Multi-drug Resistant Uropathogenic Escherichia coli and Its Treatment by Chinese Medicine

Objectives： To investigate the resistance and virulence profiles of uropathogenic Escherichia coli（UPEC） and its treatment by Chinese medicine（CM） Fuzheng Qingre Lishi Formula（扶正清热利湿方, FQLF）. Methods： UPEC strains were isolated from recurrent urinary tract infections（UTIs） patients. Patient sensitivities to 17 antibiotics were tested by the disk diffusion method. Virulence genes were screened by plolymerase chain reaction. A mouse model was constructed using a multi-drug resistant and virulent UPEC strain and treated with FQLF or the antibiotic imipenem. The treatment efficacy was evaluated by bacterial clearance from urine and the urinary organs. Results： A total of 90 UPEC strains were collected, and 94.4% of the isolates were resistant to at least 1 antibiotic. Approximately 66.7% of the UPEC strains were multi-drug resistant. More than one virulence gene was found in 85.6% of the isolates. The extended-spectrum β-lactamases（ESBL）-positive strains were more resistant than the negative ones. The virulence gene number was positively correlated with the resistance number（P〈0.05）. A mouse model was successful y constructed using UPEC10. Treatment with either FQLF or antibiotics significantly cleared bacteria from the mouse urine after 14 days. In the untreated control, the bacteria lasted for 28 days. FQLF treatment of the UTI mouse model greatly reduced the bacterial number in the kidney and bladder, but could not completely clear the bacteria. Conclusions： Multi-drug resistance is common among UPEC isolates, and the resistance is positively related with virulence. FQLF could treat UPEC UTIs, but could not completely clear the bacteria from the host.

Large intelligent surface/antennas(LISA),a two-dimensional artificial structure with a large number of reflective-surface/antenna elements,is a promising reflective radio technology to construct programmable wireless environments in a smart ***,each element of the LISA adjusts the reflection of the incident electromagnetic waves with unnatural properties,such as negative refraction,perfect absorption,and anomalous reflection,thus the wireless environments can be software-defined according to various design *** this paper,we introduce the reflective radio basics,including backscattering principles,backscatter communication,reflective relay,the fundamentals and implementations of LISA ***,we present an overview of the state-of-the-art research on emerging applications of LISA-aided wireless ***,the limitations,challenges,and open issues associated with LISA for future wireless applications are discussed.

The skin is the most common site of Staphylococcus aureus infection,which can lead to various diseases,including invasive and life-threatening infections,through evasion of host ***,little is known about the host factors that facilitate the innate immune evasion of *** in the ***,which is abundantly expressed in the skin and can be activated by proteases derived from ***,has both direct bacteria-killing activity and immunomodulatory effects via interactions with its receptor ***,we demonstrate that a lack of the chemerin/CMKLR1 axis increases the neutrophil-mediated host defense against *** in a mouse model of cutaneous infection,whereas chemerin overexpression,which mimics high levels of chemerin in obese individuals,exacerbates *** cutaneous ***,we identified keratinocytes that express CMKLR1 as the main target of chemerin to suppress ***-induced IL-33 expression,leading to impaired skin neutrophilia and bacterial ***1 signaling specifically inhibits IL-33 expression induced by cell wall components but not secreted proteins of *** by inhibiting Akt activation in mouse ***,our study revealed that the immunomodulatory effect of the chemerin/CMKLR1 axis mediates innate immune evasion of *** in vivo and likely increases susceptibility to *** infection in obese individuals.

Biological productivity and carbon cycling in the Arctic Ocean

Primary production, bacterial production, par-ticulate organic carbon fluxes and organic carbon burial rates were quantified during the summer period of 1999 in the Arctic Ocean via 14C uptake, 3H uptake, 234Th/238U disequilibrium and 210Pbex dating, respectively. The integrated primary production in the water column was as high as 197 mmolC/(m2·d) in the Chukchi shelf and was 3.8 mmolC/(m2·d) in the Canada Basin. These rates are higher than those reported previously. The ratios of bacterial production to primary production in the study region were higher than 0.5, indicating that microbial activity is not depressed but important in cold Arctic waters. 234Th/238U dis-equilibria were evident at the station in the Canada Basin. The presence of significant 234Th deficiency suggested that scavenging and removal processes are also important to bio-geochemical cycles of trace elements in the Arctic Ocean. We report a passively Q-switched and mode-locked erbium-doped fiber laser(EDFL) based on PtSe_2, a new two-dimensional material, as a saturable absorber(SA). Self-started Q-switching at 1560 nm in the EDFL was achieved at a threshold pump power of 65 mW, and at the maximum pump power of 450 m W, the maximum single Q-switched pulse energy is 143.2 n J. Due to the polarization-dependent characteristics of the PtSe_2-based SA, the laser can be switched from the Q-switched state to the mode-locked state by adjusting the polarization state. A mode-locked pulse train with a repetition rate of 23.3 MHz and a pulse width of 1.02 ps can be generated when the pump power increases to about 80 m W, and the stable mode-locked state is maintained until the pump power reaches its maximum 450 m W. The maximum single mode-locked pulse energy is0.53 nJ. This is the first time to our knowledge that successful generation of stable Q-switched and mode-locked pulses in an Er-doped fiber laser has been obtained by using PtSe_2 as a saturable absorber.

