^(68)Gallium-labelled PSMA-PET/cT as a diagnostic and clinical decision-making tool in Asian prostate cancer patients following prostatectomy

Objective: Prostate cancers(Pca) in Asian individuals are molecularly distinct from those found in their caucasian *** is no risk stratification tool for Asian men with rapid biochemical recurrence(BcR) following radical prostatectomy(Rad P). This study aims to assess the detection rate of ^(68)Ga-prostate-specific membrane antigen-positron emission tomography/computed tomography(PSMA-PET/cT) for diagnosis of clinical recurrence and as a treatment decision making tool in Asian patients with BcR post-Rad ***: ^(68)Ga PSMA-PET and cT body with/without bone scan [conventional workup(cWU)] were performed in 55 Asian patients with BcR within 36 months post-Rad P. Two blinded reviewers assessed the images. Detection rates of ^(68)Ga PSMAPET/cT were evaluated, and impact on management was reviewed by comparison with ***: Median time to BcR post-Rad P was 8.1 months. Detection rate for ^(68)Ga PSMA-PET/cT was 80%(44/55). A positive scan was significantly associated with increasing prostate-specific antigen(PSA) level [odds ratio(OR) = 1.13(95% cI 1.05–1.30), P =0.017], but not with higher Gleason grade or shorter PSA doubling time. compared to cWU, ^(68)Ga PSMA-PET/cT detected an additional 106 lesions in 33/44 patients with a positive scan, resulting in a change in management in 25/44(56.8%) patients: 10 to hormonal therapy(HT) and whole pelvis radiotherapy(RT) in addition to bed RT, and 15 to palliative HT ***: In the present report, we demonstrated the diagnostic and treatment decision utility of ^(68)Ga PSMA-PET/cT in Asian men with rapid BcR. Detection of small volume nodal and systemic recurrences at low PSA levels(l) highlights the role of the tool in assigning patients to treatment intensification with HT-RT or palliative HT in polymetastatic disease.

Dear editor,Prostate cancer(Pca)remains a major healthcare burden in men globally[1].Most patients present with localized disease,and treatment is recommended based on risk classification systems like the National comprehensive cancer Network(NccN)[2].However,these methods are imprecise for estimating metastasis-free survival and prostate cancer-specific mortality and thus biomarkers that can predict tumor aggression are needed[3–5].Several studies have since characterized the molecular landscape of localized Pca in White[4,5]and Black/African-American men[6],but data is lacking in Asian *** chinese Prostate cancer Genome and Epigenome Atlas(cPGEA)reported on the genomic and epigenomic landscape of 208 Pca of men from china[7].comparative analyses between the cPGEA cohort and data from The cancer Genome Atlas(TcGA)revealed higher frequencies of Forkhead box A1(FOXA1)and chromodomain-helicase DNA-binding 1(cHD1)mutations,and lower frequencies of phosphatase and tensin homolog(PTEN)mutations and transmembrane protease serine 2-E26 transformationspecific related gene(TMPRSS2-ERG)fusion in chinese compared with White men[7].These preliminary findings highlight the presence of race-specific differences in molecular phenotypes of Pca.

NEcAB family of neuronal calcium-binding proteins in health and disease

The N-terminal EF-hand calcium-binding proteins 1–3(NEcAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized c-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NEcAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NEcAB2 interacts with predominantly striatal G-protein-coupled receptors, while NEcAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NEcAB proteins exhibit distinct subcellular localizations: NEcAB1 is found in the nucleus and cytosol, NEcAB2 resides in endosomes and the plasma membrane, and NEcAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NEcAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NEcAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. Scalable Ir-Doped NiFe₂O₄/TiO₂ Heterojunction Anode for Decentralized Saline Wastewater Treatment and H₂ Production

Wastewater electrolysis cells (WEcs) for decentralized wastewater treatment/reuse coupled with H-2 production can reduce the carbon footprint associated with transportation of water, waste, and energy carrier. This study reports Ir-doped NiFe2O4 (NFI, similar to 5 at% Ir) spinel layer with TiO2 overlayer (NFI/TiO2), as a scalable heterojunction anode for direct electrolysis of wastewater with circumneutral pH in a single-compartment cell. In dilute (0.1 M) Nacl solutions, the NFI/TiO2 marks superior activity and selectivity for chlorine evolution reaction, outperforming the benchmark IrO2. Robust operation in near-neutral pH was confirmed. Electroanalyses including operando X-ray absorption spectroscopy unveiled crucial roles of TiO2 which serves both as the primary site for cl- chemisorption and a protective layer for NFI as an ohmic contact. Galvanostatic electrolysis of NH4+-laden synthetic wastewater demonstrated that NFI/TiO2 not only achieves quasi-stoichiometric NH4+-to-N-2 conversion, but also enhances H-2 generation efficiency with minimal competing reactions such as reduction of dissolved oxygen and reactive chlorine. The scaled-up WEc with NFI/TiO2 was demonstrated for electrolysis of toilet wastewater.

