Autophagy in colorectal cancer:An important switch from physiology to pathology

Colorectal cancer(CRC) remains a leading cause of cancer death in both men and women *** the factors and mechanisms that are involved in the multifactorial etiology of CRC,autophagy is an important transformational switch that occurs when a cell shifts from normal to *** recent years,multiple hypotheses have been considered regarding the autophagy mechanisms that are involved in *** currently accepted hypothesis is that autophagy has dual and contradictory roles in carcinogenesis,but the precise mechanisms leading to autophagy in cancer are not yet fully defined and seem to be context *** is a surveillance mechanism used by normal cells that protects them from the transformation to malignancy by removing damaged organelles and aggregated proteins and by reducing reactive oxygen species,mitochondrial abnormalities and DNA ***,autophagy also supports tumor formation by promoting access to nutrients that are critical to the metabolism and growth of tumor cells and by inhibiting cellular death and increasing drug *** studies in CRC have focused on several molecules,mainly microtubule-associated protein 1 light chain 3,beclin 1,and autophagy related 5,with conflicting *** effects were observed for some agents that modulate autophagy in CRC either alone or,more often,in combination with other *** extensive studies are needed in the future to clarify the roles of autophagy-related genes and modulators in colorectal carcinogenesis,and to develop potential beneficial agents for the prognosis and treatment of CRC.

Male gender and increased body mass index independently predicts clinically relevant morbidity after spleen-preserving distal pancreatectomy

AIM To identify risk factors for clinically relevant complications after spleen-preserving distal pancreatectomy(SPDP). No previous studies explored potential predictors of morbidity after *** The data of 41 patients who underwent a SPDP in a single surgical center between 2000 and 2015 were retrospectively reviewed from a prospectively maintained electronic database established in our Department of Surgery. The database included demographic, clinical, bioumoral, pathological, intraoperative and postoperative parameters. Uni-and multivariate ana-lyses were performed to assess potential predictors of clinically relevant morbidity. Postoperative morbidity was defined as in-hospital complications and mortality was assessed at 90 d. Clinically relevant morbidity was defined as complication ≥ grade 2 *** Overall morbidity rate was 34.1%(14 patients): grade Ⅰ(6 patients, 14.6%), grade Ⅱ(2 patients, 4.8%), grade Ⅲa(1 patient, 2.4%), and grade Ⅲb(5 patients, 12.2%). A number of 5 patients(12.2%) required re-laparotomy for postoperative complications. There was no postoperative mortality. Thus, at least one clinically relevant complication occurred in 8 patients(19.5%). Univariate analysis identified male gender(P = 0.034), increased body mass index(P = 0.002) and neuroendocrine pathology(P = 0.013) as statistically significant risk factors. Multivariate analysis identified male gender [odds ratio(OR): 1.29, 95%CI: 1.07-1.55, P = 0.005] and increased body mass index(OR: 23.18, 95%CI: 1.72-310.96, P = 0.018) as the only independent risk factors of clinically relevant morbidity after *** Male gender and increased body mass index are independently associated with increased risk of clinically relevant morbidity after SPDP. These findings may assist a surgeon in clinical decision-making to better select patients suitable for SPDP.

