Objective The aim of the study was to establish the contemporary epidemiological characteristics of Japanese encephalitis （JE） in Guizhou Province. Methods A retrospective study of National Notifiable Disease Reporting System （NNDRS） data from 2971 through 2009, was conducted to ascertain the geographical, seasonal, and age distributions of JE incidence in Guizhou Province, China. Results A total of 68 425 JE cases were reported in Guizhou from 1971-2009. The JE cases occurred sporadically in all 9 prefectures of Guizhou, mostly among residents of rural areas. Seasonal distribution of JE remained consistent over the period from 1971-2009 with the main transmission season starting from June to September and peaking in August. JE occurred mainly in children under the age of 15 years with peak incidence in the 0-6-year age group. Pearson＇s correlation analysis showed that JE vaccine distribution had a negative correlation with JE incidence rates during 1971-2009 （coefficient of correlation=-0.475, P〈O.01）. Conclusion Over the period of 1971-2009, the JE incidence rate had declined dramatically in terms of geographical and age distributions due to JE vaccination to children at risk.

The objective of this study was to evaluate the effectiveness of Chinese massage therapy in patients with knee osteoarthritis(OA) by measuring lower-limb gait parameters. We recruited 20 women with knee OA,who then underwent Chinese massage therapy three times per week for 2 weeks. The patients underwent gait evaluation using a six-camera infrared motion analysis system. They completed Western Ontario and Mc Master Universities Osteoarthritis Index questionnaires before and after treatment. We calculated the forward speed,step width,step length,total support time percentage,initial double support time percentage,and single support time percentage. We also measured the angles at the knee,hip,and ankle during the stance phase of walking. The results showed statistically significant mean differences in knee pain relief,alleviation of stiffness,and physical function enhancement after therapy(P < 0.05). The patients gained significantly faster gait speed,greater step width,and increased total support time percentage after the Chinese massage therapy(P < 0.05). There were no significant differences in the range of motion or initial contact angles of the knee,hip,or ankle during the stance phase of walking. We concluded that Chinese massage is a beneficial complementary treatment and an alternative therapy choice for patients with knee OA for short-term pain relief. Chinese massage may improve walking ability for these patients.

HIV-specific chimeric antigen receptor(CAR)T-cells have been developed to target HIV-1 infected CD4t T-cells that express HIV Env ***,T cell exhaustion and the patient-specific autologous paradigm of CAR-T cell hurdled clinical ***,we created HIV-specific CAR-T cells using human peripheral blood mononuclear cells and a 3BNC117-E27(3BE)CAR construct that enabled the expression of programmed cell death protein(PD-1)-blocking scFv E27 and the single-chain variable fragment of the HIV-1-specific broadly neutralizing antibody 3BNC117 to target native HIV *** with T cells expressing 3BNC117-CAR alone,3BE CAR-T cells showed greater cytotoxic activity against HIV Envt cells with stronger proliferation capability,higher killing efficiency,and enhanced cytokine secretion in the presence of HIV Env-expressing ***,we manufactured TCR-deficient 3BE CAR-T cells through gene editing and demonstrated that these CAR-T cells could effectively kill HIV Env^(+) cells in vivo without the occurrence of severe graft-versus-host disease(GvHD)in NSG *** data suggest that we have provided a feasible approach to the generation of“off-theshelf”anti-HIV CAR-T cells in combination with PD-1 checkpoint blockade immunotherapy,which can be a powerful therapeutic candidate for the functional cure of HIV.

目的:观察电针“肠病方”对葡聚糖硫酸钠(DSS)诱导的急性溃疡性结肠炎(UC)大鼠肠黏膜屏障和核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎性小体的影响,探讨电针“肠病方”治疗UC的作用机制。方法:将32只SPF级健康雄性SD大鼠随机分成空白组、模型组、西药组和电针组,每组8只。除空白组外,大鼠饮用5%DSS溶液7 d制备UC模型。造模后,西药组予美沙拉嗪混悬液(200 mg/kg)灌胃;电针组予针刺双侧“天枢”“上巨虚”和“中脘”穴,同侧“天枢”与“上巨虚”连接电子针疗仪电极,予疏密波,频率10 Hz/50 Hz,每次干预20 min。两组均每日1次,干预3 d。每天观察各组大鼠一般情况。干预后,计算大鼠疾病活动指数(DAI)评分;HE染色法观察大鼠结肠组织形态;ELISA法检测血清促炎性细胞因子[白细胞介素(IL)-18、IL-1β]含量;Western blot法检测大鼠结肠组织NLRP3、凋亡相关斑点样蛋白(ASC)、半胱氨酸天冬氨酸蛋白酶-1(Caspase-1)蛋白表达;免疫荧光法检测结肠组织闭锁连接蛋白-1(ZO-1)、闭锁蛋白(Occludin)阳性表达。结果:与空白组比较,模型组大鼠一般状况较差,体质量增长缓慢,结肠长度缩短(P0.05)。与空白组比较,模型组结肠黏膜形态破坏,腺体排列紊乱,隐窝萎缩,有明显的炎性细胞浸润;与模型组比较,西药组和电针组大鼠结肠黏膜形态相对完好,腺体与隐窝结构得到了恢复与改善,炎性细胞浸润减少。结论:电针“肠病方”对DSS诱导的急性UC具有显著的改善作用,可能是通过调节NLRP3炎性小体和肠黏膜屏障相关蛋白的表达,从而减轻溃疡性结肠炎的症状。

