Hepatic reconstruction from fetal porcine liver cells using a radial flow bioreactor

AIM： To examine the efficacy of the radial flow bioreactor （RFB） as an extracorporeal bioartificial liver （BAL） and the reconstruction of liver organoids using embryonic pig liver cells. METHODS： We reconstructed the liver organoids using embryonic porcine liver cells in the RFB. We also determined the gestational time window for the optimum growth of embryonic porcine liver cells. Five weeks of gestation was designated as embryonic day （E） 35 and 8 wk of gestation was designated as E56. These cells were cultured for one week before morphological and functional examinations. Moreover, the efficacy of pulsed adminisbation of a high concentration hepatocyte growth factor （HGF） was examined. RESULTS： Both cell growth and function were excellent after harvesting on E35. The pulsed administration of a high concentration of HGF promoted the differentiation and maturation of these fetal hepatic cells. Microscopic examination of organoids in the RFB revealed palisading and showed that bile duct-like structures were well developed, indicating that the organoids were mini livers. Transmission electron microscopy revealed microvilli on the luminal surfaces of bile duct-like structures and junctional complexes, which form the basis of the cytoskeleton of epithelial tissues. Furthermore, strong expression of connexin （Cx） 32, which is the main protein of hepatocyte gap junctions, was observed. With respect to liver function, ammonia detoxification and urea synthesis were shown to be performed effectively. CONCLUSION： Our system can potentially be applied in the fields of BAL and transplantation medicine.

Molecules such as water,proteins and lipids that are contained in biological tissue absorb mid-infrared(MR)light,which allows such light to be used in laser surgical ***,amides and water exhibit strong absorption bands in the 5-7μm wavelength range,but at present there are no lasers in clinical use that can emit in this ***,the present study focused on thequantum cascade laser(QCL),which is a new type of semiconductor laser that can emit at MIRwavelengths and has recentiy achieved high output power.A high-power QCL with a peakwavelength of 5.7μm was evaluated for use as a laser scalpel for ablating biological soft *** interaction of the laser beam with chicken breast tissue was compared to a conventional CO_(2) laser,based on surface and cross-sectional *** QCL was found to have sufficient power to ablate soft tissue,and its coagulation,carbonization and ablation effects were similar to those forthe CO_(2) *** QCL also induced comparable photothermal effects because it acted as apseudo-continuous wave laser due to its low peak power.A QCL can therefore be used as an effective laser scalpel,and also offers the possibility of less invasive treatment by targeting specificabsorption bands in t he MIR region.

We have read the position paper by *** Freeman and the members of the Pancreas Nutrition Committee of the North American and the European Societies for Pediatric Gastroenterology,Hepatology,and Nutrition(NASPGHAN and ESPGHAN)(1).That paper focused on chronic pancreatitis(CP)in children and endoscopic procedures about which NASPGHAN published a second position paper(2).There are clinical practice guidelines for the management of CP in adults but not for CP in children(3).

To the Editor:Pancreatic and peripancreatic collections(PCs)develop from acute pancreatitis(AP),chronic pancreatitis,surgery,or *** 2012 revised Atlanta classification[1]of AP classified local complications into the following 4 PC types:acute peripancreatic fluid collection(<4 weeks after the onset of acute interstitial edematous pancreatitis),acute necrotic collection(<4 weeks after the onset of acute necrotizing pancreatitis),pancreatic pseudocyst(PPC;≥4 weeks after the onset of acute interstitial edematous pancreatitis),and walled-off necrosis(WON;≥4 weeks after the onset of acute necrotizing pancreatitis).Endoscopic ultrasonographyguided transmural drainage(EUS-TD)has been reported to be a minimally invasive procedure for patients with PCs[2–4].This study aimed to investigate the clinical outcomes of EUS-TD for PCs.

Examination of in vivo mutagenicity of sodium arsenite and dimethylarsinic acid in gpt delta rats

Arsenic is a well-known human bladder and liver carcinogen, but its exact mechanism of carcinogenicity is not fully understood. Dimethylarsinic acid（DMAV） is a major urinary metabolite of sodium arsenite（i As~Ⅲ） and induces urinary bladder cancers in rats. DMAVand i As~Ⅲare negative in in vitro mutagenicity tests. However, their in vivo mutagenicities have not been determined. The purpose of present study is to evaluate the in vivo mutagenicities of DMAVand i As~Ⅲin rat urinary bladder epithelium and liver using gpt delta F344 ***-week old male gpt delta F344 rats were randomized into 3 groups and administered 0,92 mg/L DMAV, or 87 mg/L i As~Ⅲ（each 50 mg/L As） for 13 weeks in the drinking water. In the mutation assay, point mutations are detected in the gpt gene by 6-thioguanine selection（gpt assay） and deletion mutations are identified in the red/gam genes by Spi-selection（Spi-assay）. Results of the gpt and Spi-assays showed that DMAVand i As~Ⅲhad no effects on the mutant frequencies or mutation spectrum in urinary bladder epithelium or liver. These findings indicate that DMAVand i As~Ⅲare not mutagenic in urinary bladder epithelium or liver in rats.