Endoscopic knot tying:In vitro assessment in a porcine stomach model

AIM:To determine if surgical knotting performed via endoscopy is an effective closure method for natural orifice translumenal endoscopic surgery. METHODS:The proposed method was tested on an in vitro pig stomach model using standard endoscopy suite materials. A single use laparoscopy trocar (Versaport Plus manufactured by Tyco Healthcare) was fixed onto a plastic rectangular box in a horizontal position. A fresh pig stomach was tightly attached via its esophageal end to the trocar opening on the inner side of the box. The stomach cavity was closed at the duodenal end with Kocher forceps. A standard upper gastrointestinal endoscope fitted at its tip with a transparent plastic cap was introduced into the stomach through the outer trocar opening, so that the passage of the surgical trocar would mimic the passage of an esophagus. The stomach was subsequently inflated, followed by irrigation and washing. A neutral electrode of an electrocautery unit was placed inside the plastic box, un-derneath the pig stomach. The stomach's outer surface was kept moist using normal saline in order to maintain the natural elasticity and to ensure good contact with the electrode. RESULTS:The submucosal space on the anterior face of the stomach was accessed using the technique of endoscopic submucosal dissection. First, a site on the anterior face of the stomach was chosen, near the angle. Then, saline was injected into the submucosa with a standard endoscopic needle, so as to create a 20 mm diameter elevation. A linear 15 mm vertical incision was created at its center using a Dual Knife (KD650U manu-factured by Olympus). This incision was used to access the submucosal space, and about 10 mm was dissected on both sides of the incision. The endoscope was then pushed through to the outside of the stomach after dilating a small puncture made by the Dual Knife in the muscularis propria , which simulated the peritoneoscopy procedure. Multi-physics analysis and optimisation of high-speed train pantograph-catenary systems allowing for velocity skin effect

A pantograph-catenary system (PCS) - an essential component to supply a high-speed train (HST) - faces a variety of new challenges due to the continuously increasing train speeds. The HST traction system receives power via an electrical contact between the pantograph strip and the high-voltage contact wire. This electrical contact is subject to serious mechanical shocks and significant electrochemical corrosion, making the modelling of the dynamic processes complicated, especially under high-speed and heavy-load conditions. The damage to the PCS - which is particularly noticeable at the edges of the pantograph strip - may become severe as the speed of the train rises. Moreover, as the speed increases, the distribution of the electrical current in the strip becomes uneven due to the velocity skin effect (VSE). To assess the impact of the VSE on the performance of PCSs, a multi-physics model has been created and is reported in this study. The model has been validated through experiments and the main aspects of its functionality - such as the VSE, friction, and air convection - have been identified and analysed at different speeds. The impact of speed on the traction current and the behaviour of thermal sources have been explored. With the increasing speed, the phenomenon of current clustering at the trailing edge of the strip becomes quite dramatic, resulting in a thermal surge in the region of the strip with high current density. To mitigate the negative impact caused by VSE in the PCSs, an improved kriging optimisation methodology has been utilised to optimise the parameters of the PCS. Recommendations regarding the optimal design of the PCS are put forward to improve the current-carrying performance and reduce the local temperature rise in the strip.

Phase Structures of Nascent Polyethylene Powder Studied by Wideline Proton NMR

The wideline proton NMR spectra of polyethylene powder samples were analyzed in terms of contributions from three components：（1） a rigid part with immobile chains, （2） a soft region with liquid-like character which produces a Lorentzian contribution to the spectrum, and （3） an intermediate region in which the rotation of methylene groups about C-C bonds is partially hindered. The relative mass fractions as well as chain mobilities varied greatly among samples produced by different polymerization techniques. The NMR crystallinity agreed well with that estimated by WAXD and was much higher than DSC crystallinity, indicating an inclusion of the contribution from a crystalline-amorphous interphase. The crystalline defects in the rigid part could be significantly affected by processing parameters when employing the same type of polymerization technique. The intermediate region in the NMR spectra was analyzed according to the comparison between bimodal high density polyethylene and corresponding linear unimodal one. It was found that the mass fraction of the NMR interphase could be an indication of the percentage of tie molecules between crystalline lamellae and thus may significantly affect the mechanical properties of polymeric material.

Vibration Control for Two Air Compressor Units Used in Tandem

The paper presents the vibration control for two air compressors used in tandem to an oxygen generating station. The compressors are identical in terms of constructive and functional properties, and are both located on similar foundations. Each compressor is supported on a foundation made from one massive block of concrete, provided with some elastic and damping parts. This anti vibrating structure insulates the machines compared to other station equipment. During operation, were observed dangerous levels of vibration at one of the machines, which forced some unwanted stoppages for the station. Two hypotheses have been promoted. One of them referred to a pronounced wear of one compressor, although this was relatively new. The second hypothesis has taken in consideration the appearance of some cracks in the foundation massive concrete block. Experimental research conducted on the spot led to the identification of some errors done at foundation fabrication. The experimental results and some theoretical investigations are presented in this paper.
