Mechanism of Qili Qiangxin Capsule for Heart Failure Based on miR133a-Endoplasmic Reticulum Stress

Objective:To investigate the pharmacological mechanism of Qili Qiangxin Capsule(QLQX)improvement of heart failure(HF)based on miR133a-endoplasmic reticulum stress(ERS)pathway.Methods:A left coronary artery ligation-induced HF after myocardial infarction model was used in this study.Rats were randomly assigned to the sham group,the model group,the QLQX group[0.32 g/(kg·d)],and the captopril group[2.25 mg/(kg·d)],15 rats per group,followed by 4 weeks of medication.Cardiac function such as left ventricular ejection fraction(EF),fractional shortening(FS),left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),the maximal rate of increase of left ventricular pressure(+dp/dt max),and the maximal rate of decrease of left ventricular pressure(–dp/dt max)were monitored by echocardiography and hemodynamics.Hematoxylin and eosin(HE)and Masson stainings were used to visualize pathological changes in myocardial tissue.The m RNA expression of mi R133a,glucose-regulated protein78(GRP78),inositol-requiring enzyme 1(IRE1),activating transcription factor 6(ATF6),X-box binding protein1(XBP1),C/EBP homologous protein(CHOP)and Caspase 12 were detected by RT-PCR.The protein expression of GRP78,p-IRE1/IRE1ratio,cleaved-ATF6,XBP1-s(the spliced form of XBP1),CHOP and Caspase 12 were detected by Western blot.Td T-mediated d UTP nick-end labeling(TUNEL)staining was used to detect the rate of apoptosis.Results:QLQX significantly improved cardiac function as evidenced by increased EF,FS,LVSP,+dp/dt max,-dp/dt max,and decreased LVEDP(P<0.05,P<0.01).HE staining showed that QLQX ameliorated cardiac pathologic damage to some extent.Masson staining indicated that QLQX significantly reduced collagen volume fraction in myocardial tissue(P<0.01).Results from RT-PCR and Western blot showed that QLQX significantly increased the expression of mi R133a and inhibited the m RNA expressions of GRP78,IRE1,ATF6 and XBP1,as well as decreased the protein expressions of GRP78,cleaved-ATF6 and XBP1-s and decreased p-IRE1/IRE1 rati

The respiratory system's complex cellular heterogeneity presents unique challenges to researchers in this field.Although bulk RNA sequencing and single-cell RNA sequencing(scRNA-seq)have provided insights into cell types and heterogeneity in the respiratory system,the relevant specific spatial localization and cellular interactions have not been clearly elucidated.Spatial transcriptomics(ST)has filled this gap and has been widely used in respiratory studies.This review focuses on the latest iterative technology of ST in recent years,summarizing how ST can be applied to the physiological and pathological processes of the respiratory system,with emphasis on the lungs.Finally,the current challenges and potential development directions are proposed,including high-throughput full-length transcriptome,integration of multi-omics,temporal and spatial omics,bioinformatics analysis,etc.These viewpoints are expected to advance the study of systematic mechanisms,including respiratory studies.

BACKGROUND For the first time,we investigated the oncological role of plexin domain-containing 1(PLXDC1),also known as tumor endothelial marker 7(TEM7),in hepatocellular carcinoma(HCC).AIM To investigate the oncological profile of PLXDC1 in HCC.METHODS Based on The Cancer Genome Atlas database,we analyzed the expression of PLXDC1 in HCC.Using immunohistochemistry,quantitative real-time polymerase chain reaction(qRT-PCR),and Western blotting,we validated our results.The prognostic value of PLXDC1 in HCC was analyzed by assessing its correlation with clinicopathological features,such as patient survival,methylation level,tumor immune microenvironment features,and immune cell surface checkpoint expression.Finally,to assess the immune evasion potential of PLXDC1 in HCC,we used the tumor immune dysfunction and exclusion(TIDE)website and immunohistochemical staining assays.RESULTS Based on immunohistochemistry,qRT-PCR,and Western blot assays,overexpression of PLXDC1 in HCC was associated with poor prognosis.Univariate and multivariate Cox analyses indicated that PLXDC1 might be an independent prognostic factor.In HCC patients with high methylation levels,the prognosis was worse than in patients with low methylation levels.Pathway enrichment analysis of HCC tissues indicated that genes upregulated in the high-PLXDC1 subgroup were enriched in mesenchymal and immune activation signaling,and TIDE assessment showed that the risk of immune evasion was significantly higher in the high-PLXDC1 subgroup compared to the low-PLXDC1 subgroup.The high-risk group had a significantly lower immune evasion rate as well as a poor prognosis,and PLXDC1-related risk scores were also associated with a poor prognosis.CONCLUSION As a result of this study analyzing PLXDC1 from multiple biological perspectives,it was revealed that it is a biomarker of poor prognosis for HCC patients,and that it plays a role in determining immune evasion status.

Qinghuang Powder(QHP),an oral arsenic,has become an effective drug in the treatment of myelodysplastic syndromes(MDS)in Xiyuan Hospital,China Academy of Chinese Medical Sciences for many years,and the action mechanism of the compound or active ingredient AsSof QHP has been elucidated.Considering the relatively safety,chemotherapy-free and convenient oral profile,QHP is widely used in the clinical treatment for MDS patients,especially for elderly patients.In this review,the authors document the efficacy and safety of oral arsenic-containing compound QHP in the treatment of MDS,with a special focus on the association of efficacy of QHP with the cytogenetics,prognostic risk,DNA methylation,gene mutation,blood arsenic concentration,mechanism of action of AsSand the countermeasures against adverse reactions of gastrointestinal tract.

