation(P>0.05).Conclusion:Feeding peroxidized lipids negatively impacted growth performance and antioxidant capacity of nursery *** of vitamin E and po

AIM： To study the expression of endothelial and inducible nitric oxide synthases （enOS and inOS） and their role in inflammatory bowel disease （IBD）. METHODS： We examined the effect of sera obtained from patients with active Crohn＇s disease （CD） and ulcerative colitis （UC） on the function and viability of human umbilical vein endothelial cells （HUvEC）. HUvECs were cultured for 0-48 h in the presence of a medium containing pooled serum of healthy controls, or serum from patients with active CD or UC. Expression of enOS and inOS was visualized by immunofluorescence, and quantified by the densitometry of Western blots. Proliferation activity was assessed by computerized image analyses of Ki-67 immunoreactive cells, and also tested in the presence of the nOS inhibitor, 10^-4 mol/L L-nAME. Apoptosis and necrosis was examined by the annexin-v-biotin method and by propidium iodide staining, respectively. RESULTS： In HUvEC immediately after exposure to UC, serum enOS was markedly induced, reaching a peak at 12 h. In contrast, a decrease in enOS was observed after incubation with CD sera and the enOS level was minimal at 20 h compared to control （18%±16% vs 23%± 15% P〈0.01）. UC or CD serum caused a significant increase in inOS compared to control （UC： 300%±21%; CD： 275% ± 27% vs 108% ± 14%, P〈0.01）. Apoptosis/necrosis characteristics did not differ significantly in either experiment. Increased proliferation activity was detected in the presence of CD serum or after treatment with L-nAME. Cultures showed tube-like formations after 24 h treatment with CD serum. COnCLUSIOn： IBD sera evoked changes in the ratio of enOS/inOS, whereas did not influence the viability of HUvEC. These involved down-regulation of enOS and up-regulation of inOS simultaneously, leading to increased proliferation activity and possibly a reduced antiinflammatory protection of endothelial cells.

Postoperative complications and weight loss following jejunostomy tube feeding after total gastrectomy for advanced adenocarcinomas

Objective： Patients undergoing total gastrectomy for cancer are at risk of malnourishment. The aim of this self- controlled study was to examine the effect of jejunostomy tube feeding （JTF） and other factors on postoperative weight and the incidence of jejunostomy-related complications in patients undergoing total gastrectomy for cancer. Methods： All consecutive patients who underwent total gastrectomy for gastric cancer with jejunostomy plaeement were included from a prospective single-center database （2003-2014）. Jejunostomy-related complications and postoperative weight changes were evaluated up to 12 months after surgery. Multivariable linear regression analysis was performed to identify factors associated with weight loss 12 months after gastreetomy. Results： Of 113 patients operated in the study period, 65 received JTF after total gastrectomy for a median duration of 18 d [interquartile range （IQR）, 10-55 d]. Jejunostomy-related complieations occurred in 11 （17%） patients, including skin leakage （n=3） and peritoneal leakage （n=2）, luxation （n=3）, occlusion （n=2）, infection （n=l） and torsion （n=l）. In 2 （3%） patients, a reoperation was needed due to jejtmostomy-related complications. The mean preoperative weight of patients was 71.8 kg （100%）, and remained stable during JTF （73.9 kg, 103%, P=0.331）. After JTF was stopped, the mean weight of patients decreased to 64.9 kg （90%） at 12 months after surgery （P〈0.001）. A high preoperative body mass index （BMI）（〉_25 kg/m2） was associated with high postoperative weight loss compared to patients with a low BMI （〈25 kg/m2）（16.3% vs. 8.6%, P=0.016）. Conclusions： JTF can prevent weight loss in the early postoperative phase. However, this is at the prize of possible complications. As weight loss in the long term is not prevented, routine JTF should be re-evaluated and balanced against the selected use in preoperatively malnourished patients.
