The role of innate immunity in diabetic nephropathy and their therapeutic consequences

Diabetic nephropathy (DN) is an enduring condition that leads to inflammation and affects a substantial number of individuals with diabetes worldwide. A gradual reduction in glomerular filtration and emergence of proteins in the urine are typical aspects of DN, ultimately resulting in renal failure. Mounting evidence suggests that immunological and inflammatory factors are crucial for the development of DN. Therefore, the activation of innate immunity by resident renal and immune cells is critical for initiating and perpetuating inflammation. Toll-like receptors (TLRs) are an important group of receptors that identify patterns and activate immune responses and inflammation. Meanwhile, inflammatory responses in the liver, pancreatic islets, and kidneys involve inflammasomes and chemokines that generate pro-inflammatory cytokines. Moreover, the activation of the complement cascade can be triggered by glycated proteins. This review highlights recent findings elucidating how the innate immune system contributes to tissue fibrosis and organ dysfunction, ultimately leading to renal failure. This review also discusses innovative approaches that can be utilized to modulate the innate immune responses in DN for therapeutic purposes.

Mainstream partial nitritation-anammox(PNA)process easily suffers from performance instability and even reactor collapse in application.Thus,it is of great significance to unveil the characteristic of performance recovery,understand the intrinsic mechanism and then propose operational strategy.In this study,we combined long-term reactor operation,batch tests,and metagenomics to reveal the succession of microbial community and functional metabolism variation from system collapse to recovery.Proper aeration control(0.10-0.25mg O_(2)/L)was critical for performance recovery.It was also found that Candidatus Brocadia became the dominant flora and its abundance increased from 3.5%to 11.0%.Significant enhancements in carbon metabolism and phospholipid biosynthesis were observed during system recovery,and the genes abundance related to signal transduction was dramatically increased.The up-regulation of sdh and suc genes showed the processes of succinate dehydrogenation and succinyl-CoA synthesis might stimulate the production of amino acids and the synthesis of proteins,thereby possibly improving the activity and abundance of AnAOB,which was conducive to the performance recovery.Moreover,the increase in abundance of hzs and hdh genes suggested the enhancement of the anammox process.Changes in the abundance of key genes involved in nitrogen metabolism indicated that nitrogen removal pathway was more diverse after system recovery.The achievement of performance recovery was driven by anammox,nitrification and denitrification coupled with dissimilatory nitrate reduction to ammonium.These results provide deeper insights into the recovery mechanism of PNA system and also provide a potential regulation strategy for the stable operation of the mainstream PNA process.

Antibiotic fermentation residue(AFR)is nutrient-rich solid waste generated from fermentative antibiotic production process.It is demonstrated that AFR contains high-concentration of remaining antibiotics,and thus may promote antibiotic resistance development in receiving environment or feeding farmed animals.However,the dominate microorganisms and antibiotic resistance genes(ARGs)in AFRs have not been adequately explored,hampering understanding on the potential antibiotic resistance risk development caused by AFRs.Herein,seven kinds of representative AFRs along their production,storage,and treatment processes were collected,and multiple methods including amplicon sequencing,metagenomic sequencing,and bioinformatic approaches were adopted to explore the biological characteristics of AFRs.As expected,antibiotic fermentation producer was found as the predominant species in raw AFRs,which were collected at the outlet of fermentation tanks.However,except for producer species,more environment-derived species persisted in stored AFRs,which were temporarily stored at a semi-open space.Lactobacillus genus,classified as Firmicutes phylum and Bacilli class,became predominant bacterial taxa in stored AFRs,which might attribute to its tolerance to high concentration of antibiotics.Results from metagenomic sequencing together with assembly and binning approaches showed that these newly-colonizing species(e.g.,Lactobacillus genus)tended to carry ARGs conferring resistance to the remaining antibiotic.However,after thermal treatment,remaining antibiotic could be efficiently removed from AFRs,and microorganisms together with DNA could be strongly destroyed.In sum,the main risk from the AFRs was the remaining antibiotic,while environment-derived bacteria which tolerate extreme environment,survived in ARFs with high content antibiotics,and may carry ARGs.Thus,hydrothermal or other harmless treatment technologies are recommended to remove antibiotic content and inactivate bacteria before recycling of AFRs in pharmaceutical industry.