Elevational distribution of forests and its spatiotemporal dynamics in subtropical China from 2000 to 2019

The subtropical region of China possesses abundant forest resources and features a mountainous terrain. Under the implementation of policies such as natural forest protection, the Grain for Green Project, and other initiatives since the beginning of the 21st century, coupled with climate change, the forest resources in this region have undergone significant changes compared to historical periods. In addition, forest resources distributing mainly in mountainous areas also implies that these changes may vary significantly with elevation. To explore the spatiotemporal dynamics of forest distribution in subtropical China since 2000,especially the trend of changes with elevation, we analyzed the data from two land cover products focusing on forest cover and forest types. We used a hierarchical approach, in which coarse-classification forest cover data with relatively small uncertainties impose constraints on forest type data with larger uncertainties, to achieve a reasonable balance between obtaining more details and reducing data uncertainty. We first divided the forest cover data into ‘unchanged' and ‘changed' categories. With the constraints by the forest cover results, we further analyzed the ‘unchanged' and ‘changed' forest types. The results indicated that,since the implementation of ecological engineering and management policies, 54% of the area in the subtropical region had maintained unchanged forest cover attributes over the past 20 years, which implied the good state of ecological environment. The results also showed that dynamic conversions existed in the long term between forests and lands for essential production needs like croplands. The elevational variations of forest cover suggested that the dominant changes came from the conversion between forests and croplands in low-elevation regions below 700 m, the conversion between forests and shrublands in mid-elevation regions of 700–1500 m, and the conversion between forests and grasslands in high-elevation regions above 2000 m. 提单管辖条款是承运人寻求对其最有利的法院和准据法以规避诉讼风险的产物。我国现行法律部分承认提单管辖条款的效力,但零散模糊,缺乏可预见性。该条款是协议管辖的特殊形式,以意思自治为基础,其法律效力的争议核心是对当事人合意的解释与限制。我国司法实践中选择域外法院的大多无效,缺乏实际联系、不符合格式条款要求、不能反映当事人合意是常见理由。我国续造提单管辖规则的路径应当是,在基本立场上平衡司法主权和国际礼让原则,确立以客观联系为基础的实际联系标准;在具体规则上对当事人意思自治予以合理尊重,参照国际公约标准明确格式条款的具体要求。纾解提单管辖理论争议,有助于发展我国协议管辖制度,并逐步协调司法实践。

Layered oxide cathodes for sodium-ion batteries:microstructure design,local chemistry and structural unit

Because of the low price and abundant reserves of sodium compared with lithium,the research of sodium-ion batteries(SIBs)in the field of large-scale energy storage has returned to the research spotlight.Layered oxides distinguish themselves from the mains cathode materials of SIBs owing to their advantages such as high specific capacity,simple synthesis route,and environmental benignity.However,the commercial development of the layered oxides is limited by sluggish kinetics,complex phase transition and poor air stability.Based on the research ideas from macro-to micro-scale,this review systematically summarizes the current optimization strategies of sodium-ion layered oxide cathodes(SLOC)from different dimensions:microstructure design,local chemistry regulation and structural unit construction.In the dimension of microstructure design,the various structures such as the microspheres,nanoplates,nanowires and exposed active facets are prepared to improve the slow kinetics and electrochemical performance.Besides,from the view of local chemistry regulation by chemical element substitution,the intrinsic electron/ion properties of SLOC have been enhanced to strengthen the structural stability.Furthermore,the optimization idea of endeavors to regulate the physical and chemical properties of cathode materials essentially is put forward from the dimension of structural unit construction.The opinions and strategies proposed in this review will provide some inspirations for the design of new SLOC in the future.

Theoretical characterization of the adsorption configuration of pyrrole on Si(100)surface by x-ray spectroscopy

The possible configurations of pyrrole absorbed on a Si(100)surface have been investigated by x-ray photoelectron spectroscopy(XPS)and near-edge x-ray absorption fine structure(NEXAFS)spectra.The C-1s XPS and NEXAFS spectra of these adsorption configurations have been calculated by using the density functional theory(DFT)method and fullcore hole(FCH)approximation to investigate the relationship between the adsorption configurations and the spectra.The result shows that the XPS and NEXAFS spectra are structurally dependent on the configurations of pyrrole absorbed on the Si(100)surface.Compared with the XPS,the NEXAFS spectra are relatively sensitive to the adsorption configurations and can accurately identify them.The NEXAFS decomposition spectra produced by non-equivalent carbon atoms have also been calculated and show that the spectral features vary with the diverse types of carbon atoms and their structural environments.

为了探索产品质量对互联网消费信贷产品扩散的影响,引入网络结构调节变量因素,根据互联网消费信贷产品的特点,构建了基于产品质量的互联网消费信贷产品扩散随机阈值模型。在无标度网络仿真环境下,从扩散速度、扩散深度和扩散规模三个维度探讨产品质量如何影响互联网消费信贷产品的扩散。研究结果表明:产品质量能够对互联网消费信贷产品扩散产生显著影响。在相同网络结构环境中,产品质量越高,则互联网消费产品的扩散速度越快、扩散深度越高、扩散规模也越大,而且较高的产品质量完全可以弥补网络结构改变带来的不利因素影响。同时,在相同产品质量的条件下,改变网络规模、网络平均度也可以显著影响互联网消费信贷产品的扩散,凸显网络结构变量对于互联网消费信贷产品扩散的重要调节作用。通过对2017-2021年我国互联网消费信贷行业数据进行实证分析,有效验证了仿真结果的有效性和科学性。本文的研究不仅可以进一步丰富和完善金融产品扩散的相关理论,也可以为金融机构推广互联网消费信贷产品提供决策参考。