Detonation Propagation in Narrow Gaps with Various Configurations

In general all detonation waves have cellular structure formed by the trajectory of the triple points. This paper aims to investigate experimentally the propagation of detonation in narrow gaps for hydrogen-oxygen-argon mixtures in terms of various gap heights and gap widths. The gap of total length 1500 mm was constructed by three pair of stainless plates, each of them was 500 mm in length, which were inserted in a detonation tube. The gap heights were varied from 1.2 mm to 3.0 mm while the gap widths were varied from 10 mm to 40 mm. Various argon dilution rates were tested in the present experiments to change the size of cellular structure. Attempts have been made by means of reaction front velocity, shock front velocity, and smoked foil to record variations of cellular structure inside the gaps. A combination probe composed of a pressure and an ion probe detected the arrival of the shock and the reaction front individually at one measurement point. Experimental results show that the number of the triple points contained in detonation front decreases with decrease in the gap heights and gap widths, which lead to larger cellular structures. For mixtures with low detonability, cell size is affected by a certain gap width although conversely cell size is almost independent of gap width. From the present result it was found that detonation propagation inside the gaps is strongly governed by the gap height and effects of gap width is dependent on detonability of mixtures.

Effects of Cultivation Conditions on Chlorophyll Biosynthesis and Carotenoid Accumulation of the Microalga Chlorella Fusca

The purpose of the present study is to optimize the cultivation conditions for the productivities of chlorophyll and carotenoid of the microalga chlorellafusca in a flow bioreactor. The effects of CO2 concentration in aeration and light intensity as the culture condition were firstly examined. After the cultivation conditions as to CO2 concentration and light intensity were determined, the effects of the concentrations of KNO3, CH3COONa, NaC1 and MgSO4 in the culture medium on the chlorophyll biosynthesis and carotenoid accumulation ofC. fusca were examined. KNO3 and CH3COONa were used as the nitrogen and the organic carbon sources, respectively. They are very important nutrient sources to the microalgae. In fact, when the concentration of the nutrient sources was increased, the syntheses of chlorophyll and the carotenoid accumulation of the microalgae were promoted. Cfusca can take an organic carbon as the nutrient sources for its own growth, so the cell is activated compared to the case only in an inorganic nourishment sources. Mg2＋ is known as a raw material of chlorophyll and it can promote carotenoid accumulation. However, when it exists in excessive quantity, the microalgae receive a stress and it was difficult for microalgae to uptake such nutrient sources. In this study, it was found that there existed the optimum amounts of KNO3, CH3COONa, NaCI and MgSO4 which should be added to the culture medium.

Characteristics of In-Hospital Patients with Congenital Heart Disease Requiring Rapid Response System Activations: A Japanese Database Study

Objectives:This study aimed to study the characteristics of in-hospital deterioration in patients with congenital heart disease who required rapid response system activation and identify risk factors associated with 1-month ***:We retrospectively analysed data from a Japanese rapid response system registry with 35 participating *** included consecutive patients with congenital heart disease who required rapid response system activation between January 2014 and March *** regression analyses were performed to examine the associations between 1-month mortality and other patient-specific ***:Among 9,607 patients for whom the rapid response system was activated,only 82(0.9%)had congenital heart *** few patients with congenital heart disease were being treated at the cardiology and cardiovascular surgery departments(12.3%and 9.9%,respectively).Moreover,the incidences of rapid-response events after intensive care unit discharge or surgery were low(6.8% and 12.2%,respectively).The most common reason for rapid response system activation was respiratory dysfunction(desaturation:35.4%,tachypnoea:25.6%,and new dyspnoea:19.5%).Rapid response system interventions and intensive care unit transfers were required for 65.9% and 20.7%of patients,*** mortality rate was 1.2% at the end of the rapid response system intervention and 11.0% after 1 ***,decreased respiratory rate and decreased heart rate at rapid response system activation were associated with increased 1-month *** adjusted odds ratio was 1.10(95% confidence interval 1.02–1.19)and 1.02(95% confidence interval,1.00–1.04) for respiratory rate and heart rate,***:Rapid response systems were rarely activated after cardiac surgery and intensive care unit discharge,which were situations with a high risk of sudden deterioration in patients with congenital heart ***,encouraging the use of the rapid response sys

Driving skill classification in curve driving scenes using machine learning

Driver support and infotainment systems can be adapted to the specific needs of individual drivers by assessing driver skill and state. In this paper, we present a machine learning approach to classifying the skill at maneuvering by drivers using both longitudinal and lateral controls in a vehicle. Conceptually, a model of drivers is constructed on the basis of sensor data related to the driving environment, the drivers＇ behaviors, and the vehi- cles＇ responses to the environment and behavior together. Once the model is built, the driving skills of an unknown driver can be classified automatically from the driving data. In this paper, we demonstrate the feasibility of using the proposed method to assess driving skill from the results of a driving simulator. We experiment with curve driving scenes, using both full curve and segmented curve sce- narios. Six curves with different radii and angular changes were set up for the experiment. In the full curve driving scene, principal component analysis and a support vector machine-based method accurately classified drivers in 95.7 % of cases when using driving data about high- and low/average-skilled driver groups. In the cases with seg- mented curves, classification accuracy was 89 %.