Animal manures have been demonstrated to enhance antibiotic resistance in agricultural soils.However,little is known about the effects of plant-derived fertilizer on soil antibiotic resistome.Herein,metagenomic sequencing was used to investigate the effects of a plantderived fertilizer processed from sugarcane and beet on soil antibiotic resistance genes(ARGs)in a soybean field along crop growth stages.ARG profiles in the soils amended by plant-derived fertilizer were compared with those in the soils amended by chicken manure.The abundance and diversity of total ARGs in the soils amended by plant-derived fertilizer were significantly(P<0.05)elevated at the sprout stage,to a level comparable to that in the manured soils.Whereas,unlike chicken manure mainly introducing manure-borne ARGs to soil,the plant-derived fertilizer was indicated to mainly enrich multidrug resistance genes in soil by nourishing indigenous bacteria.ARGs with abundances in amended soils significantly(P<0.05)higher than in unamended soils at the sprout stage of soybean were considered as enriched ARGs.Decrease in the abundance of the enriched ARGs was observed in both the amended soils from the sprout to the harvest.Network analysis further identified Proteobacteria and Bacteroidetes as the primary bacterial taxa involved in the temporal variation of the enriched ARGs in the soils amended by plant-derived fertilizer,while in manured soils were Firmicutes and Actinobacteria.As revealed by multivariate statistical analyses,variation of the enriched ARGs in the soils amended by plant-derived fertilizer was majorly attributed to the response of co-occurred bacteria to depleting nutrients,which was different from the failed establishment of manure-borne bacteria in the manured soils.Our study provided field-based evidence that plant-derived fertilizer stimulated the intrinsic antibiotic resistome,and proposed attention to the un-perceived risk since some clinically relevant ARGs originate and evolve from natural resistome.

The biosynthesis of isoprene offers a more sustainable alternative to fossil fuel-based approaches,yet its success has been largely limited to pure organic compounds and the cost remains a challenge.This study proposes a waste-to-wealth strategy for isoprene biosynthesis utilizing genetically engineered E.coli bacteria to convert organic waste from real food wastewater.The impact of organic compounds present in wastewater on E.coli growth and isoprene production was systematically investigated.The results demonstrated that with filtration pretreatment of wastewater,isoprene yield,and production achieved 115 mg/g COD and 7.1 mg/(L·h),respectively.Moreover,even without pretreatment,isoprene yield only decreased by~24%,indicating promising scalability.Glucose,maltose,glycerol,and lactate are effective substrates for isoprene biosynthesis,whereas starch,protein,and acetate do not support E.coli growth.The optimum C/N ratio for isoprene production was found to be 8:1.Furthermore,augmenting essential nutrients in wastewater elevated the isoprene yield increased to 159 mg/g COD.The wastewater biosynthesis significantly reduced the cost(44%–53%decrease,p-value<0.01)and CO_(2)emission(46%–55%decrease,p-value<0.01)compared with both sugar fermentation and fossil fuel–based refining.This study introduced a more sustainable and economically viable approach to isoprene synthesis,offering an avenue for resource recovery from wastewater.

Predictability of the 7·20 extreme rainstorm in Zhengzhou in stochastic kinetic-energy backscatter ensembles

The scale-dependent predictability of the devastating 7·20 extreme rainstorm in Zhengzhou,China in 2021 was investigated via ensemble experiments,which were perturbed on different scales using the stochastic kinetic-energy backscatter(SKEB)scheme in the WRF model,with the innermost domain having a 3-km grid spacing.The daily rainfall(RAIN24h)and the cloudburst during 1600-1700 LST(RAIN1h)were considered.Results demonstrated that with larger perturbation scales,the ensemble spread for the rainfall maximum widens and rainfall forecasts become closer to the observations.In ensembles with mesoscale or convective-scale perturbations,RAIN1h loses predictability at scales smaller than 20 km and RAIN24h is predictable for all scales.Whereas in ensembles with synoptic-scale perturbations,the largest scale of predictability loss extends to 60 km for both RAIN1h and RAIN24h.Moreover,the average positional error in forecasting the heaviest rainfall for RAIN24h(RAIN1h)was 400 km(50-60)km.The southerly low-level jet near Zhengzhou was assumed to be directly responsible for the forecast uncertainty of RAIN1h.The rapid intensification in low-level cyclonic vorticity,mid-level divergence,and upward motion concomitant with the jet dynamically facilitated the cloudburst.Further analysis of the divergent,rotational and vertical kinetic spectra and the corresponding error spectra showed that the error kinetic energy at smaller scales grows faster than that at larger scales and saturates more quickly in all experiments.Larger-scale perturbations not only boost larger-scale error growth but are also conducive to error growth at all scales through a downscale cascade,which indicates that improving the accuracy of larger-scale flow forecast may discernibly contributes to the forecast of cloudburst intensity and position.

