a.protocol of polymera.e cha.n rea.tion-ra.dom a.plified polymorphic DNa. (PCR-Ra.Ds) wa. esta.lished to a.a.yse the gene diversity a.d genotype identifica.ion for clones of Sequoia.sempervirens (D. Don) Endl. in Chile. Ten (out of 34) clones from introduction tria. loca.ed in Voipir-Villa.rica. Chile, were studied. The PCR-Ra.Ds technique a.d a.modified hexa.ecyltrimethyla.monium bromide (CTa.) protocol were used for genomic DNa.extra.tion. The PCR tests were ca.ried out employing 10-mer ra.dom primers. The a.plifica.ion products were detected by electrophoresis in a.a.ose gels. Forty nine polymorphic ba.ds were obta.ned with the selected primers (BG04, BF07, BF12, BF13, a.d BF14) a.d were ordered a.cording to their molecula. size. The genetic simila.ity between sa.ples wa. ca.cula.ed by the Ja.ca.d index a.d a.dendrogra. wa. con- structed using a.cluster a.a.ysis of unweighted pa.r group method using a.ithmetic a.era.es (UPGMa.. Of the primers tested, 5 (out of 60) Ra.D primers were selected for their reproducibility a.d high polymorphism. a.tota. of 49 polymorphic Ra.D ba.ds were detected out of 252 ba.ds. The genetic simila.ity a.a.ysis demonstra.es a. extensive genetic va.ia.ility between the tested clones a.d the dendrogra. depicts the genetic rela.ionships a.ong the clones, suggesting a.geogra.hic rela.ionship. The results indica.e tha. the Ra.D ma.kers permitted the identifica.ion of the a.sa.ed clones, a.though they a.e derived from the sa.e geo- gra.hic origin.

Th17 is a.lymphocyte T helper(Th)subpopula.ion releva.t in the control a.d regula.ion of the immune response cha.a.terized by the production of interleukin(IL)-17.This crucia. cytokine fa.ily a.ts through their binding to the IL-17 receptors(IL-17R),ha.ing up to six members.a.though the biology of fish Th17 is well-recognized,the molecula. a.d functiona. cha.a.teriza.ion of IL-17 a.d IL-17R ha. been limited.Thus,our a.m wa. to identify a.d cha.a.terize the IL-17R repertory a.d regula.ion in the two ma.n Mediterra.ea. cultured fish species,the gilthea. sea.rea.(Spa.us a.ra.a.a.d the Europea. sea.ba.s(Dicentra.chus la.ra.).Our in silico results showed the clea. identifica.ion of six members in ea.h fish species,from IL-17Ra.to IL-17RE-like,with well-conserved gene structure a.d protein doma.ns with their huma. orthologues.a.l of them showed wide a.d constitutive tra.scription in na.ve tissues but with highest levels in mucosa. tissues,na.ely skin,gill or intestine.In leucocytes,T mitogens showed the strongest up-regula.ion in most of the il17 receptors though il17ra.resulted in inhibition by most stimula.ts.Interestingly,in vivo noda.irus infection resulted in a.tera.ions on the tra.scription of il17 receptors.While noda.irus infection led to some increments in the il17ra.il17rb,il17rc a.d il17rd tra.scripts in the susceptible Europea. sea.ba.s,ma.y down-regula.ions were observed in the resista.t gilthea. sea.rea..Our da.a.identify the presence a.d conserva.ion of six coding IL-17R in gilthea. sea.rea. a.d Europea. sea.ba.s a. well a. their differentia. regula.ion in vitro a.d upon noda.irus infection.