There are debates regarding whether a wet and warm climate or a dry and cold climate dominated Holocene fire activity in northern China on the millennial timescale,and when human activities overtook climate change as the dominant control on fire occurrence in the *** we present a high-resolution fire history for the past~15,500 years from a sediment core in Dali Lake,located in the foothills of the Greater Hinggan Mountains,one of the areas of highest fire risk in *** results demonstrate that fire activity was rare during the last deglaciation(~15,500–11,700 yr BP),gradually increased at the beginning of the Holocene,and reached its highest level during~9000–5000 yr BP,after which there was a decreasing ***,after~2000 yr BP this decreasing trend ended,and the most prominent feature is a peak in fire activity during the Medieval Warm Period(MWP).Overall,fire activity corresponded well to changes in the East Asian summer monsoon(EASM)precipitation on the millennial timescale during~15,500–2000 yr BP,but this relationship changed after~2000 yr *** propose that fire activity in northern China on the millennial timescale during~15,500–2000 yr BP was dominated by the biofuels reserve under the control of the EASM *** contrast,with the intensification of human activities after~2000 yr BP,human activity caused a~62%–73%increase in fire activity,which altered the fire-climate relationship that had previously prevailed in northern *** results indicate that a wet-warm climate(increased EASM intensity),rather than a dry-cold climate,was the dominant control on fire activity in northern China during 15,500–2000 yr BP on the millennial timescale,but that human activities played an important role in fire occurrence after~2000 yr BP.

Photosystem I(PSI)is a large protein supercomplex that catalyzes the light-dependent oxidation of plastocyanin(or cytochrome c6)and the reduction of *** catalytic reaction is realized by a transmembrane electron transfer chain consisting of primary electron donor(a special chlorophyll(Chl)pair)and electron acceptors A_(0),A_(1),and three Fe_(4)S_(4) clusters,F_(X),F_(A),and F_(B).Here we report the PSI structure from a Chl d-dominated cyanobacterium Acaryochloris marina at 3.3Åresolution obtained by single-particle cryo-electron *** *** PSI exists as a trimer with three identical ***,the structure reveals a unique composition of electron transfer chain in which the primary electron acceptor A_(0) is composed of two pheophytin a rather than Chl a found in any other well-known PSI structures.A novel subunit Psa27 is observed in the *** PSI *** addition,77 Chls,13α-carotenes,two phylloquinones,three Fe-S clusters,two phosphatidyl glycerols,and one monogalactosyl-diglyceride were identified in each PSI *** results provide a structural basis for deciphering the mechanism of photosynthesis in a PSI complex with Chl d as the dominating pigments and absorbing far-red light.

OBJECTIVE To investigate whether SPRY4-IT1 is involved in the promotion of breast cancer stem cells(BCSCs). METHODS We used q RT-PCR to detect the expression of SPRY4-IT1 in MCF-7 cells and MCF-7 cancer stem cells(MCF-7 CSCs). The effects of SPRY4-IT1 on the proliferation and renewal ability of breast cancer cells were investigated by in vitro and in vivo assays(ie, in situ hybridization, colony formation assay,sphere formation assay, flow cytometry assay, Western blotting, xenograft model and immunohistochemistry).The mechanism of SPPRY4-IT1 as a ce RNA was studied by a dual-luciferase reporter assay and bioinformatic analysis. RESULTS SPRY4-IT1 was upregulated in MCF-7 CSCs compared with MCF-7 cells, and high SPRY4-IT1 expression was related to reduced breast cancer patient survival. Furthermore, SPRY4-IT1 overexpression promoted breast cancer cell proliferation and stemness in vitro and in vivo. In addition, SPRY4-IT1 knockdown suppressed BCSC renewal ability and stemness maintenance in vivo and in vitro. The dual-luciferase reporter assays indicated that SPRY4-IT1 as a sponge for mi R-6882-3 p repressed transcription factor7-like 2(TCF7 L2) expression. CONCLUSION SPRY4-IT1 promotes proliferation and stemness of breast cancer cells as well as renewal ability and stemness maintenance of BCSCs by increasing the expression of TCF7 L2 through targeting mi R-6882-3 p and may be a potential therapeutic target and biomarker for treating breast cancer.