Two-phase micro-and macro-time scales in particle-laden turbulent channel flows

The micro- and macro-time scales in two-phase turbulent channel flows are investigated using the direct nu- merical simulation and the lagrangian particle trajectory methods for the fluid- and the particle-phases, respectively. lagrangian and Eulerian time scales of both phases are cal- culated using velocity correlation functions. Due to flow anisotropy, micro-time scales are not the same with the theo- retical estimations in large Reynolds number （isotropic） tur- bulence. lagrangian macro-time scales of particle-phase and of fluid-phase seen by particles are both dependent on particle Stokes number. The fluid-phase lagrangian inte- gral time scales increase with distance from the wall, longer than those time scales seen by particles. The Eulerian inte- gral macro-time scales increase in near-wall regions but de- crease in out-layer regions. The moving Eulerian time scales are also investigated and compared with lagrangian integral time scales, and in good agreement with previous measure- ments and numerical predictions. For the fluid particles the micro Eulerian time scales are longer than the lagrangian ones in the near wall regions, while away from the walls the micro lagrangian time scales are longer. The lagrangian integral time scales are longer than the Eulerian ones. The results are useful for further understanding two-phase flow physics and especially for constructing accurate prediction models of inertial particle dispersion.

The effect of milling additives on powder properties and sintered body microstructure of NiO

The evolution of powder particle size,crystal structure,and surface chemistry was evaluated for micron scale NiO powders subjected to impact milling with commonly employed milling additives:methanol,Vertrel XF,and amorphous *** effect of the different comminution protocols on sintered body microstructure was evaluated for high temperature sintering in inert atmosphere(N_(2)).X-ray photoelectron spectroscopy showed that NiO powder surface chemistry is surprisingly sensitive to milling additive *** particular,the proportion of powder surface defect sites varied with additive,and methanol left an alcohol or alkoxy residue even after *** sintering to intermediate temperatures(1100℃),scanning electron microscopy(SEM)showed that slurry milled NiO powders exhibit hindered sintering *** effect was amplified for NiO milled with methanol,in which sub-500 nm grain sizes dominated even after sintering to 1100℃.Upon heating to high temperatures(1500℃),simultaneous differential scanning calorimetry/thermogravimetric analysis(DSc/TGA)showed that the powders containing carbon residues undergo carbothermal reduction,resulting in a melting transition between 1425 and 1454℃.Taken together,the results demonstrated that when processing metal oxide powders for advanced ceramics,the choice of milling additive is crucial as it exerts significant control over sintered body microstructure.

Experimental and Numerical Investigation of an End Plate connection with Using long Shank SMA Bolts

Superelastic shape memory alloys( SMAs) have the ability to recover their original shape after experiencing large strains. End-plate connection with using superelastic long shank SMA bolts is proposed in this paper. By using strong beam-weak bolt cluster design methodology and special configurations to strength beam ends,a superelastic hinge is expected to form in column flange. To validate the mechanical behavior of the proposed beam-to-column connection,both quasi-static tests and numerical analysis are conducted with cyclic transverse loads applied on the beam ends. The results indicate the connection deformations concentrate on the long shank SMA bolts upon loading and the bolt cluster rotates around the axis near beam flange. By using the super elastic effect,the SMA bolts recover most elongations and the connection recovers to its original shape after experiencing 0. 02 rad interstorey drift angle. The connection shows obvious self-centering properties.

The antibody repertoire of Bos taurus is characterized by a subset of variable heavy(VH)chain regions with ultralong third complementarity determining regions(cDR3)which,compared to other species,can provide a potent response to challenging antigens like HIV *** unusual cDR3 can range to over seventy highly diverse amino acids in length and form uniqueβ-ribbon‘stalk’and disulfide bonded‘knob’structures,far from the typical antigen binding *** genetic components and processes for forming these unusual cattle antibody VH cDR3 are not well *** we analyze sequences of Bos taurus antibody VH domains and find that the subset with ultralong cDR3 exclusively uses a single variable gene,IGHV1-7(VHBUl)rearranged to the longest diversity gene,*** eight nucleotide duplication at the 3′end of IGHV1-7 encodes a longer V-region producing an extended Fβ-strand that contributes to the stalk in a rearranged cDR3.A low amino acid variability was observed in cDR1 and cDR2,suggesting that antigen binding for this subset most likely only depends on the *** a novel,potentially AID mediated,deletional diversification mechanism of the *** VH ultralong cDR3 knob was discovered,in which interior codons of the IGHD8-2 region are removed while maintaining integral structural components of the knob and descending strand of the stalk in *** deletions serve to further diversify cysteine positions,and thus disulfide bonded ***,both germline and somatic genetic factors and processes appear to be involved in diversification of this structurally unusual cattle VH ultralong cDR3 repertoire.