In recent years,space-division multiplexing(SDM)technology,which involves transmitting data information on multiple parallel channels for efficient capacity scaling,has been widely used in fiber and free-space optical communication sys-tems.To enable flexible data management and cope with the mixing between different channels,the integrated reconfig-urable optical processor is used for optical switching and mitigating the channel crosstalk.However,efficient online train-ing becomes intricate and challenging,particularly when dealing with a significant number of channels.Here we use the stochastic parallel gradient descent(SPGD)algorithm to configure the integrated optical processor,which has less com-putation than the traditional gradient descent(GD)algorithm.We design and fabricate a 6×6 on-chip optical processor on silicon platform to implement optical switching and descrambling assisted by the online training with the SPDG algorithm.Moreover,we apply the on-chip processor configured by the SPGD algorithm to optical communications for optical switching and efficiently mitigating the channel crosstalk in SDM systems.In comparison with the traditional GD al-gorithm,it is found that the SPGD algorithm features better performance especially when the scale of matrix is large,which means it has the potential to optimize large-scale optical matrix computation acceleration chips.

Background Rosemary extract(RE)has been reported to exert antioxidant property.However,the application of RE in late-phase laying hens on egg quality,intestinal barrier and microbiota,and oviductal function has not been systematically studied.This study was investigated to detect the potential effects of RE on performance,egg quality,serum parameters,intestinal heath,cecal microbiota and metabolism,and oviductal gene expressions in late-phase laying hens.A total of 21065-week-old“Jing Tint 6”laying hens were randomly allocated into five treatments with six replicates and seven birds per replicate and fed basal diet(CON)or basal diet supplemented with chlortetracycline at 50 mg/kg(CTC)or RE at 50 mg/kg(RE50),100 mg/kg(RE100),and 200 mg/kg(RE200).Results Our results showed that RE200 improved(Pmino acid metabolism of cecal microbiota and increased butyrate synthesizing enzymes,including 3-oxoacid Co A-transferase and butyrate-acetoacetate Co A-transferase.Moreover,transcriptomic analysis revealed that RE200 improved gene expressions and functional pathways related to immunity and albumen formation in the oviductal magnum.Conclusions Dietary supplementation with 200 mg/kg RE could increase egg quality of late-p

The sudden onset of the coronavirus disease 2019(COVID-19)in January 2020 has affected essential global health services.Cancer-screening services that can reduce cancer mortality are strongly affected.However,the specific role of COVID-19 in cancer screening is not fully understood.This study aimed to assess the efficiency of global cancer screening programs before and during the COVID-19 pandemic and to promote potential cancer-screening strategies for the next pandemic.Electronic searches in PubMed,Embase,and Web of Science,and manual searches were performed between January 1,2020 and March 1,2023.Cohort studies that reported the number of participants who underwent cancer screening before and during the COVID-19 pandemic were included.The methodological quality of the included studies was assessed using the Newcastle-Ottawa Scale.Differences in cancer-screening rates were estimated using the incidence rate ratio(IRR).Fifty-five cohort studies were included in this meta-analysis.The screening rates of colorectal cancer using invasive screening methods(Pooled IRR=0.52,95%CI:0.42 to 0.65,p<0.01),cervical cancer(Pooled IRR=0.56,95%CI:0.47 to 0.67,p<0.01),breast cancer(Pooled IRR=0.57,95%CI:0.49 to 0.66,p<0.01)and prostate cancer(Pooled IRR=0.71,95%CI:0.56 to 0.90,p<0.01)during the COVID-19 pandemic were significantly lower than those before the COVID-19 pandemic.The screening rates of lung cancer(Pooled IRR=0.77,95%CI:0.58 to 1.03,p=0.08)and colorectal cancer using noninvasive screening methods(Pooled IRR=0.74,95%CI:0.50 to 1.09,p=0.13)were reduced with no statistical differences.The subgroup analyses revealed that the reduction in cancer-screening rates varied across economies.Our results suggest that the COVID-19 pandemic has had a noteworthy impact on colorectal,cervical,breast,and prostate cancer screening.Developing innovative cancer-screening technologies is important to promote the efficiency of cancer-screening services in the post-COVID-19 era and prepare for the next pandemic.