Background Initially, Cryptococcus （C.） neoformans was previously divided into two varieties comprising C. neoformans var. neoformans and C. neoformans var. gattii. Currently, taxonomic studies defined C. neoformans as C. species complex, which contains C. neoformans var. neoformans （serotype D）, the hybrid isolates （serotype AD）, C. neoformans var. grubfi （serotype A） and C. gattii （serotypes B and C）. However, Liao and his team once isolated a unique C. gattii isolate, namely strain S8012 with unique phenotype from cerebrospinal fluid （CSF） of a 43-year-old male patient in the Shanghai Changzheng Hospital and described as C. neoformans var. shanghaiensis in 1980s. The aim of this study was to explore the genetic background and polymorphism of Chinese clinical C. gattii isolates. Methods S8012 was analyzed as representative strain using the M13-polymerase chain reaction （PC R） fingerprinting pattern and multilocus sequence analysis including internal transcribed spacers of rDNA （ITS region）, the intergenic spacer 1 regions （IGS1）, RPB1, RPB2, CNLACl, and TEF1 genes. Results The PCR fingerprinting pattern results showed strain S8012 belonged to molecular types VGI, and phylogenetic analysis suggested strain S8012 was grouped into the cluster of C. gattii environmental isolates originated from Eucalyptus camaldulensis trees in Australia. Conclusion C. gattii isolates from Chinese patients expresses high polymorphism on the phenotype, and molecular type VGI isolates from China have a close genetic relationship with the C. gattii isolates from Australia.

Objective To study the transfer of paralytic shellfish toxins （PST） using four simulated marine food chains： dinoflagellate Alexandrium tamarense→Artemia Artemia salina→Mysid shrimp Neomysis awatschensis; A. tamarense→N. awatschensis; A. tamarense→A, salina→Perch Lateolabrax japonicus; and A. tamarense→L, japonicus. Methods The ingestion of A. tamarense, a producer of PST, by L. japonicus, N. awatschensis, and A. salina was first confirmed by microscopic observation of A. tamarense cells in the intestine samples of the three different organisms, and by the analysis of Chl.a levels in the samples. Toxin accumulation in L. japonicus and N. awatschensis directly from the feeding on A. tamarense or indirectly through the vector of A. salina was then studied, The toxicity of samples was measured using the AOAC mouse bioassay method, and the toxin content and profile of A. tamarense were analyzed by the HPLC method. Results Both A. salina and N. awatschensis could ingest A. tamarense cells. However, the ingestion capability of A. salina exceeded that of N. awatschensis. After the exposure to the culture of A. tamarense （2 000 cells·mL^-1） for 70 minutes, the content of Chl.a in A. salina and N. awatschensis reached 0.87 and 0.024 μ***^-1, respectively. Besides, A. tamarense cells existed in the intestines of L. japonicus, N. awatschensis and A. salina by microscopic observation. Therefore, the three organisms could ingest A. tamarense cells directly. A. salina could accumulate high content of PST, and the toxicity of A. salina in samples collected on days 1, 4, and 5 of the experiment was 2.18, 2.6, and 2.1 MU.g^-1, respectively. All extracts from the samples could lead to death of tested mice within 7 minutes, and the toxin content in anemia sample collected on the 1st day was estimated to be 1.65×10 ^5 μg STX equal/individual. How supermassive black holes(SMBHs) are spun-up is a key issue in modern astrophysics. As an extension to the study in Wang et al., here we address the issue by comparing the host galaxy properties of nearby(z ming that radio emission from the jet reflects a high SMBH spin, which stems from the well-known Blandford-Znajek mechanism of jet production, high-mass SMBHs(i.e., MBH > 10^7.9 M⊙)have a preference for being spun-up in classical bulges, and low-mass SMBHs(i.e., MBH = 10^6-10^7 M⊙)in pseudo-bulges. This dichotomy suggests and confirms that high-mass and low-mass SMBHs are spun-up in different ways, i.e., a major "dry" merger and a secular evolution respectively.
