Alterations of Gefitinib Pharmacokinetics by Co-administration of Herbal Medications in Rats

Objective： To evaluate the potential pharmacokinetic interactions of the anticancer agent gefitinib （Iressae） and the oriental medications Guipi Decoction （归脾汤, GPD, Guibi-tang in Korean） and Bawu Decoction （八物汤, BWD, Palmul-tang in Korean）. Methods： Methylceliulose （MC, control）, GPD （1,200 mg/kg）, or BWD （6,000 mg/kg） was orally administered to rats either as a single dose or multiple doses prior to gefitinib administration. To examine the effects of a single dose of the herbal medicines, gefitinib （10 mg/kg） was orally administered after 5 min or 1 h of MC or the herbal medicine pretreatments. To examine the effects of the multiple doses of the herbal medicines, gefltinib （10 mg/kg） was orally administered following 7 consecutive days of the administration of MC or each herbal medicine. The plasma concentrations of gefitinib were determined with liquid chromatography-tandem mass spectrometry assay. The plasma concentration-time profiles of gefitinib were analyzed with a noncompartmental analysis. Results： Gefitinib was rapidly absorbed and showed a mono- exponential decline with an elimination half-life of 3.7-4.1 h. The pharmacokinetics of gefitinib was not affected by GPD pretreatment. However, a significantly lower maximum plasma concentration （Crux, P〈0.05） and area under the curve （P〈0.05）, and a delayed time to reach Cmax （Tmax, P〈0.01） were observed in both single- and multiple- dose BWD-pretreated rats compared with the control rats. Conclusions： BWD and not GPD might delay and interfere with gefitinib absorption. Further evaluations of the clinical significance of these findings are needed.

Cholecystocolic fistula caused by gallbladder carcinoma:Preoperatively misdiagnosed as hepatic colon carcinoma

Cholecystocolic fistula secondary to gallbladder carcinoma is extremely rare and has been reported in very few studies. Most cholecystocolic fistulae are late complications of gallstone disease, but can also develop following carcinoma of the gallbladder when the necrotic tumor penetrates into the adjacent colon. Although no currently available imaging technique has shown great accuracy in recognizing cholecystocolic fistula, abdominopelvic computed tomography may show fistulous communication and anatomical *** we report an unusual case of cholecystocolic fistula caused by gallbladder carcinoma, which was preoperatively misdiagnosed as hepatic flexure colon carcinoma.

Identification of Aristolactam Derivatives That Act as Inhibitors of Human Immunodeficiency Virus Type 1 Infection and Replication by Targeting Tat-Mediated Viral Transcription

Despite the success of antiretroviral therapy(ART),efforts to develop new classes of antiviral agents have been hampered by the emergence of drug ***-indole-bearing aristolactams are compounds that have been isolated from various plants species and which show several clinically relevant effects,including anti-inflammatory,antiplatelet,and antimycobacterial ***,the effect of these compounds on human immunodeficiency virus type 1(HIV-1)infection has not yet been *** this study,we discovered an aristolactam derivative bearing dibenzo[cd,f]indol-4(5 H)-one that had a potent anti-HIV-1 effect.A structure-activity relationship(SAR)study using nine synthetic derivatives of aristolactam identified the differing effects of residue substitutions on the inhibition of HIV-1 infection and cell *** the compounds tested,1,2,8,9-tetramethoxy-5-(2-(piperidin-1-yl)ethyl)-dibenzo[cd,f]indol-4(5 H)-one(Compound 2)exhibited the most potent activity by inhibiting HIV-1 infection with a half-maximal inhibitory concentration(IC50)of 1.03 lmol/L and a half-maximal cytotoxic concentration(CC50)of 16.91 lmol/L(selectivity index,16.45).The inhibitory effect of the compounds on HIV-1 infection was linked to inhibition of the viral replication ***-of-action studies showed that the aristolactam derivatives did not affect reverse transcription or integration;instead,they specifically inhibited Tat-mediated viral *** together,these findings show that several aristolactam derivatives impaired HIV-1 infection by inhibiting the activity of Tat-mediated viral transcription,and suggest that these derivatives could be antiviral drug candidates.

In this study,the texture of a rolled Mg alloy is effectively modified through the application of precompression and subsequent annealing treatment,leading to a remarkable improvement in the bending formability of the alloy at room *** induces lattice reorientation through{10-12}twinning,and annealing treatment reduces the stored strain energy of the precompressed material,which results in the formation of a stable grain structure with two dominant texture *** an increase in precompression,the tensile strain in the outer region of the bending samples is accommodated to a greater extent due to more pronounced{10-12}twinning and basal *** a result,the bending formability of the material at room temperature improves with greater *** variation in microstructure,texture,and bending behavior in relation to the degree of precompression is discussed in detail.

