The recent identification of homozygous WNT1 mutations in individuals with osteogenesis imperfecta type XV(OI-XV)has suggested that WNT1 is a key ligand promoting the differentiation and function of bone-forming *** such aninfluence was supported by subsequent studies,a mouse model of OI-XV remained to be ***,we introduced a previously identified disease-causing mutation(G177C)into the murine Wnt1 *** Wnt1^(G177C/G177C)mice were viable and did not display defects in brain development,but the majority of 24-week-old Wnt1^(G177C/G177C)mice had skeletal *** increased bone fragility was not fully explained by reduced bone mass but also by impaired bone matrix ***,the homozygous presence of the G177C mutation did not interfere with the osteoanabolic influence of either parathyroid hormone injection or activating mutation of LRP5,the latter mimicking the effect of sclerostin ***,transcriptomic analyses revealed that short-term administration of WNT1 to osteogenic cells induced not only the expression of canonical WNT signaling targets but also the expression of genes encoding extracellular matrix *** together,our data demonstrate that regulating bone matrix quality is a primary function of *** further suggest that individuals with WNT1 mutations should profit from existing osteoanabolic therapies.

Technology based on high-peak-power lasers has the potential to provide compact and intense radiation sources for a wide range of innovative *** particular,electrons that are accelerated in the wakefield of an intense laser pulse oscillate around the propagation axis and emit *** betatron source,which essentially reproduces the principle of a synchrotron at the millimeter scale,provides bright radiation with femtosecond duration and high spatial ***,despite its unique features,the usability of the betatron source has been constrained by its poor control and *** this article,we demonstrate the reliable production of X-ray beams with tunable *** ionization-induced injection in a gas mixture,the orbits of the relativistic electrons emitting the radiation are reproducible and *** observe that both the signal and beam profile fluctuations are significantly reduced and that the beam pointing varies by less than a tenth of the beam *** polarization ratio reaches 80%,and the polarization axis can easily be *** anticipate a broad impact of the source,as its unprecedented performance opens the way for new applications.

AIM:To investigate the effect of short-chain fatty ac-ids (SCFAs) on production of prostaglandin E2 (PGE2),cytokines and chemokines in human ***:Human neutrophils and monocytes were isolated from human whole blood by using 1-Step Polymorph and RosetteSep Human Monocyte Enrich-ment Cocktail,respectively. Human GPR41 and GPR43 mRNA expression was examined by quantitative real-time polymerase chain reaction. The calcium flux assay was used to examine the biological activities of SCFAs in human neutrophils and monocytes. The effect ofSCFAs on human monocytes and peripheral blood mononuclear cells (PBMC) was studied by measuring PGE2,cytokines and chemokines in the supernatant. The effect of SCFAs in vivo was examined by intraplantar injection into rat ***:Human GPR43 is highly expressed in human neutrophils and monocytes. SCFAs induce robust calcium flux in human neutrophils,but not in human monocytes. In this study,we show that SCFAs can in-duce human monocyte release of PGE2 and that this effect can be enhanced in the presence of lipopolysac-charide (LPS). In addition,we demonstrate that PGE2 production induced by SCFA was inhibited by pertussis toxin,suggesting the involvement of a receptor-mediated mechanism. Furthermore,SCFAs can specifi cally inhibit constitutive monocyte chemotactic protein-1 (MCP-1) production and LPS-induced interleukin-10 (IL-10) production in human monocytes without affecting the secretion of other cytokines and chemokines examined. Similar activities were observed in human PBMC for the release of PGE2,MCP-1 and IL-10 after SCFA treatment. In addition,SCFAs inhibit LPS-induced production of tumor necrosis factor-α and interferon-γ in human PBMC. Finally,we show that SCFAs and LPS can induce PGE2 production in vivo by intraplantar injection into rat paws (P < 0.01). CONCLUSION:SCFAs can have distinct antiinflammatory activities due to their regulation of PGE2,cytokine and chemokine release from human immune cells.

＜正＞A variety of xenobiotics in the environment has been shown to induce adverse effects in animals and humans by interfering with their endocrine functions at different modes of action. However, our knowledge about the effects of these so-called endocrine disrup-tors, especially on invertebrates, is still limited. Many of the substances are considered to be rather persistent, which is partly due to their strong adsorption to suspended particles and sediments in aquatic systems. Besides, they reveal a substantial bioaccu-mulation potential. Hence, it is highly relevant to conduct experiments in which exposure via sediments takes place. Therefore, we developed an invertebrate test system with the sediment-dwelling freshwater mudsnail Potamopyrgus antipodarum （Gastropoda, Prosobranchia） to identify and assess endocrine-mimetic test compounds.