Molecular mechanisms of ZD1839 (Iressa)-induced apoptosis in human leukemic U937 cells

Aim:To investigate the molecular mechanisms of ZD1839-induced apoptosis inhuman leukemic U937 ***:The inhibition of human leukemic U937 cellgrowth was assessed by 3-（4,5-dimethyl-2-thiazolyl）-2,5-diphnyl-2H-tetrazolimbromide（MTT）assays,lactate dehydrogenase（LDH）release,and cell *** expression of anti- and pro-apoptotic proteins was detected byWestern blot ***:This study demonstrated that ZD 1839 inducedapoptosis in leukemic U937 cells by the downregulation of Bcl-2,caspase activa-tion and subsequent apoptotic *** with ZD 1839 and the caspase-3 inhibitor z-DEVD-fmk blocked apoptosis,indicating that caspase-3 activation isat least partially responsible for ZD1839-induced *** ectopic expres-sion of Bcl-2 attenuated caspase-3 activation,PARP cleavage,and subsequentindicators of apoptosis,including sub-G1DNA content and LDH *** indicate that the downregulation of Bcl-2 plays a major role in the initiationof ZD1839-induced apoptosis,and that the activation of a caspase cascade isinvolved in the execution of ***,ZD 1839 treatment triggeredthe activation of p38 mitogen-activated protein kinase（MAPK）and the down-regulation of c-Jun-N-terminal kinase（JNK）,extracellular signal-regulated kinase（ERK）and phosphatidyl inositol 3-kinase（PI3K）/*** inhibition of the ERKand PI3K/Akt pathways also significantly increased cellular ***:ZD 1839 activated caspase-3 and the inhibited Bcl-2 in human leukemic U937 cellsthrough the downregulation of the ERK and PI3K/Akt pathways.

Creep and tensile properties of press molding joined GMT-sheets

GMT-sheet is used in automobile bumper with high rigidity and strength, and its joining strength is influenced by lap length, one of the joined molding conditions. Fracture strength was calculated by dividing fracture load with cross-sectional area. Total five repeated measurements were made to obtain the average value. Tensile test was conducted at room temperature for 10 specimens. In addition, the effect of compression ratio on creep and tensile performance during lap joined molding was discussed. With increasing lap length, the lap joining efficiency of GMT-sheet was increased. However, higher compression ratio reduced the joining efficiency. Creep test on GMT-sheet showed abrupt fracture without tertiary creep. This can be explained by the weak thermal resistance of the resin. If GMT-sheet was exposed to high temperature for a long time, it was easily failed by external force.

Nanostructure and corrosion behaviors of nanotube formed Ti-Zr alloy

In order to investigate the nanostructures and corrosion behaviors of Ti-Zr alloys,nanotube formed Ti-Zr(10%,20%,30% and 40% in mass fraction) alloys were prepared by arc melting and the condition of controlling nanostructure was at 1 000 ℃ for 24 h in argon atmosphere;formation of nanotubes was conducted by anodizing a Ti-Zr alloy in H3PO4 electrolyte with a small amount of fluoride ions at room *** corrosion properties of specimens were examined through potentiodynamic test(potential range of -1 500-2 000 mV) in 0.9% NaCl solution by using *** of the alloys were observed by optical microscope(OM),field emission scanning electron microscope(FE-SEM) and X-ray diffractometer(XRD).Diameter of nanotube does not depend on Zr content,but interspace of nanotube predominantly depends on Zr content,which confirms that ZrO2 oxides play a role to increase the interspace of nanotube formed on the surface.

Synthesis of Al-doped ZnO Nanorods via Microemulsion Method and Their Application as a CO Gas Sensor

Aluminum doped ZnO(AZO) nanorods were synthesized by microemulsion method with different types of *** phase and the morphology of the above nanorods were investigated by scanning electron microscopy(SEM) and X-ray diffraction(XRD).SEM observations showed that the ZnO nanorods had diameters around about 50—200 nm and lengths up to several *** CO gas sensing properties of the AZO nanorods were tested at operating temperatures of 200,300,350 and 400 ℃.It was found that AZO nanorods based sensor exhibited the highest sensitivity to CO at 350℃.

Surgical repair of intractable chylous ascites following laparoscopic anterior resection

Chylous ascites is the accumulation of a milk-like peritoneal fluid rich in triglycerides and it is an unusual complication following surgical treatment of colorectal cancer. Conservative management is usually sufficientin patients with chylous ascites after surgery. However,we describe a patient with intractable chylous ascites after laparoscopic anterior resection for sigmoid colon cancer who failed initial conservative treatment. This patient was successfully managed by surgery.
